Age‐related diseases such as cancer, cardiovascular disease, kidney failure, and osteoarthritis have universal features: Their incidence rises exponentially with age with a slope of 6–8% per year and decreases at very old ages. There is no conceptual model which explains these features in so many diverse diseases in terms of a single shared biological factor. Here, we develop such a model, and test it using a nationwide medical record dataset on the incidence of nearly 1000 diseases over 50 million life‐years, which we provide as a resource. The model explains incidence using the accumulation of senescent cells, damaged cells that cause inflammation and reduce regeneration, whose level rise stochastically with age. The exponential rise and late drop in incidence are captured by two parameters for each disease: the susceptible fraction of the population and the threshold concentration of senescent cells that causes disease onset. We propose a physiological mechanism for the threshold concentration for several disease classes, including an etiology for diseases of unknown origin such as idiopathic pulmonary fibrosis and osteoarthritis. The model can be used to design optimal treatments that remove senescent cells, suggeting that treatment starting at old age can sharply reduce the incidence of all age‐related diseases, and thus increase the healthspan. 相似文献

Mesophotic coral ecosystems (MCEs) host a thriving community of biota that has remained virtually unexplored. Here we report for the first time on a large population of the endangered coral species Euphyllia paradivisa from the MCEs of the Gulf of Eilat/Aqaba (GOE/A), Red Sea. The mesophotic zone in some parts of the study site harbors a specialized coral community predominantly comprising E. paradivisa (73 % of the total coral cover), distributed from 36 to 72 m depth. Here we sought to elucidate the strict distribution but high abundance of E. paradivisa in the MCEs at the GOE/A. We present 4 yr of observations and experiments that provide insight into the physiological plasticity of E. paradivisa: its low mortality rates at high light intensities, high competitive abilities, successful symbiont adaptation to the shallow-water environment, and tolerance to bleaching conditions or survival during prolonged bleaching. Despite its ability to survive under high irradiance in shallow water, E. paradivisa is not found in the shallow reef of the GOE/A. We suggest several factors that may explain the high abundance and exclusivity of E. paradivisa in the MCE: its heterotrophic capabilities; its high competition abilities; the possibility of it finding a deep-reef refuge there from fish predation; and its concomitant adaptation to this environment. 相似文献

Monte Carlo simulation (MCS) is a common methodology to compute pathways and thermodynamic properties of proteins. A simulation run is a series of random steps in conformation space, each perturbing some degrees of freedom of the molecule. A step is accepted with a probability that depends on the change in value of an energy function. Typical energy functions sum many terms. The most costly ones to compute are contributed by atom pairs closer than some cutoff distance. This paper introduces a new method that speeds up MCS by exploiting the facts that proteins are long kinematic chains and that few degrees of freedom are changed at each step. A novel data structure, called the ChainTree, captures both the kinematics and the shape of a protein at successive levels of detail. It is used to efficiently detect self-collision (steric clash between atoms) and/or find all atom pairs contributing to the energy. It also makes it possible to identify partial energy sums left unchanged by a perturbation, thus allowing the energy value to be incrementally updated. Computational tests on four proteins of sizes ranging from 68 to 755 amino acids show that MCS with the ChainTree method is significantly faster (as much as 10 times faster for the largest protein) than with the widely used grid method. Effective translation of the viral genome during the infection cycle most likely enhances its fitness. In this study, we reveal two different strategies employed by cyanophages, viruses infecting cyanobacteria, to enhance their translation efficiency. Cyanophages of the T7-like Podoviridae family adjust their GC content and codon usage to those of their hosts. In contrast, cyanophages of the T4-like Myoviridae family maintain genomes with low GC content, thus sometimes differing from that of their hosts. By introducing their own specific set of tRNAs, they appear to modulate the tRNA pools of hosts with tRNAs that fit the viral low GC preferred codons. We assessed the possible effects of those viral tRNAs on cyanophages and cyanobacterial genomes using the tRNA adaptation index, which measures the extent to which a given pool of tRNAs translates efficiently particular genes. We found a strong selective pressure to gain and maintain tRNAs that will boost translation of myoviral genes when infecting a high GC host, contrasted by a negligible effect on the host genes. Thus, myoviral tRNAs may represent an adaptive strategy to enhance fitness when infecting high GC hosts, thereby potentially broadening the spectrum of hosts while alleviating the need to adjust global parameters such as GC content for each specific host. 相似文献

Photoplethysmography is a well‐established technique for the noninvasive measurement of blood pulsation. However, photoplethysmographic devices typically need to be in contact with the surface of the tissue and provide data from a single contact point. Extensions of conventional photoplethysmography to measurements over a wide field‐of‐view exist, but require advanced signal processing due to the low signal‐to‐noise‐ratio of the photoplethysmograms. Here, we present a noncontact method based on temporal sampling of time‐integrated speckle using a camera‐phone for noninvasive, widefield measurements of physiological parameters across the human fingertip including blood pulsation and resting heart‐rate frequency. The results show that precise estimation of these parameters with high spatial resolution is enabled by measuring the local temporal variation of speckle patterns of backscattered light from subcutaneous skin, thereby opening up the possibility for accurate high resolution blood pulsation imaging on a camera‐phone.

Mounting effective innate and adaptive immune responses are critical for viral clearance and the generation of long lasting immunity. It is known that production of inhibitory factors may result in the inability of the host to clear viruses, resulting in chronic viral persistence. Fibrinogen-like protein 2 (FGL2) has been identified as a novel effector molecule of CD4+CD25+ Foxp3+ regulatory T (Treg) cells that inhibits immune activity by binding to FCγRIIB expressed primarily on antigen presenting cells (APC). In this study, we show that infection of mice with Lymphocytic Choriomeningitis Virus WE (LCMV WE) leads to increased plasma levels of FGL2, which were detected as early as 2 days post-infection (pi) and persisted until day 50 pi. Mice deficient in FGL2 (fgl2−/−) had increased viral titers of LCMV WE in the liver early p.i but cleared the virus by day 12 similar to wild type mice. Dendritic cells (DC) isolated from the spleens of LCMV WE infected fgl2−/− had increased expression of the DC maturation markers CD80 and MHC Class II compared to wild type (fgl2+/+). Frequencies of CD8+ and CD4+ T cells producing IFNγ in response to ex vivo peptide re-stimulation isolated from the spleen and lymph nodes were also increased in LCMV WE infected fgl2 −/− mice. Increased frequencies of CD8+ T cells specific for LCMV tetramers GP33 and NP396 were detected within the liver of fgl2−/− mice. Plasma from fgl2−/− mice contained higher titers of total and neutralizing anti-LCMV antibody. Enhanced anti-viral immunity in fgl2−/− mice was associated with increased levels of serum alanine transaminase (ALT), hepatic necrosis and inflammation following LCMV WE infection. These data demonstrate that targeting FGL2 leads to early increased viral replication but enhanced anti-viral adaptive T & B cell responses. Targeting FGL2 may enhance the efficacy of current anti-viral therapies for hepatotropic viruses. 相似文献

Beyond pollinator attraction: extra-floral displays deter herbivores in a Mediterranean annual plant

The evolution of several floral traits is thought to be driven by multiple selective agents, including pollinators and herbivores.
Similar combinations of selection pressures may have shaped extra-floral traits. The conspicuous purple tufts of leaves (“flags”),
which often terminate vertical inflorescences in the Mediterranean annual Salvia viridis, were shown to attract insect pollinators to the flowering patch. Here we test whether they also function as anti-herbivore
signals. We determined the aposematic potential of S. viridis flags on three levels: concentrations of anthocyanins, suggested to function as aposematic visual signals, in leaves and
flags; spectrometry to estimate whether the color-vision system of two common Mediterranean generalist herbivores (locusts
and goats) can discriminate flags from leaves; and choice experiments to determine food preferences of the same herbivores.
Anthocyanin concentrations in flags were >10-fold higher than in leaves. Flags exhibited peak reflectance at 450 and 700 nm
wavelengths, while leaves reflected maximally at 550 nm. According to the Vorobyev-Osorio color vision model, these differences
in color reflection are likely to allow visual discrimination by herbivores. Goats preferred feeding on clipped inflorescences
over control inflorescences. Locusts preferred leaves over flags. To test whether this was due to deterrence from the flags’
coloration, we also offered them choice between leaves and a rare, white morph, of the flags. The locusts chose both equally
immediately after presentation, but leaves attracted more individuals after 5 min of feeding. The locusts also preferred green
cabbage over anthocyanin-rich red cabbage. These results support the function of colorful extra-floral displays as warning

Elevated levels of extracellular glutamate have been linked to reactive oxygen species mediated neuronal damage and brain disorders. Lipoic acid is a potent antioxidant that has previously been shown to exhibit neuroprotection in clinical studies. A new positively charged water soluble lipoic acid amide analog, 2-(N,N-dimethylamine) ethylamido lipoate HCl (LA-plus), with a better cellular reduction and retention of the reduced form was developed. This novel antioxidant was tested for protection against glutamate induced cytotoxicity in a HT4 neuronal cell line. Glutamate treatment for 12 h resulted in significant release of LDH from cells to the medium suggesting cytotoxicity. Measurement of intracellular peroxides showed marked (up to 200%) increase after 6 h of glutamate treatment. Compared to lipoic acid, LA-plus was more effective in (1) protecting cells against glutamate induced cytotoxicity, (2) preventing glutamate induced loss of intracellular GSH, and (3) disallowing increase of intracellular peroxide level following the glutamate challenge. The protective effect of LA-plus was found to be independent of its stereochemistry. The protective function of this antioxidant was synergistically enhanced by selenium. These results demonstrate that LA-plus is a potent protector of neuronal cells against glutamate-induced cytotoxicity and associated oxidative stress. 相似文献
