The development of the CNS in vertebrate embryos involves the generation of different sub-types of neurons and glia in a complex but highly-ordered spatio-temporal manner. Zebrafish are commonly used for exploring the development, plasticity and regeneration of the CNS, and the recent development of reliable protocols for isolating and culturing neural stem/progenitor cells (NSCs/NPCs) from the brain of adult fish now enables the exploration of mechanisms underlying the induction/specification/differentiation of these cells. Here, we refined a protocol to generate proliferating and differentiating neurospheres from the entire brain of adult zebrafish. We demonstrated via RT-qPCR that some isoforms of ip3r, ryr and stim are upregulated/downregulated significantly in differentiating neurospheres, and via immunolabelling that 1,4,5-inositol trisphosphate receptor (IP3R) type-1 and ryanodine receptor (RyR) type-2 are differentially expressed in cells with neuron- or radial glial-like properties. Furthermore, ATP but not caffeine (IP3R and RyR agonists, respectively), induced the generation of Ca2+ transients in cells exhibiting neuron- or glial-like morphology. These results indicate the differential expression of components of the Ca2+-signaling toolkit in proliferating and differentiating cells. Thus, given the complexity of the intact vertebrate brain, neurospheres might be a useful system for exploring neurodegenerative disease diagnosis protocols and drug development using Ca2+ signaling as a read-out.  相似文献

Insecticide resistance is a major obstacle to the management of disease‐vectoring mosquitoes worldwide. The genetic changes and detoxification genes involved in insecticide resistance have been extensively studied in populations of insecticide‐resistant mosquitoes, however few studies have focused on the resistance genes upregulated upon insecticide exposure and the possible regulation pathways involved in insecticide resistance. To characterize the changes in gene expression during insecticide exposure, and to investigate the possible connection of known regulation pathways with insecticide resistance, we conducted RNA‐Seq analysis of a highly permethrin‐resistant strain of Culex quinquefasciatus following permethrin exposure. Gene expression profiles revealed a total of 224 upregulated and 146 downregulated genes when compared to a blank acetone carrier treated control, respectively, suggesting that there were multiple, but specific genes involved in permethrin resistance. Functional enrichment analysis showed that the upregulated genes contained multiple detoxification genes including a glutathione S‐transferase and multiple cytochrome P450 genes, as well as several immune‐related genes, while the downregulated genes consisted primarily of proteases and carbohydrate metabolism and transport. Further analysis showed that permethrin exposure resulted in a decrease in the expression of serum storage proteins and likely represented a delay in the development of the fourth instar possibly due to a decrease in feeding. This effect was more pronounced in an insecticide‐resistant strain than in an insecticide‐susceptible strain and may represent a behavioral mechanism of insecticide resistance in Culex mosquitoes.  相似文献

Recurring drying and wetting events are likely to increase in frequency and intensity in predicted future droughts in the central USA and alter the regeneration potential of species. We explored the resistance of seed banks to successive droughts in 53 sites across the nine locations in baldcypress swamps in the southeastern USA. Along the Mississippi River Alluvial Valley and northern Gulf of Mexico, we investigated the capacity of seed banks to retain viable seeds after successive periods of drying and wetting in a greenhouse study. Mean differences in species richness and seed density were compared to examine the interactions of successive droughts, geographical location and water regime. The results showed that both species richness and total density of germinating seedlings decreased over repeated drought trials. These responses were more pronounced in geographical areas with higher annual mean temperature. In seed banks across the southeastern swamp region, most species were exhausted after Trial 2 or 3, except for semiaquatic species in Illinois and Tennessee, and aquatic species in Texas. Distinct geographical trends in seed bank resistance to drought demonstrate that climate-induced drying of baldcypress swamps could influence the regeneration of species differently across their ranges. Despite the health of adult individuals, lack of regeneration may push ecosystems into a relict status. Seed bank depletion by germination without replenishment may be a major conservation threat in a future with recurring droughts far less severe than megadrought. Nevertheless, the protection of moist refugia might aid conservation.  相似文献

Stathmin (STMN) plays a vital role in maintaining the malignant behavior of cancer through directly regulating microtubule dynamics equilibrium. Taxol, an effective chemotherapeutics mainly acting to promote microtubule polymerization, has been commercially applied in treating solid tumors, which results in serious drug resistance. Our study demonstrated that STMN RNA interference (RNAi) enlarged taxol-induced inhibitions in cellular proliferation, colony formation, and multidimensional spaces of cell immigration and decreased half maximal inhibitory concentration (IC50) of taxol in nonsmall cell lung cancer (NSCLC) NCI-H1299 cells; STMN RNAi and taxol jointly attenuated the expressions of extracellular regulated kinase (ERK), nuclear factor kappa B (NF-κB) and B cell lymphoma-2 (Bcl-2), but up regulated Bax expression and initiated intrinsic cell death pathway by activating caspase-3 and caspase-9, while inhibited interleukin 10 (IL-10) autocrine from cell culture supernatant and xenografted mouse serum, as well as intracellular expressions of IL-10 protein and mRNA in vitro; additionally, neutralizing IL-10 alone would incur cell apoptosis to some degree; the further study confirmed that RNAi targeting STMN promoted the sensitivity of taxol in different NSCLC cells. In vivo animal experiments proved that STMN RNAi and taxol cooperatively inhibited the tumorigenicity of NCI-H1299 cells and histological atypia and Ki-67 proliferative index of xenografted tumors and promoted cell differentiation to a higher grade with well-differentiated indicators of glandular lumen-like structure and proliferative fibroblasts. These findings suggest that silencing STMN alleviates the resistance to taxol and collectively contributes to induce the dysfunction of multiple signals and down regulate the malignancy of tumors; thus, STMN is a promising target in treating refractory tumors.  相似文献

Cotton ovules were aseptically removed from ovaries 48 hr after anthesis and floated on the surface of liquid medium. Plant growth substances were filter sterilized and added to the medium in which glucose was the principal source of carbohydrate and KNO3 the sole source of nitrogen. The amount of total fiber produced from the ovule surface was determined colorimetrically from the intensity of a destaining solution to which the ovules had been transferred following their 15-second emersion in a solution of toluidine blue O. Gibberellic acid induced a marked stimulation and kinetin and abscisic acid a marked inhibition of fiber production from fertilized cotton ovules. Indoleacetic acid afforded only a trend of stimulation of fiber production. Gibberellic acid overcame the inhibition of total fiber production induced by both kinetin and abscisic acid. Indoleacetic acid did not overcome the inhibition of total fiber production induced by kinetin but did, to a great extent, overcome the inhibition due to abscisic acid.  相似文献
