Recently, microalgae have been considered as a promising alternative for the production of biofuels from CO2. For the efficient cultivation of these microalgae, several types of photobioreactors have been designed and Pilot scale photobioreactors have been used to assess the performance of these reactors. Therein the primarily investigated reactor type is the Raceway Pond. However, the less researched Thin‐Layer Cascade Photobioreactor (TLC) shows a high potential for efficient production processes. Unfortunately, for low‐value products like biofuels costs must be kept to a minimum for an economic operation. To facilitate this, 3D Computational Fluid Dynamic simulations can be employed to estimate performance of reactor variants e.g. with respect to power input and mixing. Since up to now little effort has been put into the modelling of TLC reactors, this report aims to present a simulation approach for these reactors types that allows simple adaptation to different geometric or operational boundary conditions. All models have been generated for a two‐phase mixture in OpenFOAM. To demonstrate its applicability, validation measurements with a physical unit have been performed and were compared to the simulation results. With errors in the order of 10 % a successful simulation of the reactor geometry could be proven.  相似文献

Local adaptation may cause thermal tolerance to vary between nearby but distinct populations of a species. During the summer of 2013, alligator gar Atractosteus spatula spawned from broodstock collected from three populations within the Mississippi River drainage separated by a 5° latitudinal gradient were acclimated to three temperatures (25, 30, and 35°C). Ten fish from each population were acclimated at each temperature. CTMax was determined at each temperature for each population, using five fish for each population‐acclimation temperature pairing. CTMax for each population‐acclimation temperature pairing was compared using two‐factor anova . CTMax increased significantly with acclimation temperature (F2,40 = 600.5, P < 0.001) but population had no significant effect (F2,40 = 1.882, P = 0.166). Temperature tolerance appears to be consistent across populations of alligator gar, with no evidence of local adaptation.  相似文献

Tall shrubs and trees are advancing into many tundra and wetland ecosystems but at a rate that often falls short of that predicted due to climate change. For forest, tall shrub, and tundra ecosystems in two pristine mountain ranges of Alaska, we apply a Bayesian, error‐propagated calculation of expected elevational rise (climate velocity), observed rise (biotic velocity), and their difference (biotic inertia). We show a sensitive dependence of climate velocity on lapse rate and derive biotic velocity as a rigid elevational shift. Ecosystem presence identified from recent and historic orthophotos ~50 years apart was regressed on elevation. Biotic velocity was estimated as the difference between critical point elevations of recent and historic logistic fits divided by time between imagery. For both mountain ranges, the 95% highest posterior density of climate velocity enclosed the posterior distributions of all biotic velocities. In the Kenai Mountains, mean tall shrub and climate velocities were both 2.8 m y?1. In the better sampled Chugach Mountains, mean tundra retreat was 1.2 m y?1 and climate velocity 1.3 m y?1. In each mountain range, the posterior mode of tall woody vegetation velocity (the complement of tundra) matched climate velocity better than either forest or tall shrub alone, suggesting competitive compensation can be important. Forest velocity was consistently low at 0.1–1.1 m y?1, indicating treeline is advancing slowly. We hypothesize that the high biotic inertia of forest ecosystems in south‐central Alaska may be due to competition with tall shrubs and/or more complex climate controls on the elevational limits of trees than tall shrubs. Among tall shrubs, those that disperse farthest had lowest inertia. Finally, the rapid upward advance of woody vegetation may be contributing to regional declines in Dall's sheep (Ovis dalli), a poorly dispersing alpine specialist herbivore with substantial biotic inertia due to dispersal reluctance.  相似文献

Chromobacterium violaceum DSM 30191 accumulated a homopolyester of 3-hydroxyvaleric acid (3HV) up to 65% of the cellular dry matter during cultivation in fed-batch cultures with valeric acid as sole carbon source and during cell starvation of the nitrogen source. From fructose, gluconate, propionate or hexanoate a homopolyester of 3-hydroxybutyrate (3HB) was accumulated. Poly(3HV) homopolyster was also accumulated by two different strains of C. violaceum, whereas two other strains of C. violaceum and three strains of Janthinobacterium lividum accumulated poly(3HB-co-3HV) copolyesters from valerate. The composition of the biosynthetic poly(3HV) was confirmed by various nuclear magnetic resonance spectroscopic methods. Differential scanning calorimetry analysis of four poly(3HV) samples that were isolated from different batches of cells revealed glass transition temperatures between –10 and –12°C and melting points between 107 and 112°C. Viscosity measurements gave intrinsic viscosities between 62.5 and 124.8 × 10–2 dl/g for these samples, indicating approximate relative molecular masses between 60 000 and 145 000 of the biosynthetic poly(3HV). Cloud computing is an emerging technology and is being widely considered for resource utilization in various research areas. One of the main advantages of cloud computing is its flexibility in computing resource allocations. Many computing cycles can be ready in very short time and can be smoothly reallocated between tasks. Because of this, there are many private companies entering the new business of reselling their idle computing cycles. Research institutes have also started building their own cloud systems for their various research purposes. In this paper, we introduce a framework for virtual cluster system called vcluster which is capable of utilizing computing resources from heterogeneous clouds and provides a uniform view in computing resource management. vcluster is an IaaS (Infrastructure as a Service) based cloud resource management system. It distributes batch jobs to multiple clouds depending on the status of queue and system pool. The main design philosophy behind vcluster is cloud and batch system agnostic and it is achieved through plugins. This feature mitigates the complexity of integrating heterogeneous clouds. In the pilot system development, we use FermiCloud and Amazon EC2, which are a private and a public cloud system, respectively. In this paper, we also discuss the features and functionalities that must be considered in virtual cluster systems.  相似文献

Leishmania spp. is a protozoan parasite and the causative agent of leishmaniasis. Thymidine kinase (TK) catalyses the transfer of the γ-phosphate of ATP to 2’-deoxythymidine (dThd) forming thymidine monophosphate (dTMP). L. major Type II TK (LmTK) has been previously shown to be important for infectivity of the parasite and therefore has potential as a drug target for anti-leishmanial therapy. In this study, we determined the enzymatic properties and the 3D structures of holo forms of the enzyme. LmTK efficiently phosphorylates dThd and dUrd and has high structural homology to TKs from other species. However, it significantly differs in its kinetic properties from Trypanosoma brucei TK since purines are not substrates of the enzyme and dNTPs such as dUTP inhibit LmTK. The enzyme had Km and kcat values for dThd of 1.1 μM and 2.62 s-1 and exhibits cooperative binding for ATP. Additionally, we show that the anti-retroviral prodrug zidovudine (3-azido-3-deoxythymidine, AZT) and 5’-modified dUrd can be readily phosphorylated by LmTK. The production of recombinant enzyme at a level suitable for structural studies was achieved by the construction of C-terminal truncated versions of the enzyme and the use of a baculoviral expression system. The structures of the catalytic core of LmTK in complex with dThd, the negative feedback regulator dTTP and the bi-substrate analogue AP5dT, were determined to 2.74, 3.00 and 2.40 Å, respectively, and provide the structural basis for exclusion of purines and dNTP inhibition. The results will aid the process of rational drug design with LmTK as a potential target for anti-leishmanial drugs.  相似文献

A promoter sequence, PAN, was isolated from Mycobacterium paratuberculosis and characterized. This promoter lies adjacent to, and outside, the 3' end of an IS900 insertion element. IS900 contains an open reading frame, ORF2, on the complementary strand which codes for the putative transposase of this insertion sequence. A DNA fragment containing PAN and part of ORF2 was fused to the lacZ gene and inserted into the replicative shuttle vector pRR3. Mycobacterium smegmatis and Mycobacterium bovis BCG (BCG) transformed with this plasmid exhibited beta-galactosidase activity. However, lacZ was only expressed in Escherichia coli under the control of PAN, when ORF2 was deleted. Immunization of mice with the recombinant M. bovis BCG expressing lacZ resulted in the induction of a high humoral and cellular response directed against beta-galactosidase. The PAN-ORF2 expression system may prove to be particularly useful for cloning and expression of heterologous genes in the BCG vaccine strain.  相似文献
