Urban biodiversity in local newspapers: a historical perspective 总被引:2,自引:1,他引:2
Due to rapid urbanization the conservation and values of urban wildlife are becoming increasingly important study objects. Unfortunately, the lack of historical data makes it difficult to assess the effects of long-term urban land-use changes and human attitudes on local biodiversity. In this paper we show that old newspapers may in some cases provide useful historical data on both urban wildlife and local attitudes towards it. In the city of Turku, southwest Finland, the leading newspaper published 316 observations or reports on local urban mammals, birds and reptiles in 1890–1920, and in addition to these a lot of information on contemporary attitudes towards urban wildlife. At least 12 species of noncaptive mammals, 32 species of birds, and three reptile species were documented in newspapers. The newspaper data seem fairly reliable, and provide valuable information on the development of urban biodiversity. In the study period animals visiting urban areas were often persecuted, in which both adults and children (mainly schoolboys) participated actively. Birds were persecuted less frequently than mammals or reptiles. Some bird species, especially 'song-birds', were actively supported by winter-feeding and by constructing nest-boxes. According to newspapers local people were interested in local biodiversity, especially on phenological events, and benefited from it by getting esthetic pleasure, pet animals, and information on seasonal changes. 相似文献

The active stalactites from Grotta dei Cervi, Porto Badisco, southeastern Italy, were sampled to investigate the microbial communities present in these speleothems. Sampling was carried out in a transect about 150 m long in the central gallery, where numerous Gram-positive bacteria were isolated. Actinomycetes of the genus Streptomyces were the most abundant, followed by members of the genus Bacillus. Further isolates were assigned to the genera Amycolatopsis, Arthrobacter; Agromyces. Micrococcus, Nocardiopsis and Rhodococcus of the order Actinomycetales. The ability of actinomycetes to colonize subterranean environments is discussed. 相似文献

We combine evidence from small-scale experiments with a large-scale field survey to clarify the roles of biotic resistance
and pre-adapted habitat niche segregation to the invasion success of the North American brook trout (Salvelinus fontinalis) in North European streams previously dominated by brown trout (Salmo trutta). Interspecific aggressions among the two species were negligible, yet there was distinct habitat niche segregation between
them: brook trout occupied mainly pool habitats while brown trout tended to reside in fast-flowing riffles. Habitat niche

At present, Salmonella is considered to express two peroxiredoxin-type peroxidases, TsaA and AhpC. Here we describe an additional peroxiredoxin, Tpx, in Salmonella enterica and show that a single tpx mutant is susceptible to exogenous hydrogen peroxide (H2O2), that it has a reduced capacity to degrade H2O2 compared to the ahpCF and tsaA mutants, and that its growth is affected in activated macrophages. These results suggest that Tpx contributes significantly to the sophisticated defense system that the pathogen has evolved to survive oxidative stress.Salmonella is an important human pathogen which causes a variety of diseases, including gastroenteritis, septicemia, and typhoid fever. In the host, salmonellae reside inside phagocytic cells and are exposed to various host defense mechanisms, including oxidative stress (13). The production of superoxide anion (O2−) is crucial, as individuals with chronic granulomatous disease, which is due to a defective phagocyte NADPH oxidase, are more susceptible to infections with Salmonella (10). Likewise, diminished NADPH oxidase activity leads to increased susceptibility to Salmonella in murine macrophages (20-22, 25). Superoxide anion (O2−) is weakly reactive and fails to pass through the bacterial cell wall. After conversion to H2O2 by either spontaneous or enzymatic dismutation by superoxide dismutases, it readily diffuses into the bacterial cell and forms reactive hydroxyl radicals (OH) that damage macromolecules such as DNA, proteins, and lipids (12, 17).In principle, Salmonella possesses two classes of enzymes to degrade H2O2. Catalases degrade H2O2 to water and molecular oxygen independent of an additional reductant. Peroxiredoxin-type peroxidases (peroxiredoxins) reduce organic hydroperoxides to alcohols and hydrogen peroxide to water at the expense of NADH or NADPH. In a recent study by Hébrard et al., three members of the catalase family, KatG, KatE, and KatN, and two members of the peroxiredoxin family, AhpC and TsaA, were characterized in Salmonella (14). Previously it had been shown that single katE, katG, and katN Salmonella mutants did not show increased susceptibility to exogenous H2O2 (3, 24). In macrophages a katG katE katN triple mutant had no growth defect, whereas an ahpCF tsaA double mutant showed a reduced growth rate in macrophages (14). These observations point out the multiple routes that have evolved in Salmonella to protect the pathogen against oxidative stress and suggest that peroxiredoxins play a dominant role in the antioxidant defense during infection. In this study we characterized a third peroxiredoxin-type peroxidase, Tpx. Surprisingly, a simple tpx mutant of Salmonella enterica serovar Typhimurium (S. Typhimurium) was more susceptible to exogenous H2O2 than the wild type (WT). The mutant grew less well in activated macrophages and showed a reduced peroxidase activity toward H2O2. 相似文献

Processing of antigens by proteases in the endocytic compartments of antigen presenting cells (APC) is essential to make them suitable for presentation as antigenic peptides to T lymphocytes. Several proteases of the cysteine, aspartyl and serine classes are involved in this process. It has been speculated, that the aspartyl protease cathepsin E (CatE) is involved in antigen processing in B cell line, monocyte-derived dendritic cells (DC) and murine DC. Here we show the expression of CatE in primary human B cells and DC, which was only elevated in B cells after induction with phorbol 12-myristate 13-acetate (PMA), resulted in enhanced presentation of tetanus toxin C-fragment (TTC) to the respective T cells. Inhibition of aspartyl proteases using pepstatin-A-penetratin (PepA-P), a highly efficient, cell-permeable aspartyl protease inhibitor, reduced significantly T cell activation in PMA activated B cells but not in PMA activated myeloid DC (mDC). Thus we suggest that CatE is important in the processing of TTC in primary human B cells. 相似文献

of Kongsfjorden (Svalbard, Norway) at ≤5 m water depth and at two stations at five different water depths (5, 10, 15, 20,
30 m). Oxygen planar optode sensor spots were used ex situ to determine oxygen exchange in the overlying water of intact sediment
cores under controlled light (ca. 100 μmol photons m−2 s−1) and temperature (2–4°C) conditions. Patches of microalgae (mainly diatoms) covering sandy sediments at water depths down
to 30 m showed high biomass of up to 317 mg chl a m−2. In spite of increasing water depth, no significant trend in “photoautotrophic active biomass” (chl a, ratio living/dead cells, cell sizes) and, thus, in primary production was measured at both stations. All sites from ≤5 to
30 m water depth exhibited variable rates of net production from −19 to +40 mg O2 m−2 h−1 (−168 to +360 mg C m−2 day−1) and gross production of about 2–62 mg O2 m−2 h−1 (17–554 mg C m−2 day−1), which is comparable to other polar as well as temperate regions. Background
Abnormalities in the circadian clockwork often characterize patients with major depressive and bipolar disorders. Circadian clock genes are targets of interest in these patients. CRY2 is a circadian gene that participates in regulation of the evening oscillator. This is of interest in mood disorders where a lack of switch from evening to morning oscillators has been postulated.Principal Findings
We observed a marked diurnal variation in human CRY2 mRNA levels from peripheral blood mononuclear cells and a significant up-regulation (P = 0.020) following one-night total sleep deprivation, a known antidepressant. In depressed bipolar patients, levels of CRY2 mRNA were decreased (P = 0.029) and a complete lack of increase was observed following sleep deprivation. To investigate a possible genetic contribution, we undertook SNP genotyping of the CRY2 gene in two independent population-based samples from Sweden (118 cases and 1011 controls) and Finland (86 cases and 1096 controls). The CRY2 gene was significantly associated with winter depression in both samples (haplotype analysis in Swedish and Finnish samples: OR = 1.8, P = 0.0059 and OR = 1.8, P = 0.00044, respectively).Conclusions
We propose that a CRY2 locus is associated with vulnerability for depression, and that mechanisms of action involve dysregulation of CRY2 expression. 相似文献88.

We have established a versatile method for studying the interaction of the oleosin gene product with oil bodies during oil body biogenesis in plants. Our approach has been to transiently express a green fluorescent protein (GFP)-tagged Arabidopsis oleosin gene fusion in tobacco leaf cells containing bona fide oil bodies and then to monitor oleosin-GFP expression using real-time confocal laser scanning microscopy. We show that normally non-oil-storing tobacco leaf cells are able to synthesize and then transport oleosin-GFP fusion protein to leaf oil bodies. Synthesis and transport of oleosin-GFP fusion protein to oil bodies occurred within the first 6 h posttransformation. Oleosin-GFP fusion protein exclusively associated with the endoplasmic reticulum and was trafficked in a Golgi-independent manner at speeds approaching 0.5 microm sec(-1) along highly dynamic endoplasmic reticulum positioned over essentially static polygonal cortical endoplasmic reticulum. Our data indicate that oil body biogenesis can occur outside of the embryo and that oleosin-GFP can be used to monitor early events in oil body biogenesis in real-time. 相似文献

The aim of the study was to differentiate the impact of lighting conditions and feeding times on the regulation of lipid metabolism of goats under different photoperiods throughout the year. Seven Finnish landrace goats were kept under artificial lighting that simulated the annual changes of photoperiod at 60 degrees N (the longest light period 18 h, the shortest 6 h). Ambient temperature and feeding regime were kept constant. Blood samples were collected six times a year at 2-h intervals for 2 days, first in light/dark (LD) conditions and then after 3 days in constant darkness (DD). Significant daily variations were detected in the concentrations of plasma free fatty acids (FFA) and glycerol throughout the year. The nocturnal decrease and morning rise of FFA levels were related to the photoperiod, while the trough levels of glycerol were associated with the concentrate meal times. In DD conditions, FFA and glycerol rhythms were unstable. A significant seasonal variation was detected in the overall FFA and glycerol levels suggesting decreased lipogenesis in winter, increased lipolysis in spring and high lipogenesis in summer and fall. There was no significant daily rhythm in serum leptin levels, nor did the profiles in LD and DD conditions differ. The leptin level was slightly lower in early fall than in the other seasons, paralleling a small decrease of body mass in the goats after the grazing season. The daily or annual variations of FFA and glycerol levels were not clearly related to leptin concentrations. The results suggest that lipid metabolism of goats is regulated by light even in constant temperature and feeding conditions; however, no significant contribution of leptin levels could be shown. 相似文献

A 25-year time series for a vendace (Coregonus albula (L.)) population was analysed with mathematical models to reveal the factors that regulate its recruitment. The fitted recruitment model was an age-structured model incorporating the compensatory effects of the spawning population and the abundance of the previous year class. Wind forcing index was also added as an example of an external source of recruitment variability. The auto-correlation analysis revealed a tendency for 2-year generation cyclicity in recruitment. The compensatory effects of both spawning biomass and previous year class abundance on recruitment had to be incorporated into the model to remove cyclicity from residuals. Wind forcing during the larval and early juvenile periods negatively affected recruitment. Re-estimating the parameters of the Cushing recruitment function and the effect of previous year-class from simulated data revealed that the time series structure and measurement errors induce strong biases in parameters exaggerating the density independent population growth rate parameter and the amount of compensation. The negative effect of previous year class was also exaggerated but less severely. Simulations with the artificially perturbed deterministic model skeleton revealed a tendency for cyclicity in recruitment. The model typically generated dampening oscillations, but the dynamics appeared as limit cycles when assuming high mortality, a low level of compensation by spawning biomass and a considerable negative effect of the previous year-class. No single ultimate mechanism for inter-stage effects causing cyclicity can be presently designated despite the rather extensive studies on vendace population dynamics.
