This study examines the effects of a radical bariatric surgery-induced weight loss on the gait of obese subjects. We performed a three-dimensional motion analysis of lower limbs, and collected force platform data in the gait laboratory to calculate knee and hip joint moments. Subjects (n=13) performed walking trials in the laboratory before and 8.8 months (SD 4.2) after the surgical procedure at two gait speeds (1.2m/s and 1.5m/s). The average weight loss was 26.7kg (SD 9.2kg), corresponding to 21.5% (SD 6.8%) of the initial weight. We observed a decrease in step width at both gait speeds, but no changes in relative double support or swing time or stride length. A significant decrease was noted in the absolute values of peak knee abductor, peak knee flexor and peak hip extensor moments. However, the moment values normalized by the body weight and height remained unchanged in most cases. Thus, we conclude that weight loss reduces hip and knee joint moments in proportion to the amount of weight lost. 相似文献

Summary Rabbits were immunized using human erythroxyte carbonic anhydrase B (HCA B) purified. by the modified methods of Armstrong et al. (1966) and Bernstein and Schraer (1972). The globulin fraction was isolated by ammonium sulphate precipitation. The anti-HCA B globulin was specific, when judged using the double diffusion technique of Ouchterlony and immunoelectrophoresis. No cross reaction with human erythrocyte carbonic anhydrase C was found, but cross reactions with erythrocyte carbonic anhydrase from rat, mouse and guinea pig were observed. Gamma-aminobutyric acid (GABA) is synthesized by two isoforms of the pyridoxal 5'-phosphate-dependent enzyme glutamic acid decarboxylase (GAD65 and GAD67). GAD67 is constitutively active and is responsible for basal GABA production. In contrast, GAD65, an autoantigen in type I diabetes, is transiently activated in response to the demand for extra GABA in neurotransmission, and cycles between an active holo form and an inactive apo form. We have determined the crystal structures of N-terminal truncations of both GAD isoforms. The structure of GAD67 shows a tethered loop covering the active site, providing a catalytic environment that sustains GABA production. In contrast, the same catalytic loop is inherently mobile in GAD65. Kinetic studies suggest that mobility in the catalytic loop promotes a side reaction that results in cofactor release and GAD65 autoinactivation. We investigate olfactory associative learning in larval Drosophila. A reciprocal training design is used, such that one group of animals receives a reward in the presence of odor X but not in the presence of odor Y (Train: X+ // Y), whereas another group is trained reciprocally (Train: X // Y+). After training, differences in odor preference between these reciprocally trained groups in a choice test (Test: X - Y) reflect associative learning. The current study, after showing which odor pairs can be used for such learning experiments, 1) introduces a one-odor version of such reciprocal paradigm that allows estimating the learnability of single odors. Regarding this reciprocal one-odor paradigm, we show that 2) paired presentations of an odor with a reward increase odor preference above baseline, whereas unpaired presentations of odor and reward decrease odor preference below baseline; this suggests that odors can become predictive either of reward or of reward absence. Furthermore, we show that 3) innate attractiveness and associative learnability can be dissociated. These data deepen our understanding of odor-reward learning in larval Drosophila on the behavioral level, and thus foster its neurogenetic analysis. 相似文献

A spectrophotometer-fluorometer was constructed for simultaneous measurement of reflectance spectrum changes and surface fluorescence of intact tissues and organs. The apparatus employs monochromators for dual-wavelength spectrophotometry and optical filters for fluorometry. Three light beams are time-shared by a novel type of modulator to hit the same area of the tissue to be measured. The analog circuitry was controlled by a digital logic circuitry clocked by an optically derived frequency based on the angular velocity of the rotating light modulator and produced a maximum of three sets of differences in the parameters measured. The optical design has the advantage of allowing the use of monochromators rather than filters in a multichannel apparatus. The electronics accept a wide range of light modulation frequencies without difficulties in the phasing of the detector gates, so that signal sampling can be optimized for the recording of fast and slow phenomena. The apparatus was used successfully for the simultaneous monitoring of the mitochondrial redox state and tissue oxygenation in isolated perfused rat hearts. 相似文献

Summary The intracellular ATP of baker's yeast (Saccharomyces cerevisiae) was measured using the bioluminescent firefly luciferase assay. Benzalkonium chloride and trichloro-acetic acid served in the experiments as extracting agents and optimal conditions for the extraction and assay of the intracellular ATP are reported. Using the results obtained from manually performed experiments two continuous flow systems were designed for the measurement of ATP in yeast cells during cell growth. Good correlation between the amount of cellular ATP and cell growth was found during the exponential growth phase. 相似文献

Relationships between the elemental composition, the microfibril angle (MFA) distribution and the average shape of the cell
cross-section of irrigated-fertilised and untreated Norway spruce (Picea abies [L.] Karst.) earlywood were studied. Sample material was obtained from Flakaliden, Sweden. The elemental composition was
studied by determining the relative mass fractions of the elements P, S, Cl, K, Ca and Mn by X-ray fluorescence and by determining
the mass absorption coefficients for X-rays. X-ray diffraction was used to determine the MFA distribution and the average
shape of the cell cross-section. The latter was also determined by light microscopy. In transition from juvenile wood to mature
wood, a decrease of the mode of the MFA distribution from 13°–24° to 3°–6° was connected to a change in the shape of the cell
cross-section from circular to rectangular. The irrigation-fertilisation treatment caused no change in the MFA distribution
or in the shape of the cell cross-section, whereas the mass absorption coefficient was higher and the density was smaller
in irrigated-fertilised wood. Larger proportion of the elements S, Cl and K, but smaller proportion of the element Mn, were
observed due to the treatment. The results indicate that the shape of the cell cross-section or the MFA distribution are not
directly linked to the growth rate of tracheids or to the nutrient-element content in the xylem and only show notable changes
