Factors affecting the isolation of CCC DNA from Streptomyces lividans and Escherichia coli

Based on the results of a systematic study of factors affecting plasmid yield and purity, a procedure suitable for the rapid screening for and isolation of covalently closed circular DNA from Streptomyces lividans and Escherichia coli was developed. The method consists of lysis of lysozyme-treated bacteria combined with alkaline denaturation of DNA at high temperature. Renaturation of CCC DNA and precipitation of single-stranded DNA together with protein is achieved by the addition of a minimal amount of phenol/chloroform. The screening procedure uses only a single tube and the samples can be analyzed by agarose gel electrophoresis about 30 min after lysis. Removal of phenol and further purification of the plasmid preparation is achieved by consecutive precipitations with isopropanol and spermine, followed by extraction with ethanol, producing samples suitable for restriction endonuclease digestion, ligation, and transformation of S. lividans protoplasts or competent E. coli cells in about 2 h. All steps of the procedure are explained in detail with information about the effects of changing parameters. This should help the experimenter to obtain reproducible results and may be useful if the method has to be adapted to new strains or plasmids.     

Biochemical pharmacology of the gamma-aminobutyric acid receptor/ionophore protein

The synaptic receptor sites for the neurotransmitter gamma-aminobutyric acid (GABA) can be assayed in vitro with several radiolabeled agonists and one antagonist. Numerous criteria of specificity have been met for these binding sites. All of the ligands show heterogeneity in binding affinities. The subpopulations thus defined have a remarkably similar specificity for GABA analogs, which suggests an intimate relationship and possible interconvertibility. Modulation of GABA receptor binding by barbiturates, anions, and other membrane treatments that affect agonists and antagonists in an opposite manner suggests a three-state model of interconvertible affinities. The complex of GABA receptor and chloride ion channel contains modulatory sites for barbiturates and benzodiazepines, drugs that enhance GABA responses in neurons. The receptor complex can be solubilized in detergent with the three mutually interacting receptor activities intact. The complex has an apparent molecular weight of 355,000 and has been partially purified. GABA agonist function has been assayed at the biochemical level by measuring the activation of 36Cl- efflux from preloaded hippocampal slices by GABA, muscimol, and barbiturates. This response is blocked by the antagonists of the GABA site (bicuculline) and the barbiturate site (picrotoxin). Comparison of binding and function on the same tissue should be useful in analyzing the mechanism of action of GABA.     

Hormone-sensitive stages in the sexual differentiation of laryngeal muscle fiber number in Xenopus laevis

The number of muscle fibers in the vocal organ of the adult male African clawed frog, Xenopus laevis, exceeds that of adult females. This sex difference is the result of rapid fiber addition in males between the end of metamorphosis, post-metamorphic stage 0 (PM0) and PM2. At PM0, male and female frogs have similar numbers of laryngeal muscle fibers. Males then add more muscle fibers than females and achieve an adult value that is 1.7 times the female number. Males castrated at PM0 have the same fiber number as females. Ovariectomy at PM0 does not alter muscle fiber addition in females. Gonadectomy at PM2 has no effect on fiber addition in either sex. Females attain masculine muscle fiber number if their ovaries are replaced with a testis at metamorphosis. Exogenous testosterone treatment at PM0 significantly increases fiber number in females but not in males. Exogenous testosterone given at PM2 has no effect on fiber number in females but decreases fiber number in males. We conclude that the testes are necessary for the marked addition of laryngeal muscle fibers seen in male X. laevis between PM0 and PM2. The masculine pattern of muscle fiber addition can be induced in females provided with a testis. Androgen secretion from the testes most probably accounts for masculinization of laryngeal muscle fiber number. After PM2, androgens are no longer necessary for muscle fiber addition and cannot increase fiber number in females.     

Extracellular matrix disruption and pain after eccentric muscle action

Pain, stiffness, and indicators of muscle damage occur at different times after eccentric muscle action. After a single bout of maximal resisted lengthening of the elbow flexors, elbow position, pain perception, and indicators of cellular damage were measured. Immediately postexercise, a significant decrease in resting muscle length was observed that continued to 48 h. At this time, an increase in perceived muscle soreness was noted (P less than 0.05), and a biopsy of the biceps brachii revealed mast cell degranulation, separations of the extracellular matrix from myofibers, and increased plasma constituents in the extracellular space. It is proposed that myofiber disruption allows intracellular proteins to escape and extracellular proteins and ions to enter, causing swelling, whereas the disrupted extracellular matrix initiates the inflammatory response, which includes the release of mast cell granules seen at 48 h postexercise. Thus the delayed sensation of pain (soreness) after repeated eccentric muscle actions probably results from inflammation in response to extracellular matrix disruption.     

Characterization and affinity cross-linking of receptors for human recombinant lymphotoxin (tumor necrosis factor-beta) on a human histiocytic lymphoma cell line, U-937

Recombinant human lymphotoxin (rhLT) expressed in a mammalian cell line was purified and used to examine its receptors on the human histiocytic lymphoma cell line U-937. rhLT was radioiodinated by the IODO-GEN method to a specific activity of 60 microCi/micrograms; the labeled protein was biologically active in the cytolytic assay, and displaceable binding to U-937 cells was observed. The specific binding reached a plateau within 10, 60, and 180 min at 37, 23, and 4 degrees C, respectively. Scatchard analysis of the binding data revealed the presence of a single class of high affinity receptors with an apparent Kd of 0.6 nM and a capacity of 33,000 +/- 7,000 binding sites/cell. The binding of 125I-rhLT to U-937 cells could be inhibited by excess unlabeled rhLT or recombinant human tumor necrosis factor (rhTNF), suggesting a common receptor for both molecules. As competitive inhibitor of the binding, rhTNF was equal in its potency to rhLT. Bacterial derived rhLT lacking carbohydrate was also found equipotent to cell line-derived rhLT for cell binding, indicating that carbohydrate plays no significant role in receptor interaction. Additionally, 125I-rhLT was covalently attached to the cell surface via a bifunctional cross-linking reagent, ethylene glycol bis(succinimidyl succinate), solubilized, and analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The cross-linking of the receptor to rhLT revealed two distinct bands at approximate molecular masses of 100,000 and 120,000 daltons. Both bands were absent when unlabeled rhLT or rhTNF was used for competition, indicating the specificity. Affinity cross-linking of U-937 cells with 125I-rhTNF, however, provided only a single band with a molecular mass of about 100,000 daltons. These results suggest that the manner in which rhLT interacts with its receptor is perhaps somewhat different from that of rhTNF.

Zusammenfassung Mit Hilfe eines Stopfpräparates () wurde die Aggressivität von Neuntötern in einer Entfernung von 10–20 m vom Nest gestestet. In jedem Brutabschnitt gab es , die die Attrappe ignorierten (Intensität=0) oder sich nur annäherten (1 und 2), während andere den Balg schwach (3) oder heftig (4 und 5) attackierten. zeigten bis auf zwei Ausnahmen keine Angriffstendenz. Während der Nestbauphase und der Eiablage waren die Attacken durchschnittlich heftiger als in der Inkubationsperiode; zur Zeit der Jungenbetreuung fiel die Angriffsneigung weiter ab (Abb.). In den heftigen Reaktionen der in der fertilen Periode ihrer sehen wir eine Strategie zur Verhinderung von Fremdkopulationen.

Aggression of Red-backed Shrike (Lanius collurio) against a stuffed male

Summary Aggression of Redbacked Shrikes was tested with a stuffed male which was put up in most cases in a distance of 10–20 m from nest. In each breeding stage some males ignored the dummy (intensity scale 0) or only approached it (1 and 2), while others attacked it weakly (3) or heavily (4 and 5). With two exceptions females took no notice of the stuffed male. On average the reactions in nestbuilding and laying time were stronger than those during incubation; the decline continues in the nestling stage (Fig.). The violence of males in the fertile period of their females may be a method to prevent extra-pair copulations.

The minimum requirements for the keeping of crocodiles in private and public institutions are set up by a collective of authors. Important aspects such as the design of enclosures, secure handling and keeping, thermoregulation, protection of animals, feeding, behavioural enrichment and diseases and their prophylaxis are important topics. The authors critically deal with existing guidelines and give suggestions on how crocodiles can be kept appropriately and according to existing laws at the same time.Special attention is given to the aspects of current legislation. It is stressed that there should be given proper attention to the qualification of the keeping institution as to the biology of the animals as well as to security aspects.A table of species completes the text with a short array of important features to be considered in the keeping of crocodiles.
