We have carried out detailed structural studies of the glycopeptides of glycoprotein gD of herpes simplex virus types 1 and 2. We first examined and compared the number of N-asparagine-linked oligosaccharides present in each glycoprotein. We found that treatment of either pgD-1 or pgD-2 with endo-β-N-acetylglucosaminidase H (Endo H) generated three polypeptides which migrated more rapidly than pgD on gradient sodium dodecyl sulfate-polyacrylamide gels. Two of the faster-migrating polypeptides were labeled with [3H]mannose, suggesting that both pgD-1 and pgD-2 contained three N-asparagine-linked oligosaccharides. Second, we characterized the [3H]mannose-labeled tryptic peptides of pgD-1 and pgD-2. We found that both glycoproteins contained three tryptic glycopeptides, termed glycopeptides 1, 2, and 3. Gel filtration studies indicated that the molecular weights of these three peptides were approximately 10,000, 3,900, and 1,800, respectively, for both pgD-1 and pgD-2. Three methods were employed to determine the size of the attached oligosaccharides. First, the [3H]mannose-labeled glycopeptides were treated with Endo H, and the released oligosaccharide was chromatographed on Bio-Gel P6. The size of this molecule was estimated to be approximately 1,200 daltons. Second, Endo H treatment of [35S]methionine-labeled glycopeptide 2 reduced the molecular size of this peptide from approximately 3,900 to approximately 2,400 daltons. Third, glycopeptide 2 isolated from the gD-like molecule formed in the presence of tunicamycin was approximately 2,200 daltons. From these experiments, the size of each N-asparagine-linked oligosaccharide was estimated to be approximately 1,400 to 1,600 daltons. Our experiments indicated that glycopeptides 2 and 3 each contained one N-asparagine-linked oligosaccharide chain. Although glycopeptide 1 was large enough to accommodate more than one oligosaccharide chain, the experiments with Endo H treatment of the glycoprotein indicated that there were only three N-asparagine-linked oligosaccharides present in pgD-1 and pgD-2. Further studies of the tryptic glycopeptides by reverse-phase high-performance liquid chromatography indicated that all of the glycopeptides were hydrophobic in nature. In the case of glycopeptide 2, we observed that when the carbohydrate was not present, the hydrophobicity of the peptide increased. The properties of the tryptic glycopeptides of pgD-1 were compared with the properties predicted from the deduced amino acid sequence of gD-1. The size and amino acid composition compared favorably for glycopeptides 1 and 2. Glycopeptide 3 appeared to be somewhat smaller than would be predicted from the deduced sequence of gD-1. It appears that all three potential glycosylation sites predicted by the amino acid sequence are utilized in gD-1 and that a similar number of glycosylation sites are present in gD-2.  相似文献

The effects of active ionic transport are included in the derivation of a general expression for the zero current membrane potential. It is demonstrated that an active transport system that transfers no net charge (nonrheogenic) may, nevertheless, directly alter the membrane potential. This effect depends upon the exchange of matter within the membrane between the active and passive diffusion regimes. Furthermore, in the presence of such exchange, the transmembrane active fluxes measured by the usual techniques and the local pumped fluxes are not identical. Several common uses of the term “electrogenic pump” are thus shown to be inconsistent with each other. These inconsistencies persist when the derivation is extended to produce a Goldman equation modified to account for active transport; however, that equation is shown to be limited by less narrow constraints on membrane heterogeneity and internal electric field than those previously required. In particular, it is applicable to idealized mosaic membranes limited by these requirements.  相似文献

The outer solar system contains many environments of interest for studies of the origin of life. Recent observations support the idea that Jupiter and Saturn have retained the mixture of elements originally present in the solar nebula. Subsequent low temperature chemistry has produced the expected array of simple molecules giving characteristic absorption bands in the spectra of these planets. Microwave and infrared observations show that the lower atmospheres are at temperatures above 300 K. Sources of energy for non-equilibrium chemistry seem available at least on Jupiter and the presence of an array of colored materials in the Jovian cloud belts has often been cited as evidence for the existence of complex abiogenic organic molecules. Further study of both planets in an exobiological context seems well worthwhile; potentially productive methods of investigation (including planned space missions) can be described and evaluated from this point of view. Uranus and Neptune are clearly deficient in light gases, but otherwise little is known with certainty about these distant planets. Again unusually high temperatures have been reported, but not above 273 K. Pluto and many of the outer planet satellites appear to represent a class of small bodies very unlike our neighbors in the inner solar system. Titan, Saturn's largest satellite, is especially interesting for our purposes because of its atmosphere. Methane and hydrogen are both present, and Titan's unusually reddish color again suggests the presence of organic compounds. The hydrogen-methane ratio is likely to be more similar to that of a primitive reducing terrestrial atmosphere than the ratios for Jupiter and Saturn, suggesting that in some respects this satellite may provide an even better model for early organic synthesis on the Earth. The problem of Titan's heat balance and atmospheric composition are currently under active investigation.  相似文献

The synthesis of desthiobiotin from 7,8-diaminopelargonic acid (DAP) was demonstrated in resting cell suspensions of Escherichia coli K-12 bioA mutants under conditions in which the biotin locus was derepressed. The biosynthetically formed desthiobiotin was identified by chromatography, electrophoresis, and by its ability to support the growth of yeast and those E. coli biotin auxotrophs that are blocked earlier in the biotin pathway. Optimal conditions for desthiobiotin synthesis were determined. Desthiobiotin synthetase activity was repressed 67% when partially derepressed resting cells were incubated in the presence of 3 ng of biotin per ml. Serine, bicarbonate, and glucose stimulated desthiobiotin synthesis apparently by acting as sources of CO(2). The results of this study are consistent with an earlier postulated pathway for biotin biosynthesis in E. coli: pimelic acid --> 7-oxo-8-aminopelargonic acid --> DAP --> desthiobiotin --> biotin.  相似文献

The theoretical approach presented in the previous paper provides an analytical method for determining the unidirectional, nonsteady-state fluxes in a three compartment system. Based on this a study was made of the sodium flux transients in the toad urinary bladder. A transient time-dependent state was generated by suddenly short-circuiting a bladder previously maintained in an open-circuited steady state. The sequence of experiments suggested by the theory provided the data required for the analysis. The results of these tracer experiments were consistent with the complex non-three compartmental structure of this tissue. As a result both of the inadequacy of the three compartment model in representing the tissue and of certain experimental difficulties, attempts at a quantitative solution were not entirely successful. Useful information was nevertheless obtained through a careful use of this model, and a qualitative analysis implied that the sodium influxes into the tissue at both of its surfaces are sensitive to changes in electrical potential while both effluxes are insensitive to this change. This suggests that both of the effluxes result from active processes while both influxes are associated with passive processes. The net transepithelial transport of sodium would then necessarily result from a more complex polarization than that proposed by Koefoed-Johnsen and Ussing.  相似文献

Two aflatoxin-producing isolates of Aspergillus flavus were grown for 5 days on Wort media at 2, 7, 13, 18, 24, 29, 35, 41, 46, and 52 C. Maximal production of aflatoxins occurred at 24 C. Maximal growth of A. flavus isolates occurred at 29 and 35 C. The ratio of the production of aflatoxin B1 to aflatoxin G1 varied with temperature. Aflatoxin production was not related to growth rate of A. flavus; one isolate at 41 C, at almost maximal growth of A. flavus, produced no aflatoxins. At 5 days, no aflatoxins were produced at temperatures lower than 18 C or higher than 35 C. Color of CHCl3 extracts appeared to be directly correlated with aflatoxin concentrations. A. flavus isolates grown at 2, 7, and 41 C for 12 weeks produced no aflatoxins. At 13 C, both isolates produced aflatoxins in 3 weeks, and one isolate produced increasing amounts with time. The second isolate produced increasing amounts through 6 weeks, but at 12 weeks smaller amounts of aflatoxins were recovered than at 6 weeks.  相似文献
