We examined the accuracy of pulmonary cytology in 224 consecutive patients being evaluated for lung cancer. The diagnostic yeild of specimens obtained by various methods, including flexible fiber optic bronchoscopy (FFB), was compared. Among 69 patients with lung cancer, a cytologic diagnosis was made in 87%, including 73% with peripheral tumors. Prebronchoscopy sputa were positive in 50%, bronchial washings in 63%, postbronchoscopy sputa in 82% and bronchial brushings in 59% of the patients. In only one patient was the bronchial brush specimen the only positive cytologic specimen. Normal FFB and small cell undifferentiated cancer were found with increased frequency (P less than 0.05) among the nine patients (13%) with false-negative cytology. Among 155 patients with nonmalignant lung disease, 16 (10%) had false-positive specimens; this finding was significantly related (P less than 0.05) to necrotizing pneumonia in 13 of the 16 patients (81%). The overall diagnostic accuracy of cytology showed 87% sensitivity and 90% specificity, and the predictive value of a positive specimen was 79%. We have determined the mode of cleavage of superhelical SV40 DNA (Form I) by restriction endonucleases EcoRI and HpaII at 37 degrees C. By analysis with agarose gel electrophoresis and direct examination with dark field electron microscopy, we found that a large amount of the single-nicked circular DNA (Form II) was produced before the linear SV40 DNA (Form III) appeared. Thus, both restriction enzymes cleave only one strand of the superhelical DNA first. The second cleavage on the complementary strand occurred after a lag period. The first order rate constant for the second cleavage by EcoRI endonuclease was determined and a kinetic reaction scheme for both enzymes is proposed. 相似文献

When slices prepared from rat corpus striatum were preincubated for 15 min in potassium-enriched Krebs Ringer-Phosphate medium (K+-KRP), the activity of glutamic acid decarboxylase measured upon reincubation in normal Krebs-Ringer-Phosphate (KRP) was doubled as compared to GAD activity in slices preincubated in normal KRP. Similarly, when striatal slices were preincubated in KRP containing 100 μM veratridine, GAD activity upon reincubation in normal KRP was increased 66% as compared to activity in slices preincubated in normal KRP. The observed increase in GAD activity was not a function of alterations in glutamate uptake by the slices. These results suggest that GABAergic neurons may regulate transmitter synthesis during the process of depolarization by increasing GAD activity. 相似文献

Colony formation by variant Chinese hamster cells highly resistant to adenine analogs and deficient in adenine phosphoribosyltransferase (APRT) activity was measured after co-cultivation with APRT+, CHO-K1 cells in medium containing one of three different adenine analogs. Depending upon the density of APRT+ cells and the specific adenine analog, large differences in the recovery of APRT? colonies were observed. The particular adenine analog and APRT+ cell density were more significant factors in the recovery of APRT? colonies than the concentration of the analog or the level of APRT activity. The number of wild-type cells (CHO-K1) required to inhibit formation of APRT? colonies by 50% (mean lethal density; MLD50) with 65 μg/ml 8-aza-adenine (AzA) as the selective drug was 8.0 × 105 cells/100 mm dish (1.5 × 104/cm2). With 100 μg/ml 2,6-diaminopurine (DAP) the MLD50 for CHO-K1 was 4.0 × 105 cells/100 mm dish (7.3 × 103/cm2). The MLD50 for CHO-K1 when the DAP concentration was decreased to 50 μg/ml was only slightly higher, 5 × 105 cells/100 mm dish (9.1 × 103/cm2). The most toxic effect was observed with 2-fluoroadenine (FA). The MLD50 for CHO-K1 in 2 μg/ml FA was 4.5 × 104 cells/100 mm dish (8.2 × 102/cm2), a cell density which permits minimal direct contact between APRT+ and APRT? cells. The toxic effects of FA on individually resistant, APRT? cells were found to be mediated by metabolites released into the medium by dying APRT+ cells. This metabolite toxicity to APRT? cells was also demonstrated in mixtures with cells having only 8% of wild-type APRT activity. The MLD50 for these APRT+ (8%) cells in 2 μg/ml FA was 7.5 × 104 cells/100 dish (1.4 × 103/cm2), a small difference from the MLD50 for cells with wild-type levels of APRT activity. The differences in the recovery of APRT? colonies from mixtures with APRT+ cells in these three adenine analogs are critical to the design of procedures for the selection of APRT? cells from populations of APRT+ cells and emphasize the importance of establishing the parameters of metabolic cooperation, not only in terms of cell density but also with regard to the particular selective agent, in any experiment designed to determine precise mutation rates or to test putative mutagens upon mammalian cells in culture. 相似文献

Identification of a transformation-specific protein induced by a Rous sarcoma virus. 总被引:15,自引:0,他引:15
Using antisera obtained from rats bearing Schmidt-Ruppin strain Rous sarcoma virus-induced tumors, we have idnetified a protein with an apparent molecular weight of 56,000 daltons and an isoelectric point of 6.3 in extracts of chick embryo fibroblasts transformed by a wild-type nondefective Rous sarcoma virus (Schmidt-Ruppin strain). This protein was not found in cells infected by trnasformation-defective mutants with either a partial or complete deletion of the src gene, nor in cells infected by a nontransforming avian leukosis virus. The 56,000 dalton molecular weight protein was found to be synthesized at both the permissive and nonpermissive temperatures in cells infected by either of two conditionallethal mutants that are temperature-sensitive in cell transformation. The amount of this protein, however, accumulated in cells infected by these temperature-sensitive mutants, relative to the structural polypeptides, differed significnatly from that seen with the nondefective virus. Pulsechase experiments indicate that the protein is extremely unstable, with a half-life of about 20 min, and does not serve as a precursor to any of the detectable virion polypeptides. Furthermore, incubation of the rat antiserum with purified, disrupted virus did not affect its immunoreactivity to this particular protein. We conclude that this 56,000 dalton molecular weight protein is a nonstructural protein specific to cells transformed by Rous sarcoma virus. 相似文献

Preparations of human malignant effusion galactosyltransferase activity purified according to previously published techniques using enzyme-specific affinity chromatography consistently produced antibodies directed toward immunoglobulins with no detectable antigalactosyltransferase. Double immunodiffusion analysis of the antigen showed the presence of both IgG and IgA. Affinity chromatography with anti-human IgG-Sepharose and anti-human serum-Sepharose resulted in a 48,000-fold purification of galactosyltransferase activity with no detectable IgG by radioimmunoassay. Immunization of rabbits with this preparation produced antibodies directed against galactosyltransferase activity and minimal anti-Ig. The persistence of immunoglobulins during the purification of soluble galactosyltransferase activity through two enzyme-specific affinity chromatographic steps suggests an association of immunoglobulins with galactosyltransferase activity. 相似文献

Treatment with PGF2α resulted in the termination of pregnancy in 16- and 19-day pregnant rats, but not in 10- or 13-day pregnant rats. Rats that aborted displayed a rapid onset of maternal behavior when tested with foster pups. Aborted rats also displayed sexual receptivity and ovulation: these phenomena resemble the sequence of events following hysterectomy on the same days of pregnancy. Both can be related to the events surrounding normal parturition in the rat. The results are interpreted as due to a pregnancy-induced deactivation of the factor in the uterus that prevents estrogen from stimulating maternal behavior in nonpregnant females. In the absence of this factor, the PGF2α-induced rise in estrogen secretion facilitates maternal behavior and sexual behavior and induces ovulation. 相似文献

The nondefective adenovirus type 2 (Ad2)-simian virus 40 (SV40) hybrid viruses, Ad2+ND2 and Ad2+ND4, have been used to determine which regions of the SV40 genome coding for the large tumor (T) antigen are involved in specific and nonspecific DNA binding. Ad2+ND2 encodes 45,000 M4 (45K) and 56,000 Mr (56K) T antigen-related polypeptides. The 45K polypeptide did not bind to DNA, but the 56K polypeptide bound nonspecifically to calf thymus DNA, Ad2+ND4 encodes 50,000 Mr (60K), 66,000 Mr (66K), 70,000 Mr (70K), 74,000 Mr (74K), and 90,000 Mr (90K) T antigen-related polypeptides, all of which bound nonspecifically to calf thymus DNA. However, in more stringent assays, where tight binding to viral origin sequences was tested, only the 90K protein specified by Ad2A+ND4 showed specific high affinity for sequences at the viral origin of replication. From these results and previously published experiments describing the SV40 DNA integrated into these hybrid viruses, it was concluded that SV40 early gene sequences located between 0.39 and 0.44 SV40 map units contribute to nonspecific DNA binding, whereas sequences located between 0.50 and 0.63 SV40 map units are necessary for specific binding to the viral origin of replication. 相似文献
