Schwannomas may develop sporadically or in association with NF2 and schwannomatosis. The fundamental aberration in schwannomas is the bi-allelic inactivation of the NF2 gene. However, clinical and molecular data suggest that these tumors share a common pathogenetic mechanism related to as yet undefined 22q-loci. Linkage studies in schwannomatosis, a condition related to NF2, have defined a candidate 22q-locus and excluded the NF2 gene as the causative germline mutation. Thus, analysis of aberrations in schwannomas may lead to the identification of putative gene(s) involved in the development of schwannoma/schwannomatosis. We profiled a series of 88 schwannomas and constitutional DNA using a tiling path chromosome 22 array. Array-CGH is a suitable method for high-resolution discrimination between germline and tumor-specific aberrations. Previously reported frequencies of 22q-associated deletions in schwannomas display large discrepancies, ranging from 30% to 80%. We detected heterozygous deletions in 53% of schwannomas and the predominant pattern was monosomy 22. In addition, three tumors displayed terminal deletions and four harbored overlapping interstitial deletions of various sizes encompassing the NF2 gene. When profiling constitutional DNA, we identified eight loci that were affected by copy number variation (CNV). Some of the identified CNVs may not be phenotypically neutral and the possible role of these CNVs in the pathogenesis of schwannomas should be studied further. We observed a correlation between the breakpoint position, present in tumor and/or constitutional DNA and the location of segmental duplications. The microwave-assisted synthesis and characterization of the ten new sildenafil (Viagra; 1) analogues 6-15 are described. A detailed structure-activity-relationship (SAR) study revealed that compounds 10 (= 4-ethoxy-N-hydroxy-3-(7-methoxy-1-methyl-3-propyl-1H-pyrazolo[4,3-d]pyrimidin-5-yl)benzenesulfonamide) and 12 (= S-(2-hydroxyethyl) 4-ethoxy-3-(7-methoxy-1-methyl-3-propyl-1H-pyrazolo[4,3-d]pyrimidin-5-yl)benzenesulfonothioate) are extremely potent mushroom tyrosinase inhibitors, with IC50 values (3.59 and 2.15 microM, resp.) below those of the standard inhibitors L-mimosine and kojic acid (IC50 = 3.68 and 16.67 microM, resp.). Compounds 10 and 12 are, thus, the currently most-effective inhibitors of tyrosinase, and bear great potential to be used for the treatment of various skin disorders such as hyperpigmentation, which is associated with high production of melanocytes. 相似文献

The aim of this work was to investigate the effects of supercritical carbon dioxide (SC-CO2) processing on the release profiles of progesterone (PGN) and Gelucire 44/14 dispersion systems. A fractional factorial design was conducted for optimization of the particles from gas-saturated suspension (PGSS) method and formulation parameters and evaluating the effects of three independent responses: PGSS process yield, in vitro dissolution extent after 20 min (E20) and t1/2 for prepared PGN dispersion systems. The experimental domain included seven factors measured at two levels to determine which factors represent the greatest amount of variation, hence the most influence on the resulting PGN dispersion systems. Variables tested were temperature (A) and pressure (B) of the supercritical fluid, sample loading (C), SC-CO2 processing time (D), sonication (E), drug-to-excipient ratio (F) and orifice diameter into the expansion chamber (G). The analysis of variance showed that the factors tested had significant effects on the responses (p value <0.05). It was found that the optimum values of the PGSS process are higher pressure (186 bar), higher temperature (60°C), a longer processing time (30 min) and lower PGN-to-excipient ratio of 1:10. The corresponding processing yield was 94.7%, extent of PGN dissolution after 20 min was 85.6% and the t1/2 was 17.7 min. The results suggest that Gelucire 44/14-based dispersion systems might represent a promising formulation for delivery of PGN. Purpose
The conventional decision-making for bridges is mostly focusing on technical, economical, and safety perspectives. Nowadays, the society devotes an ever-increased effort to the construction sector regarding their environmental performance. However, considering the complexity of the environmental problems and the diverse character of bridges, the related research for bridge as a whole system is very rare. Most existing studies were only conducted for a single indicator, part of the structure components, or a specific life stage.Methods
Life Cycle Assessment (LCA) is an internationally standardized method for quantifying the environmental impact of a product, asset, or service throughout its whole life cycle. However, in the construction sector, LCA is usually applied in the procurement of buildings, but not bridges as yet. This paper presents a comprehensive LCA framework for road bridges, complied with LCA ReCiPe (H) methodology. The framework enables identification of the key structural components and life cycle stages of bridges, followed by aggregation of the environmental impacts into monetary values. The utility of the framework is illustrated by a practical case study comparing five designs for the Karlsnäs Bridge in Sweden, which is currently under construction.Results and discussion
This paper comprehensively analyzed 20 types of environmental indicators among five proposed bridge designs, which remedies the absence of full spectrum of environmental indicators in the current state of the art. The results show that the monetary weighting system and uncertainties in key variables such as the steel recycling rate and cement content may highly affect the LCA outcome. The materials, structural elements, and overall designs also have varying influences in different impact categories. The result can be largely affected by the system boundaries, surrounding environment, input uncertainties, considered impact indicators, and the weighting systems applied; thus, no general conclusions can be drawn without specifying such issues.Conclusions
Robustly evaluating and ranking the environmental impact of various bridge designs is far from straightforward. This paper is an important attempt to evaluate various designs from full dimensions. The results show that the indicators and weighting systems must be clearly specified to be applicable in a transparent procurement. Background
The occurrence of variations in the spectrum of cardiovascular disease between different regions of the world and ethnic groups have been the subject of great interest. This study report the 24-h variation of myocardial infarction (MI) occurrence in patients recruited from CCU located in Argentina and Uruguay.Methods
A cohort of 1063 patients admitted to the CCU within 24 h of the onset of symptoms of an acute MI was examined. MI incidence along the day was computed in 1 h-intervals.Results
A minimal MI incidence between 03:00 and 07:00 h and the occurrence of a first maximum between 08:00 and 12:00 h and a second maximum between 15:00 and 22:00 h were verified. The best fit curve was a 24 h cosinor (acrophase ~ 19:00 h, accounting for 63 % of variance) together with a symmetrical gaussian bell (maximum at ~ 10:00 h, accounting for 37 % of variance). A similar picture was observed for MI frequencies among different excluding subgroups (older or younger than 70 years; with or without previous symptoms; diabetics or non diabetics; Q wave- or non-Q wave-type MI; anterior or inferior MI location). Proportion between cosinor and gaussian probabilities was maintained among most subgroups except for older patients who had more MI at the afternoon and patients with previous symptoms who were equally distributed among the morning and afternoon maxima.Conclusion

Different levels of sexual size dimorphism (SSD) have usually been explained by selective forces operating in the adult stage. Developmental mechanisms leading to SSD during the juvenile development have received less attention. In particular, it is often not clear if the individuals of the ultimately larger sex are larger already at hatching/birth, do they grow faster, or do they grow for a longer time. In the case of insects, the question about sexually dimorphic growth rates is still open because most previous studies fail to adequately consider the complexity of larval growth curve, the existence of distinct larval instars in particular. Applying an instar-specific approach, we analysed ontogenetic determination of female-biased SSD in a number of distantly related species of Lepidoptera. The species studied showed a remarkable degree of similarity: SSD appeared invariably earlier than in the final instar, and tended to accumulate during development. The higher weight of the females was shown to be primarily a consequence of longer development within several larval instars. There was some evidence of higher instantaneous growth rates of females in the penultimate instar but not in the final instar. Egg size, studied in one species, was found not to be sexually dimorphic. The high across-species similarity may be seen as an indication of constraints on the set of possible mechanisms of size divergence between the two sexes. The results are discussed from the perspective of the evolution of insect body size in general. In particular, this study confirms the idea about limited evolvability of within-instar growth increments. An evolutionary change towards larger adult size appears always to be realised via moderate changes in relative increments of several larval instars, whereas a considerable change in just one instar may not be feasible. 相似文献

We detected a significant inter- and intraspecific host preference on the level of individual host use in a system, in which three moth species (Lepidoptera: Noctuidae), feeding on a cattail Typha latifolia , are parasitized by three solitary parasitoid species (Hymenoptera: Ichneumonidae). The biology of the host species is similar but they exhibit remarkable inter- and intraspecific variance in body size. All the parasitoid species preferred the largest host species in this system whereas other host species were used only occasionally. We found that parasitoids which emerged from females of the preferred host species were larger than those which developed in males of the same species. Accordingly, two of the parasitoid species had a significant within-host-species preference: females of the largest moth species were used more often than males. No dependence of the preference pattern on host density was found. This pattern of host use is discussed in the light of the switching theory and the optimal host selection theory. Our results indicate that non-random host use by parasitoids may have significant effects on host populations and communities, and forms a potential selective factor against large body size in herbivorous insects. Fitness consequences of early-life environmental conditions are often sex-specific, but corresponding evidence for invertebrates remains inconclusive. Here, we use meta-analysis to evaluate sex-specific sensitivity to larval nutritional conditions in insects. Using literature-derived data for 85 species with broad phylogenetic and ecological coverage, we show that females are generally more sensitive to food stress than males. Stressful nutritional conditions during larval development typically lead to female-biased mortality and thus increasingly male-biased sex ratios of emerging adults. We further demonstrate that the general trend of higher sensitivity to food stress in females can primarily be attributed to their typically larger body size in insects and hence higher energy needs during development. By contrast, there is no consistent evidence of sex-biased sensitivity in sexually size-monomorphic species. Drawing conclusions regarding sex-biased sensitivity in species with male-biased size dimorphism remains to wait for the accumulation of relevant data. Our results suggest that environmental conditions leading to elevated juvenile mortality may potentially affect the performance of insect populations further by reducing the proportion of females among individuals reaching reproductive age. Accounting for sex-biased mortality is therefore essential to understanding the dynamics and demography of insect populations, not least importantly in the context of ongoing insect declines. 相似文献

The genomes are regularly targeted by epigenetic regulatory mechanisms (DNA methylation, histone modifications, binding of regulatory proteins) in infected cells. In addition, proteins encoded by microbial genomes may disturb the action of a set of cellular promoters by interacting with the same epi-regulatory machinery. The outcome of this may result in epigenetic dysregulation and subsequent cellular dysfunctions that may manifest in or contribute to the development of pathological changes. How epigenetic methylation decorations on DNA and histones are started and established remains largely unknown. The inherited nature of these processes in regulation of genes suggests that they could play key roles in chronic diseases associated with microbial persistence; they might also explain so-called hit-and-run phenomena in infectious disease pathogenesis. Microbes infecting mammals may cause diseases by causing hyper-methylation of key cellular promoters at CpG di-nucleotides and may induce pathological changes by epigenetic reprogramming of host cells they are interacting with elucidation of the epigenetic consequences of microbe–host interactions may have important therapeutic implications because epigenetic processes can be reverted and elimination of microbes inducing patho-epigenetic changes may prevent disease development. 相似文献
