1. A soluble phosphodiesterase is present in mammalian tissues which rapidly hydrolyses enantiomorphs of rac-glycerol 1:2-cyclic phosphate, producing rac-glycerol 1-phosphate. 2. The enzyme has been purified up to 1700-fold by a combination of acetone precipitation and chromatography on DEAE-Sephadex A-50, Sephadex G-150 and hydroxyapatite. 3. The Km with glycerol cyclic phosphate as substrate is 7.2 mM, and the pH optimum broad (6.9--7.5). The molecular weight (by gel filtration) of the enzyme is approx. 35500. 4. The phosphodiesterase has no requirement for Ca2+ or Mg2+, but is stimulated by reducing agents (cysteine, dithiothreitol) and Fe2+. 5. The purified phosphodiesterase preparation also hydrolysed 3':5'-cyclic AMP, producing 5'-AMP exclusively, and 2':3'-cyclic AMP, forming 3'-AMP and 2'-AMP in the ratio 7:3. Bis-(p-nitrophenyl) phosphate was slowly hydrolysed, but other phosphodiesters tested were not attacked. 6. The phosphodiesterase is inhibited by theophylline and o-phenanthroline. It is inhibited by Pi and by a variety of phosphomonoesters, of which certain aromatic primary phosphates are particularly effective.  相似文献

Purkinje cell degeneration (pcd) is a neurological mutation in the mouse that causes male sterility, but not female sterility. In order to assess the effects of this mutation on spermiogenesis, the structure of the testis and of epididymal spermatozoa was examined by transmission and scanning electron microscopy. In the mutant males, the sperm count was reduced, sperm were nonmotile, and 93% of the sperm were characterized by structural abnormalities of the head, the tail, or both. In the testes of mutant mice, Sertoli cell structure was normal, as were also the early stages of spermiogenesis. However, the elongating and maturing spermatids were characterized by abnormally shaped heads and tails with extraneous and ectopic outer dense fibers. These defects were common in the testes of the mutant mice and rare in the testes of the littermate control mice. It was concluded that the structural abnormalities of the pcd sperm occurred during spermiogenesis and were not due to degeneration of the sperm in the epididymis. These structural abnormalities are similar to those found in all other reported male sterile mutants of the mouse; therefore, although they are caused by the expression of the pcd gene, they are not unique to the expression of this gene.  相似文献

Radioiodination of immunoglobulins, particularly human cytotoxic IgG, induced a marked loss of antibody activity. Phenylisothiocyanate (PITC) readily reacts with alpha-amino groups and the epsilon-amino groups of lysines to form phenylthiocarbamyl derivatives. Since PITC is commercially available with 14C, 3H, and 35S substitutions, it provided an alternative means for labeling immunoglobulin which preserved antibody activity. There were approximately 80 PITC binding sites per IgG molecule, and 14C-PITC was bound with an efficiency in excess of 80%. With as many as 40 PITC molecules bound per IgG molecule, full cytotoxic activity was retained by both anti-HLA isoantisera and human anti-melanoma autoantisera. Even with 70 to 80 molecules of PITC bound per IgG molecule, over 80% of the antibody activity remained. Radiolabeling of antibody with 3H, 14C, or 35S-substituted PITC will greatly facilitate studies of antibody binding, particularly to cell surface antigens.  相似文献

Despite the fact that many parasitic and hemiparasitic plant species such as mistletoes are dioecious and occur in both the new and the old world, few data exist on variation in the sex ratio and allocation to reproduction in these taxa. We investigated 1) the sex-ratio of the xylem-tapping mistletoe Phoradendron juniperinum in relation to its age and position within the canopy of its host tree Juniperus osteosperma, and 2) reproductive effort in relation to the gender and age of mistletoe plants. Our surveys showed that P. juniperinum has a male-biased sex ratio. Despite this predominance of male individuals, females lived longer and had a greater reproductive effort than did males. A statistical analysis of the age distribution data indicated that the peak in the frequency of reproductively mature individuals was later in females than in males. These gender-specific distributions may have resulted 1) from sequential hermaphroditism (age-specific sex switching), or 2) because the average age of peak reproduction is later in female individuals. Because sex is genetically determined in a closely related genus of mistletoe and because we have no data to indicate sex switching in this species, we feel that our data support the interpretation that female individuals, on average, show a peak in reproductive vigor at an older age relative to males. While delayed reproduction in females may be favored because reproductive effort and success appear to be age-dependent in females of this species, both sexes can become reproductively mature relatively early in life. Further, because 1) allocation to reproduction as a function of age increases more rapidly for females of this species relative to males, and 2) because there may be a higher resource cost associated with reproduction in females, we hypothesized that female individuals would be more abundant in the best quality locations within the host tree so as to maximize the opportunity to meet those costs. In spite of the association between gender and some host characteristics, there was no indication that female plants were located in sites most favorable to either their carbon or water balance. We discuss reasons why this may be the case.  相似文献

Prostaglandin E2 synthesis and eicosanoid biosynthetic enzyme activities (arachidonyl CoA synthetase, cyclooxygenase and phospholipase A2) were measured in dermal fibroblasts from patients with metabolic disorders of peroxisomal origin and compared to those from normal subjects and patients with other metabolic disorders of lipid metabolism. Basal- as well as interleukin 1-stimulated prostaglandin E2 syntheses were higher in fibroblasts from patients with X-linked adrenoleukodystrophy, the Zellweger cerebrohepatorenal syndrome and rhizomelic chondrodysplasia punctata than in normals. Basal cyclooxygenase and phospholipase A2 activities were elevated in most of the peroxisomal disease cells. Cells from patients with adrenomyeloneuropathy, however, had significantly lower cytokine-stimulated cyclooxygenase and phospholipase A2 activities than normals, as well as lower prostaglandin E2 synthesis in response to interleukin 1. The peroxisomal disease lines exhibited dose-response curves to interleukin 1 similar to controls. Receptor-binding analysis indicated that cells from patients with rhizomelic chondrodysplasia punctata expressed 5-times fewer interleukin 1 receptors than normals and the other disease lines. Exaggerated arachidonic acid metabolism in response to interleukin 1 suggests that cells from patients with peroxisomal enzyme defects may be useful in elucidating pathways for arachidonate release and eicosanoid synthesis.  相似文献

The effect of a support composed of polymers based on poly-N-isopropyl acrylamide and poly-t-butyl acrylamide and collagen on human fibroblasts was studied. As the temperature was decreased to 4 degrees C, the polymeric support is converted to a diluted state and cells spontaneously detached from it. The presence of collagen in the support prevented the detachment of cells and increased cell growth. It was shown by microcalorimetry, that in a copolymer-collagen mixture, a microstratification takes place.  相似文献
