A specific and qualitative detection method for buckwheat in foods using the polymerase chain reaction (PCR) was developed. Trace amounts of buckwheat in commercial food products were qualitatively detected by this method. -2-haloalkanoic acids to produce the corresponding

-2-hydroxyalkanoic acids, respectively. We have constructed an overproduction system for
-2-haloacid dehalogenase from Pseudomonas putida PP3 (
-DEX 312) and purified the enzyme to analyze the reaction mechanism. When a single turnover reaction of
-DEX 312 was carried out in H218O by use of a large excess of the enzyme with
- or
-2-chloropropionate as a substrate, the lactate produced was labeled with 18O. This indicates that the solvent water molecule directly attacked the substrate and that its oxygen atom was incorporated into the product. This reaction mechanism contrasts with that of
-2-haloacid dehalogenase, which has an active-site carboxylate group that attacks the substrate to displace the halogen atom.
-DEX 312 resembles
-2-haloacid dehalogenase from Pseudomonas sp. 113 (
-DEX 113) in that the reaction proceeds with a direct attack of a water molecule on the substrate. However,
-DEX 312 is markedly different from
-DEX 113 in its substrate specificity. We found that
-DEX 312 catalyzes the hydrolytic dehalogenation of 2-chloropropionamide and 2-bromopropionamide, which do not serve as substrates for
-DEX 113.

The spatial distribution of basidiocarps provides much information on the dispersal abilities, habitat preferences, and inter- and intraspecific interactions of aphyllophoraceous fungi. To reveal the spatial distribution and resource utilization patterns of aphyllophoraceous fungi in Malaysia, we conducted field observations in a primary forest in 2006 and analyzed the relationships between the abundance of eight dominant fungal species and various environmental factors. The topographical characteristics were significantly patchily distributed at the 100-m scale, whereas woody debris and most fungal species were distributed randomly. Although the dominant fungal species differed among the decay classes and diameters of the woody debris, the abundance of a few dominant species was significantly correlated with environmental factors. Although the latter factors might affect the spatial distribution of these fungi, the effects appear to be so small that they would not create an aggregated distribution at a few 100-m scales. 相似文献

A stable mutant flocculent yeast strain of Saccharomyces cerevisiae KRM-1 was isolated during repeated-batch ethanol fermentation using kitchen refuse as the medium. The mechanism of flocculation and interaction with the medium was investigated. According to sugar inhibition assay, it was found that the mutant flocculent strain was a NewFlo phenotype. Flocculation was completely inhibited by protease, proteinase K and partially reduced by treatments with carbohydrate-hydrolyzing enzymes. Flocculation ability showed no difference for pH 3.0–6.0. Furthermore, the mutant flocculent yeast provided repeated-batch cultivations employing cell recycles by flocculation over 10 rounds of cultivation for the production of ethanol from kitchen refuse medium, resulting in relatively high productivity averaging 8.25 g/L/h over 10 batches and with a maximal of 10.08 g/L/h in the final batch. We develop discrete-time models for analyzing the long-run equilibrium outcomes on invasive species management in two-patch
environments with migration. In particular, the focus is on a situation where removal operations for invasive species are
implemented only in one patch (controlled patch). The new features of the model are that (1) asymmetry in density-dependent
migration is considered, which may originate from impact of harvesting as well as heterogeneous habitat conditions, and (2)
the effect of density-dependent catchability accounts for the fact that the required effort level to remove one individual
may rise as the existing population decreases. The model is applied to agricultural damage control in the raccoon (Procyon lotor) problem that has occurred in Hokkaido, Japan. Numerical illustrations demonstrate that the long-run equilibrium outcomes
largely depend on the degree of asymmetry in migration as well as the sensitivity of catchability in response to a change
in the population size of the invasive species. Furthermore, we characterize the conditions under which the economically optimal
effort levels are qualitatively affected by the above two factors, and conclude that aiming at local extermination of invasive
species in the controlled patch is justified.

Three morphologically and genetically distinct forms of the genus Carassius were collected from the Ob River system, Kazakhstan, Central Asia; Carassius carassius , Carassius gibelio gibelio and an unknown stock tentatively referred to as Carassius gibelio sub-species M. The last mentioned had 33–41 gill rakers, being intermediate between the other two forms (23–27 in C. carassius and 44–49 in C. g. gibelio ), and five scales in the upper transverse series, less than in the others. It also had a relatively larger erythrocyte suggesting triploidy and an mtDNA haplotype distinct from all other known crucian carps. Comparative mtDNA phylogenetic analysis suggested that C. gibelio gibelio in the Ob River system was introduced from China and the Amur River, the same possibly being true for European C. gibelio gibelio based on published haplotypes. C. gibelio sub-species M is thought to be more widely distributed in central Asia, probably extending as far west as European Russia. 相似文献
