Coatomer is the soluble precursor of the COPI coat (coat protein I) involved in traffic among membranes of the endoplasmic reticulum and the Golgi apparatus. We report herein that neomycin precipitates coatomer from cell extracts and from purified coatomer preparations. Precipitation first increased and then decreased as the neomycin concentration increased, analogous to the precipitation of a polyvalent antigen by divalent antibodies. This suggested that neomycin cross-linked coatomer into large aggregates and implies that coatomer has two or more binding sites for neomycin. A variety of other aminoglycoside antibiotics precipitated coatomer, or if they did not precipitate, they interfered with the ability of neomycin to precipitate. Coatomer is known to interact with a motif (KKXX) containing adjacent lysine residues at the carboxyl terminus of the cytoplasmic domains of some membrane proteins resident in the endoplasmic reticulum. All of the antibiotics that interacted with coatomer contain at least two close amino groups, suggesting that the antibiotics might be interacting with the di-lysine binding site of coatomer. Consistent with this idea, di-lysine itself blocked the interaction of antibiotics with coatomer. Moreover, di-lysine and antibiotics each blocked the coating of Golgi membranes by coatomer. These data suggest that certain aminoglycoside antibiotics interact with di-lysine binding sites on coatomer and that coatomer contains at least two of these di-lysine binding sites.  相似文献

Maize has been used effectively as a model organism in the development and evaluation of molecular markers for the identification, mapping and manipulation of major genes affecting the expression of quantitative traits in plants. Although quantitative geneticists have recognized the possibility of major loci, the general dogma bad emerged that quantitative traits were controlled by many loci, each with a small effect. This interpretation sent a signal to the molecular biologist not to bother with quantitative traits because it would be essentially impossible to isolate a gene responsible for the trait. Recent results from numerous mapping studies have shown that quantitative traits are controlled by, at least some, factors with major effects, and have given credibility to the conclusion that major loci exist and that one might be able to study them. Positive results from marker-facilitated selection and introgression studies have further strengthened this conclusion.  相似文献

The flavonoids of eleven species of Clibadium are all based on the flavonols kaempferol, quercetin and quercetagetin. Glucose and galactose sugar substitutions occur in all taxa, while rhamnosides, arabinosides, xylosides, rutinosides and diglucosides are less widely distributed. O-Methylated compounds provide the most meaningful taxonomic information and serve to divide the species into two groups. Infraspecific variation of flavonoids is commonplace. We investigated the effect of "psyching-up" on force production during the bench press. Twelve men (mean age +/- SD: 27.4 +/- 11.2 years) and 8 women (20.9 +/- 2.5 years) with strength-training experience performed 5 bench press repetitions on a modified Biodex isokinetic dynamometer during 3 interventions. The interventions were counterbalanced and included a free-choice psych-up, a cognitive distraction, and an attention-placebo. Peak force recorded after psyching-up (mean +/- SD: 764 +/- 269 N.m) was significantly different from both distraction (703 +/- 282 N.m, p = 0.003) and attention-placebo (708 +/- 248 N.m, p = 0.01). The mean percentage increase in peak force from distraction to psyching-up was 11.8% (6 to 18%, 95% confidence interval [CI]) and 8.1% from placebo to psyching-up (3 to 13%, 95% CI). The results of the present study indicate that psyching-up may increase force production during the bench press exercise in participants with at least 1 year strength-training experience.  相似文献

The previously insignificant and small South American subtribe Barnadesiinae of tribe Mutisieae has been shown recently to be ancient within Asteraceae. Due to absence of a 22 kb diagnostic chloroplast inversion, plus other features, this subtribe has been elevated to subfamilial status as Barnadesioideae, now containing 90 species in nine genera. Recent cladistic analyses at the generic and specific levels based on morphology and DNA sequences (ITS and trnL intron) have revealed different relationships among taxa of the subfamily. To better understand these conflicts, we analyze specific morphological features of corollas (shape and vascularization) to develop trends (trees) in these features based on minimal structural change (i.e., morphological parsimony), and to compare these with relationships among genera derived from the two recent cladistic analyses. We define six principal types of corollas in Barnadesioideae (tubular, split, double split, ligulate, subbilabiate and bilabiate) and six principal types of corolla vascularization patterns (combinations of presence or absence of central bundles, fusion of adjacent bundles and fusion of bundles at the apex of corolla lobes). In all features we assume character states in Calyceraceae to be ancestral. In corolla shape, from tubular ancestry, we hypothesize two general evolutionary trends within the subfamily: (1) splitting of the tube, and (2) flattening of the tube. In vascularization of corollas, from an ancestral condition of one central and one lateral bundle in each corolla lobe and all traces fused at the apex, we hypothesize four basic trends: (1) gain or (2) loss of lateral vascular traces in each lobe, (3) loss of the central vein, and (4) loss of fusion of traces in lobe apices. These morphological trends allow tests of the two previous phylogenetic hypotheses by (1) counting step changes (following steps in the morphological network) in the two characters on the two competing phylogenetic trees and (2) constructing an index of morphological advancement for each genus (based on morphological trees of the two characters) and correlating these with cladistic distances within the two phylogenies. Results of both tests reveal the molecular phylogeny to be more compatible with evolutionary inferences from the two morphological features. With Fulcaldea excluded, a highly significant correlation is seen between morphological advancement and cladistic distance in the molecular phylogeny.  相似文献

Dinophysis acuminata and D. norvegica were observed in plankton net samples during the summer of 2002 from the Kandalaksha Gulf in the White Sea (North European Russia). Prorocentrum lima was found as an epiphyte on subtidal macroalgae in August, but not observed in plankton net samples. Protein phosphatase 2A (PP2A) inhibition measured 127.8 ng OA-equivalent/g of mussel (Mytilus edulis) hepatopancreas from samples collected a few days after when Dinophysis was recorded at a density of 1550 cells L−1. Liquid chromatography–mass spectrometry confirmed presence of several classes of lipophilic shellfish toxins associated with Dinophysis spp. in the mussels including okadaic acid, dinophysistoxin-1, pectenotoxins and yessotoxins. No azaspiracid was detected. This represents the first identification of phycotoxicity in the White Sea.  相似文献
