Nitrile groups are catabolized to the corresponding acid and ammonia through one-step reaction involving a nitrilase. Here,
we report the use of bioinformatic and biochemical tools to identify and characterize the nitrilase (NitPf5) from Pseudomonas fluorescens Pf-5. The nitPf5 gene was identified via sequence analysis of the whole genome of P. fluorescens Pf-5 and subsequently cloned and overexpressed in Escherichia coli. DNA sequence analysis revealed an open-reading frame of 921 bp, capable of encoding a polypeptide of 307 amino acids residues
with a calculated isoelectric point of pH 5.4. The enzyme had an optimal pH and temperature of 7.0°C and 45°C, respectively,
with a specific activity of 1.7 and 1.9 μmol min−1 mg protein−1 for succinonitrile and fumaronitrile, respectively. The molecular weight of the nitrilase as determined by sodium dodecyl
sulfate-polyacrylamide gel electrophoresis and gel filtration chromatography was 33,000 and 138,000 Da, respectively, suggesting
that the enzyme is homotetrameric. Among various nitriles, dinitriles were the preferred substrate of NitPf5 with a Km = 17.9 mM and kcat/Km = 0.5 mM−1 s−1 for succinonitrile. Homology modeling and docking studies of dinitrile and mononitrile substrate into the active site of
NitPf5 shed light on the substrate specificity of NitPf5. Although nitrilases have been characterized from several other sources,
P. fluorescens Pf-5 nitrilase NitPf5 is distinguished from other nitrilases by its high specific activity toward dinitriles, which make
P. fluorescens NitPf5 useful for industrial applications, including enzymatic synthesis of various cyanocarboxylic acids. 相似文献
Effect of methylprednisolone sodium succinate (MPSS) and its comparison with dexamethasone in experimentally induced acute spinal cord compression in adult rats was studied. The rats were divided into group A (control) and group B, which was subdivided into B1, B2, B3 where MPSS was given after 1, 8 and 24 hr and B4 where dexamethasone was given after 1 hr of cord injury respectively. Proper neurological evaluation was done with mobility, running and climbing score. Recovery index was evaluated for 7 days. After sacrificing the rats, spinal cord was observed histopathologically. Mean recovery index and microscopic findings based on hemorrhage in gray and white matter, neuronal degeneration, hematomyelia and edema in white matter were recorded. The results suggested that MPSS was effective in promoting post-traumatic clinical and histological recovery and to a greater extent, when given 1 hr after trauma. MPSS is more effective than dexamethasone in reducing edema when both are given after interval of 1 hr. 相似文献
Riparian zones are among the most valuable ecosystems on the earth. They act as the ecological engineers that improve river health through delivering a range of ecosystem functions. Stream bank stabilization, pollutant and sediment buffering, temperature regulation, provision of energy to river food webs and communities, groundwater recharge and provision of ecological corridors and habitat for wildlife, are among major ecosystem functions of riparian zones that play a great role in river health. Besides these ecosystem functions, riparian zones also provide various ecosystem goods and services for human well-being. But in the current scenario, riparian zones are under severe threat due to agricultural activities, urbanization, river flow alteration, overexploitation, climate change, pollution, and biological invasion. In the present and probable future scenarios of declining river health and global environmental changes, there is a pressing need of an integrated approach for managing riparian zones. This review article aims to advocate an integrated approach for riparian zone management based on various components such as riparian condition assessment, policy framework, stakeholder’s participation, management practices, legislation, and awareness. Authors also discussed riparian zones in context of their concepts, features, functions, and threats.
Active compound oleandrin extracted from Nerium indicum (Lal Kaner) leaf has potent piscicidal activity. The piscicidal activity of oleandrin on freshwater fish C. punctatus was both time and dose dependent. Exposure to sub-lethal doses of oleandrin for 24hr and 96hr to fish caused significant alteration in the level of total protein, total free amino acid, nucleic acid, glycogen, pyruvate, lactate and enzyme protease, phosphatases, alanine aminotransferase, aspartate aminotransferase and acetylcholinesterase activity in liver and muscle tissues. The alterations in all the above biochemical parameters were also significantly time and dose dependent. The results show a significant recovery in all the above biochemical parameters, in both liver and muscle tissues of fish after the 7th day of the withdrawal of treatment. Toxicity persistence test of oleandrin on juvenile Labeo rohita shows that fish seed of common culturing carp can be released into rearing ponds after three days of oleandrin treatment. It supports the view that the oleandrin is safer and may be useful substitute of other piscicides for removing the unwanted freshwater fishes from aquaculture ponds. 相似文献
The effect of elevated carbon dioxide (600±50 cm3 m−3; C600) on growth performance, biomass production, and photosynthesis of Cenchrus ciliaris L. cv. 3108 was studied. This crop responded significantly by plant height, leaf length and width, and biomass production
under C600. Leaf area index increased triple fold in the crops grown in the open top chamber with C600. The biomass production in term of fresh and dry biomass accumulation increased by 134.35 (fresh) and 193.34 (dry) % over
the control (C360) condition where the crops were grown for 20 d. The rate of photosynthesis and stomatal conductance increased by 24.51 and
46.33 %, respectively, in C600 over C360 plants. In comparison with C360, the rate of transpiration decreased by 6.8 % under C600. Long-term exposure (120 d) to C600 enhanced photosynthetic water use efficiency by 34 %. Also the contents of chlorophylls a and b significantly increased in C600. Thus C. ciliaris grown in C600 throughout the crop season may produce more fodder in terms of green biomass. 相似文献
A rapid method of ultracentrifugation pelleting of avian adenovirus (AAV) from small volume of chloroform treated infected cell culture fluid or allantoic fluid was adapted for isolation of adenoviral DNA. The viral DNA extracted from semipurified viruses was found to be intact on agarose gel and pure enough (A260/280 = 1.85-1.92) for restriction enzyme analysis. Restriction endonuclease analysis of Indian strain of AAV serotype 1, AAV serotype 4 (group I AAVs) and egg drop syndrome-76 (EDS-76) virus genomes (group III AAV) with Hind III enzyme differentiated these viruses. The AAV serotype 1 and serotype 4 strain exhibited identical Hind III profile to European viral strains belonging to same serotypes however, the EDS-76 virus gave similar but not identical profile. The calculated genomic lengths for AAV serotype 1 and EDS-76 virus were approximately found to be 33.9 and 44.4 Kb, respectively. 相似文献
The non-coding fraction of the human genome, which is approximately 98%, is mainly constituted by repeats. Transpositions, expansions and deletions of these repeat elements contribute to a number of diseases. None of the available databases consolidates information on both tandem and interspersed repeats with the flexibility of FASTA based homology search with reference to disease genes. Repeats in diseases database (RiDs db) is a web accessible relational database, which aids analysis of repeats associated with Mendelian disorders. It is a repository of disease genes, which can be searched by FASTA program or by limitedor free- text keywords. Unlike other databases, RiDs db contains the sequences of these genes with access to corresponding information on both interspersed and tandem repeats contained within them, on a unified platform. Comparative analysis of novel or patient sequences with the reference sequences in RiDs db using FASTA search will indicate change in structure of repeats, if any, with a particular disorder. This database also provides links to orthologs in model organisms such as zebrafish, mouse and Drosophila.
