We describe an easy method to test odour detection and recognition in 1‐d‐old zebra finch hatchlings (Taeniopygia guttata). Day‐old chicks beg in a stereotypical posture, which can be induced by directing gentle puffs of air from a plastic wash bottle near the face. We used this method to experimentally test whether begging duration of chicks was indicative of nest odour recognition. We manipulated the olfactory environment of 12 nests throughout incubation and hatching with either an artificial odour (orange oil) or with a neutral control (tap water). We then presented these two stimulus odours to 25 day‐old chicks and measured the duration of the first begging bout exhibited for each odour. Zebra finches hatched in a nest environment enriched with orange oil scent begged significantly longer when exposed to orange oil odour, compared to control hatchlings. Our simple testing procedure can be used to efficiently quantify odour recognition and/or preference in altricial songbirds at a very early developmental stage.  相似文献

The Caenorhabditis elegans intestinal lumen is surrounded by a dense cytoplasmic network that is laterally attached to the junctional complex and is referred to as the endotube. It localizes to the terminal web region which anchors the microvillar actin filament bundles and is particularly rich in intermediate filaments. To examine their role in intestinal morphogenesis and function, C. elegans reporter strains were generated expressing intestine-specific CFP-tagged intermediate filament polypeptide IFB-2. When these animals were treated with dsRNA against intestinal intermediate filament polypeptide IFC-2, the endotube developed multiple bubble-shaped invaginations that protruded into the enterocytic cytoplasm. The irregularly widened lumen remained surrounded by a continuous IFB-2::CFP-labeled layer. Comparable but somewhat mitigated phenotypic changes were also noted in wild-type N2 worms treated with ifc-2 (RNAi). Junctional complexes were ultrastructurally and functionally normal and the apical domain of intestinal cells was also not altered. These observations demonstrate that IFC-2 is important for structural maintenance of the intestinal tube but is not needed for establishment of the endotube and epithelial cell polarity.  相似文献

The ongoing climate change affects biogeochemical cycling in terrestrial ecosystems, but the magnitude and direction of this impact is yet unclear. To shed further light on the climate change impact, we investigated alterations in the soil nitrogen (N) cycling in a Danish heathland after 5 years of exposure to three climate change factors, i.e. warming, elevated CO2 (eCO2) and summer drought, applied both in isolation and in combination. By conducting laboratory 15N tracing experiments we show that warming increased both gross N mineralization and nitrification rates. In contrast, gross nitrification was decreased by eCO2, an effect that was more pronounced when eCO2 was combined with warming and drought. Moreover, there was an interactive effect between the warming and CO2 treatment, especially for N mineralization: rates increased at warming alone but decreased at warming combined with eCO2. In the full treatment combination, simulating the predicted climate for the year 2075, gross N transformations were only moderately affected compared to control, suggesting a minor alteration of the N cycle due to climate change. Overall, our study confirms the importance of multifactorial field experiments for a better understanding of N cycling in a changing climate, which is a prerequisite for more reliable model predictions of ecosystems responses to climate change.  相似文献

In this work we model the extent of hypoxia in the diabetic retina as a function of the area affected by vessel disruption. We find two regimes that differ on the ratio between the area of disrupted vasculature and the area of tissue in hypoxia. In the first regime the hypoxia is localized in the vicinity of the vascular disruption, while in the second regime there is a generalized hypoxia in the affected tissue. The transition between these two regimes occurs when the tissue area affected by individual sites of vessel damage is on the order of the square of the characteristic irrigation length in the tissue (the maximum distance that an irrigated point in the tissue is from an existing vessel). We observe that very high levels of hypoxia are correlated with the rupture of larger vessels in the retina, and with smaller radii of individual sites of vessel damage. Based on this property of vascular networks, we propose a novel mechanism for the transition between the nonproliferative and the proliferative stages in diabetic retinopathy.  相似文献

Skeletal muscles are surrounded by other muscles, connective tissue and bones, which may transfer transversal forces to the muscle belly. Simple Hill-type muscle models do not consider transversal forces. Thus, the aim of this study was to examine and model the influence of transversal muscle loading on contraction dynamics, e.g. on the rate of force development and on the maximum isometric muscle force (Fim). Isometric experiments with and without transversal muscle loading were conducted on rat muscles. The muscles were loaded (1.3 N cm? 2) by a custom-made plunger which was able to move in transversal direction. Then the muscle was fully stimulated, the isometric force was measured at the distal tendon and the movement of the plunger was captured with a high-speed camera. The interaction between the muscle and the transversal load was modelled based on energy balance between the (1) work done by the contractile component (CC) and (2) the work done to lift the load, to stretch the series elastic structures and to deform the muscle. Compared with the unloaded contraction, the force rate was reduced by about 25% and Fim was reduced by 5% both in the experiment and in the simulation. The reduction in Fim resulted from using part of the work done by the CC to lift the load and deform the muscle. The response of the muscle to transversal loading opens a window into the interdependence of contractile and deformation work, which can be used to specify and validate 3D muscle models.  相似文献
