The Q strain known as v6 was tested for its ability to induce X-linked lethal mutations in male and female hybrids from crosses with M strains in the P-M system of hybrid dysgenesis. All measurements of the mutation rate were made on the X chromosome derived from the v6 strain. The lethal rate for young hybrid males from the cross M female X v6 male was 1.11% per chromosome. For older males, it was only 0.44%, suggesting that there is less mutational or more repair activity in the germ cells of the older males or that mutant cells are selectively eliminated as the hybrid males age. The lethal rate for hybrid females from comparable crosses was approximately the same for both ages that were tested. However, it was substantially less than the rate for the hybrid males--only 0.26% per chromosome. Genetically identical hybrid females from reciprocal crosses also showed a low mutation rate, 0.13% per chromosome. Again, there was no difference between young and old flies. Mapping experiments established that most of the lethal mutations that were recovered from the male and female hybrids were located in two regions on the X chromosome, one between bands 14B13 and 15A9 , the other between bands 19A1 and 20A , which encompasses the maroonlike locus. More refined mapping of the lethals in the maroonlike region demonstrated that the vast majority of these affected a single gene located in band 19C4 . Cytological analysis of the lethal chromosomes revealed that several carried rearrangements, including inversions, duplications and deficiencies. Chromosome breakage occurred primarily in bands 14D1 -3 and 18F- 20A , and most of the breaks in the latter segment were located in 19C . However, rearrangements involving 19C and mutations of the gene in 19C4 were mutually exclusive events. In situ hybridization of a P element probe to the chromosomes of v6 demonstrated that P elements reside at a minimum of five sites on the X chromosome. These P element sites correspond to the mutational and breakage hot spots on that chromosome. The combined genetic and cytological data imply that most of the X-linked lethal mutations that occur in M X v6 hybrids are due to local P element action. Consideration of these and other data suggest that v6 is a weak P strain in the P-M system of hybrid dysgenesis and that other Q strains might also be regarded in this way.  相似文献

The morphology and arrangement of pili in the P++ colony phenotype of Neisseria gonorrhoeae were examined by a variety of electron microscopic techniques. The apparent structure and organization of gonococcal pili varied depending upon the method of specimen preparation. Pili as thin, individual, unbranched structures were demonstrated by negative staining and in sections of epoxy-embedded specimens. Pili forming thick structures which branch, subdivide, and rejoin to form an irregular lattice were demonstrated in specimens processed by the critical-point drying method and by rapid freezing and low temperature sublimination. We propose that in gonococcal colonies of the P++ phenotype, pili exist as individual threadlike structures only on the bacterial surfaces; as the pili leave the bacterial surfaces, they form thick bundles which branch, subdivide, and rejoin to form a supporting framework interconnecting the colony members. This arrangement of pili is usually disrupted by the commonly used method of negative staining and cannot be clearly detected within epoxy-embedded specimens. These data are summarized in a model depicting the organization of pili in the P++ colony phenotype of N. gonorrhoeae.  相似文献

Summary Robber fly species within a Panamanian rain forest comprised distinct shade-seeking (SS) and light-seeking (LS) groups. Thoracic temperatures of LS species averaged 9.2°C greater than ambient, whereas those of SS species averaged only 1.3°C above ambient. Among SS species, attack rate decreased with increasing body mass, whereas relocation rate and attack and relocation distances increased with increasing body mass. Attack and relocation distances of LS species were similar to those of SS species of similar size. Large (>100 mg) LS species, however, had much higher attack and relocation rates than large SS species. The potential costs and benefits of basking are briefly discussed.  相似文献

The binding of phosphofructokinase and aldolase to the membrane of the intact human erythrocyte was assessed by the rapid hemolysis/filtration method of Kliman and Steck (Kliman, H. J., and Steck, T. L. (1980) J. Biol. Chem. 255, 6314-6321). We found that about 50% of the phosphofructokinase was membrane-bound in fresh red cells prior to hemolysis. Binding was not significantly altered by deoxygenation. Approximately 40% of aldolase was membrane-associated in fresh red cells. In outdated, blood-banked red cells, aldolase was 73% membrane-bound while, following metabolic repletion, 40% of the enzyme was membrane-associated. These results support the hypothesis that certain glycolytic enzymes in the red cell are membrane-bound in a rapidly reversible and metabolically sensitive fashion.  相似文献

We have isolated a BALB/c (H-2d, Mlsb) T cell clone (JTL-G12) specific for the synthetic polypeptide antigen poly(Glu60Ala30Tyr10) (GAT) in the context of self I-A determinants and for Mlsa,d antigens in the absence of GAT. JTL-G12 proliferation in response to GAT was mapped to the Kd, I-Ad subregions by using inbred H-2 congenic and recombinant strains. In addition, monoclonal antibody directed against I-Ad but not Kd or I-As determinants blocked JTL-G12 proliferation in response to GAT presented by syngeneic splenocytes, indicating I-A restriction. The Mls cross-reactivity of this clone was verified by using a panel of inbred strains bearing the Mlsa,b,c,d alleles and by using BXD recombinant inbred strains bearing the Mlsa allele or the Mlsb allele. All of the Mlsa BXD strains of the H-2d or H-2b haplotypes stimulated JTL-G12 in the absence of GAT, whereas all of the Mlsb BXD strains were nonstimulatory. This response pattern is in complete accordance with recognition of the Mlsa determinant encoded by Mls or closely linked loci on chromosome 1. JTL-G12 proliferation in response to GAT/I-Ad and Mlsa,d determinants could be blocked with a monoclonal antibody (GK1.5) directed against L3T4, a structure involved in class II major histocompatibility complex antigen recognition. These results suggest that antigen/class II responsiveness, Mls reactivity, and expression of L3T4 can be properties of a single T cell population.  相似文献

The mechanisms underlying Ir gene control of CMI were addressed by examining the DTH and Tprlf responses specific for the synthetic polymers GT, GAT, and GA. We show that BALB/c mice (GAT/GA responders, GT nonresponders) primed with GT fail to develop DTH and Tprlf responses specific for GT, GAT, or GA. GAT immunization resulted in DTH responses that could be elicited not only with GAT and GA but also with GT, demonstrating that GT-specific TDH are present in nonresponder mice. GT-specific DTH was transferred with Thy-1+ Lyt-1+2, H-2 Irestricted, nylon wool nonadherent cells. GA-primed BALB/c mice developed GAT- and GA-, but not GT-apecific DTH responses, indicating that GA and GT do not cross-react at the T-cell level. The ability of GAT [but not a mixture of GA plus GT, or GT electrostatically complexed to the immunogenic carrier MBSA (GT-MBSA)] to induce GT-specific DTH suggested a requirement for covalent linkage of stimulatory GA and nonstimulatory GT determinants present on the GAT molecule. Similarly, GT-specific in vitro Tprlf responses could be demonstrated in GAT-primed mice exhibiting significant levels of GT-specific DTH but not in GT- or GT-MBSA-primed mice. Tolerization experiments also suggested that GT-specific Th were involved in the development of GT-specific DTH in GAT-primed mice. The GT nonresponsiveness of BALB/c mice for DTH and Tprlf responses could not be reversed by treatments designed to abrogate Ts activity (priming with GT-MBSA and CY injection), nor could GT-primed cells be shown to inhibit the development or elicitation of GT-specific CMI in GAT-primed mice during the afferent and/or efferent stages of DTH. Our results suggest that GT nonresponsiveness does not result from the absence of GT-specific T cells or preferential induction of Ts. Observers judged the slants and tilts of numerous regions within shaded images of ellipsoid surfaces that varied in shape, orientation, surface reflectance, and direction of illumination. The perceived three-dimensional structure of each surface was calculated from these judgments. Much of the error in observers' responses resulted from a tendency to perceive surfaces whose axes were aligned with the display screen. The presence of specular highlights or cast shadows, in contrast, had no effect on performance. The results of the experiment indicate that several assumption of certain formal models for perception of shape from shading are not psychologically valid. The most notable of these assumptions are that the visual system initially assumes that all surfaces have Lambertian reflectance and that illuminant direction must be known before shape detection can proceed. These assumptions are often accompanied by a third assumption that surface orientation is detected locally, and global shape determined by smoothing over local surface orientation estimates. The present experiment indicates that an alternative approach offered by Koenderink and van Doorn may be more psychologically accurate, as it avoids all three assumptions.Supported in part by the Air Force Office of Scientific Research (AFOSR 82-0148). The subunit stoichiometry of the ATP synthetase (CF1-CF0) immunoprecipitated from Triton X-100 extracts of chloroplast thylakoid membranes was determined to be α3, β3, γ, δ, ? (CF1) and I0.3, II0.6–0.9, III4(6) (CF0). Antibodies against the polypeptides α, β, γ, δ, I, II and ? combined specifically with the isolated subunits as analysed by the protein blotting method. Applying this technique, antibodies against the CF1 subunits were found to form complexes with the corresponding polypeptides of thylakoids, whereas those against I (Mr 20 000) and II (Mr 17 000) combined with Mr 26 000 and Mr 24 500 membrane polypeptides, respectively. The Mr 26 000 polypeptide was identified as the major subunits of the light-harvesting chlorophyll a/b-protein (LHCP) complex and the Mr 24 500 component seems to be functionally connected with this complex. From the results it is concluded that the chloroplast ATP synthetase consists of the subunit of the α, β, γ, δ, ? and III (proteolipid only and that proteolytically altered LHCP polypeptides bind artifically to the protein complex during isolation.  相似文献
