Investigations on the activation of bovine prochymosin.   总被引:1,自引:0,他引:1  
Activation of prochymosin at pH below 2.5 results in formation of the active enzyme pseudochymosin by proteolytic cleavage of the bond 27--28. Pseudochymosin is 15 amino acid residues longer than chymosin. It is the final activation product at low pH, whereas chymosin is formed by activation between pH 4 and 5. Pseudochymosin is converted to chymosin when it is brought to pH 5.5. Our present knowledge does not allow quantitative evaluation of the possible reactions involved in formation of pseudochymosin, but the course of activation at pH 2 is in accordance with an intermolecular reaction between two zymogen molecules as the predominant reaction. We find indications of an intramolecular reaction when intermolecular reactions are prevented by immobilization of the zymogen.  相似文献

Root segments and root-soil cores (6.5-cm diameter) from fields and nurseries of winter wheat and sorghum were tested for N2 fixation by using the acetylene reduction assay. Wheat samples (approximately 1,200) from 109 sites generally had low or no activity (0 to 3.1 nmol of C2H4 produced per h per g [dry weight] of root segments), even after 24 h of incubation. However, a commercial field of Scout 66, located in western Nebraska, exhibited appreciable activity (290 nmol of C2H4 produced per h per g [dry weight] of root segments). Of 400 sorghum lines and crosses, grain sorghums (i.e., CK-60A, Wheatland A, B517, and NP-16) generally exhibited higher nitrogenase activity than forage sorghums or winter wheats. CK-60A, a male sterile grain sorghum, was sampled at four locations and had the most consistent activity of 24 to 1,100 nmol of C2H4 produced per h per core. The maximum rate extrapolated to 2.5 g of N per hectare per day. Numerous N2-fixing bacterial isolates were obtained from wheat and sorghum roots that exhibited high nitrogenase activity. Most isolates were members of the Enterobacteriacae, i.e., Klebsiella pneumoniae, Enterobacter cloacae, and Erwinia herbicola.  相似文献

Treatment of preimplantation mouse embryos in vitro with 10(-3) to 10(-1) mug actinomycin D/ml for 2 hr showed that (i) postimplantation development in vitro was inhibited most when embryos were treated at the morula stage and (ii) after the morula stage actinomycin D inhibited trophoblast outgrowth less than inner cell mass development.  相似文献

Summary The presence of EF-Tu, RNA polymerase subunit , and EF-G on the dfus-3 genome and EF-Tu, ribosomal proteins L7/L12, and RNA polymerase subunit on the drif d 18 genome has been confirmed using a two-dimensional gel electrophoresis technique sensitive to changes in isoelectric point and molecular weight. In this system two EF-Tu gene products could not be resolved. Following infection of ultraviolet light-irradiated Escherichia coli with either dfus-3 or drif d 18, the EF-Tu gene, tufA, near 65 minutes on the genetic map is expressed as 3–4 copies per EF-G molecule. The EF-Tu gene, tufB, near 79 minutes on the genetic map, is expressed at about one-third of this rate. is expressed as 1 copy per EF-G molecule, as 0.14 per EF-G molecule and L7/L12 as 2.5 per EF-G. These figures compare well with the relative amounts found in exponentially-growing cells, in which the ratio of EF-Tu to EF-G is approximately 5. Almost 90% of the total number of proteins (calculated on a molecular weight basis) which theoretically can be encoded on the drif d 18 have been identified on the two-dimensional gel.  相似文献

In vivo measurement of the oxygen saturation levels in blood may be obtained from relative amounts of backscattered monochromatic light at two different wavelengths, as measured with a fiber-optic catheter oximeter. Because of the short mean free path length of light in blood, the backscattering can be well approximated by a previously-derived, one-wavelength transport theory solution for the half-space searchlight problem. This solution, unlike simple diffusion approximations has the advantage that the boundary condition describing illumination of a localized area of blood by a monodirectional light beam can be rigorously satisfied. Sample calculations using the solution are compared with experimental values of the reflectance of blood.  相似文献

Raman difference spectrophotometry reveals that CH3HgII binds quantitatively to N(1) of inosine at pH 8, substituting for the proton. When N(1) is saturated, binding occurs at a second site. Measurements of the 1-H nuclear magnetic resonance spectra of both inosine and of CH3Hg-II are in agreement with the N(1) binding and indicate that the second site for mercuriation is N(7). This second binding reaction is observed to increase the rate of exchange of the C(8) hydrogen with solvent, consistent with results observed for alkylation at N(7). Coordination of the electrophilic CH3Hg-II to N(7) increases the acidity of H(8), facilitating OHminus--catalyzed proton abstraction and reprotonation by themedium. For comparison, the reaction of CH3Hg-II with [8-2-H]inosine has been studied. Displacement of the N(1) hydrogen upon mercuriation of inosine causes a significant electron delocalization into the ring, increasing the basicity of N(7), and accounting for the synergic effect in metal binding observed originally by Simpson. In contrast, 1-methylinosine interacts only slightly with CH3Hg-II at pH 8. Coordination appears to be at N(7), since H(8) again is observed to exchange rapidly with solvent protons. In acidic solution, pH less than 2, binding to inosine is almost quantitative and exclusively to N(7). The behavior of CH3Hg-II is compared with that of Pt(II) and with Ni(II), Co(II), AND Zn(II). A brief comparison is made among ultraviolet absorption spectrophotometry, nuclear magnetic resonance (NMR), and Raman difference spectrophotometry for studying reactions of nucleosides and nucleotides.  相似文献

Three ATP-dependent reactions catalyzed by the inner membrane of rat liver mitochondria and the ATPase reaction catalyzed by purified mitochondrial ATPase (F1), were studied with respect to kinetic properties, substrates specificity, and sensitivity to bicarbonate. The ATP-dependent transhydrogenase reaction (reduction of NADP+ by NADH) catalyzed by inner membrane vesicles displays typical Michaelis-Menten kinetics in both Tris-Cl and Tris-bicarbonate buffers, with Km (ATP) values of 0.035 mM and 0.054 mM respectively. The Vmax of transhydrogenase activity (25 nmol min-1 mg-1) is the same in Tris-bicarbonate or Tris-Cl buffer. ITP and GTP readily substitute for ATP in the transhydrogenase reaction. The ATP-P1 exchange reaction catalyzed by inner membrane vesicles displays typical Michaelis-Menten kinetics in both Tris-Cl and Tris-bicarbonate buffers with Km (ATP) values of 1.0 mM and 1.4 mM respectively. The Vmax of exchange (200 nmol min-1 mg-1) is the same in either buffer. ITP and GTP do not effectively replace ATP in the exchange reaction.  相似文献

Interplanetary space contains fluxes of fast moving atomic nuclei. The distribution of these reflects the atomic composition of the universe, and such particles may pose limitations for space flight and for life in space. Over the past 50 years, since the invention of Ernest Lawrence's cyclotron, advances in accelerator technology have permitted the acceleration of charged nuclei to very high velocities. Currently, beams of any stable isotope species up to uranium are available at kinetic energies of several hundred MeV/nucleon at the Berkeley Bevalac. Recently, new areas of particle physics research relating to the mechanisms of spallation and fission have opened up for investigation, and it is now realistic to search for nuclear super-dense states that might be produced in heavy nuclear collisions. The heavy ions hold interest for a broad spectrum of research because of their effectiveness in producing a series of major lesions in DNA along single particle tracks and because of the Bragg depth ionization properties that allow the precise deposition of highly localized doses deep in the human body. Individual heavy ions can also interrupt the continuity of membraneous regions in cells. Heavy ions, when compared to low-LET radiation, have increased effectiveness for mammalian cell lethality, chromosome mutations, and cell transformation. The molecular mechanisms are not completely understood but appear to involve fragmentation and reintegration of DNA. Cells attempt to repair these lesions, and many of the deleterious effects are due to misrepair or misrejoining of DNA. Heavy ions do not require the presence of oxygen for producing their effects, and hypoxic cells in necrotic regions have nearly the same sensitivity as cells in well-oxygenated tissues. Heavy ions are effective in delaying or blocking the cell division process. Heavy ions are also strong enhancers of viral-induced cell transformation, a process that requires integration of foreign DNA. Some cell lines, known to be radioresistant to X rays, have exhibited greater sensitivity to heavy ions. These radiobiological properties, combined with the ability to deliver highly localized internal doses, make accelerated heavy ions potentially important radiotherapeutic tools. Other novel approaches include the utilization of radioactive heavy beams as instant tracers. Heavy-ion radiography and microscopy respond to delicate changes in tissue electron density. Dose localization with helium ions has achieved excellent results for pituitary tumors, tumors adjacent to the spinal cord, and ocular melanomas. Two little known species of brown algae, Litosiphon mortensenii S. Lund and L. groenlandicus S. Lund, described from East Greenland have been compared with the type species of the genera Litosiphon Harv. and Asperococcus Lamour. They are distinguished from the genus Litosiphon by their basally sheathed hairs, hecatonematoid microthalli, mediate differentiation of embryospores, and lack of sympodial branching of the macrothallus. They are considered more primitive than Asperococcus by the absence of paraphyses and by the non-localized formation of reproductive structures. The new genus Trachynema is proposed to accommodate these plants; it finds a natural position in the Punctariaceae.  相似文献
