大豆(Glycine max)是重要的油料和蛋白作物, 其丰富的遗传变异为生物学性状挖掘和育种改良提供了重要的资源基础。然而, 单个基因组信息无法全面揭示种质资源的遗传变异, 泛基因组研究为解决这一不足提供了新方案。近日, 中国科学院遗传与发育生物学研究所田志喜和梁承志研究团队从2 898份大豆种质中选取26份代表性材料, 并整合已有的3个基因组, 构建了包含野生和栽培大豆的泛基因组和图基因组(graph-based genome), 鉴定了整个群体的绝大多数结构变异数据集, 确定了大豆种质的核心、非必需和个体特异的基因集。利用这些数据系统地揭示了生育期位点E3的等位基因变异和基因融合事件、种皮颜色基因I的单体型和演化关系以及结构变异对铁离子转运基因表达和地区适应性选择的影响。该研究为作物基因组学研究提供了一个新的模式, 同时将加速推动大豆遗传变异的鉴定、性状解析和种质创新。  相似文献

Switchgrass is considered one of the most promising energy crops. However, breeding of elite switchgrass cultivars is required to meet the challenges of large scale and sustainable biomass production. As a native perennial adapted to North America, switchgrass has lowland and upland ecotypes, where most lowland ecotypes are tetraploid (2n?=?4x?=?36), and most upland ecotypes are predominantly octoploid (2n?=?8x?=?72). Hybridization between lowland and upland switchgrass plants could identify new cultivars with heterosis. However, crossing between tetraploid and octoploid switchgrass is rare in nature. Therefore, in order to break down the cross incompatibility barrier between tetraploid lowland and octoploid upland switchgrass lines, we developed autoployploid switchgrass lines from an anueploid lowland cv. Alamo. In this study, colchicine was used in liquid and solid mediums to chemically induce chromosome doubling in embryogenic calli derived from cv. Alamo. Thirteen autopolyploid switchgrass lines were regenerated from seedlings and identified using flow cytometry. The autoplyploid switchgrass plants exhibited increased stomata aperture and stem size in comparison with the cv. Alamo. The most autooplyploid plants were regenerated from switchgrass calli that were treated with 0.04 % colchicine in liquid medium for 13 days. One autopolyploid switchgrass line, VT8-1, was successfully crossed to the octoploid upland cv. Blackwell. The autoployploid and the derived inter-ecotype hybrids were confirmed by in situ hybridization and molecular marker analysis. Therefore, the results of this study show that an autopolyploid, generated by chemically induced chromosome doubling of lowland cv. Alamo, is cross compatible with upland octoploid switchgrass cultivars. The outcome of this study may have significant applications in switchgrass hybrid breeding.  相似文献

Recent results demonstrate techniques for fully quantitative, statistical inference of the dynamics of individual neurons under the Hodgkin–Huxley framework of voltage-gated conductances. Using a variational approximation, this approach has been successfully applied to simulated data from model neurons. Here, we use this method to analyze a population of real neurons recorded in a slice preparation of the zebra finch forebrain nucleus HVC. Our results demonstrate that using only 1,500 ms of voltage recorded while injecting a complex current waveform, we can estimate the values of 12 state variables and 72 parameters in a dynamical model, such that the model accurately predicts the responses of the neuron to novel injected currents. A less complex model produced consistently worse predictions, indicating that the additional currents contribute significantly to the dynamics of these neurons. Preliminary results indicate some differences in the channel complement of the models for different classes of HVC neurons, which accords with expectations from the biology. Whereas the model for each cell is incomplete (representing only the somatic compartment, and likely to be missing classes of channels that the real neurons possess), our approach opens the possibility to investigate in modeling the plausibility of additional classes of channels the cell might possess, thus improving the models over time. These results provide an important foundational basis for building biologically realistic network models, such as the one in HVC that contributes to the process of song production and developmental vocal learning in songbirds.  相似文献

Full-length nucleoproteins from Ebola and Marburg viruses were expressed as His-tagged recombinant proteins in Escherichia coli and nucleoprotein-based enzyme-linked immunosorbent assays(ELISAs) were established for the detection of antibodies specific to Ebola and Marburg viruses. The ELISAs were evaluated by testing antisera collected from rabbit immunized with Ebola and Marburg virus nucleoproteins. Although little cross-reactivity of antibodies was observed in antiEbola virus nucleoprotein rabbit antisera, the highest reactions to immunoglobulin G(Ig G) were uniformly detected against the nucleoprotein antigens of homologous viruses. We further evaluated the ELISA's ability to detect antibodies to Ebola and Marburg viruses using human sera samples collected from individuals passing through the Guangdong port of entry. With a threshold set at the mean plus three standard deviations of average optical densities of sera tested, the ELISA systems using these two recombinant nucleoproteins have good sensitivity and specificity. These results demonstrate the usefulness of ELISA for diagnostics as well as ecological and serosurvey studies of Ebola and Marburg virus infection.  相似文献

Vascular smooth muscle cells (VSMCs) are exposed to mechanical cyclic stretch in vivo, which play important roles in maintenance of vascular homeostasis and regulation of pathological vascular remodeling. Reversible protein phosphorylation is crucial for intracellular signaling transduction. However, the dynamic phosphorylated profile induced by cyclic stretch in VSMCs is still unclear. Using the stable isotope labeling by amino acid in cell culture, VSMCs were labeled and exposed to 10% physiological cyclic stretch in vitro at 1.25 Hz for 0 min, 15 min, 30 min, 1 h and 6 h, respectively. Using TiO2 beads and liquid chromatography tandem mass spectrometry, the temporal phosphoproteomic profiles in response to cyclic stretch were then detected. Bioinformatics analysis including fuzzy c-means clustering, functional classifications, and Ingenuity Pathway Analysis were applied to further reveal the potential mechanotranduction networks. The results indicated that protein kinase C (PKCs) family, Rho-associated coiled-coil containing protein kinase 1 (ROCK1) and Akt may participate in cyclic-stretch induced VSMC functions. Cyclic stretch repressed the expression of ROCK1, while it had no significant effect on the phosphorylation of PKCα/βII, PKCζ/λ and PKCδ/θ. PKCθ was activated first at short time-phase (15 min and 30 min), and again at long time-phase (6 h, 12 h and 24 h). The activation of p-PKCμ was immediate and short-term, similar to p-Akt. Our present in vitro work hence revealed that cyclic stretch activates complex mechanotransduction networks, suggesting that novel mechanoresponsive molecules, i.e., PKCθ, PKCμ, and ROCK1, may participate in the mechanotransduction and modulation VSMC functions.  相似文献

The unique ability of intrinsically disordered proteins (IDPs) to fold upon binding to partner molecules makes them functionally well-suited for cellular communication networks. For example, the folding-binding of different IDP sequences onto the same surface of an ordered protein provides a mechanism for signaling in a many-to-one manner. Here, we study the molecular details of this signaling mechanism by applying both Molecular Dynamics and Monte Carlo methods to S100B, a calcium-modulated homodimeric protein, and two of its IDP targets, p53 and TRTK-12. Despite adopting somewhat different conformations in complex with S100B and showing no apparent sequence similarity, the two IDP targets associate in virtually the same manner. As free chains, both target sequences remain flexible and sample their respective bound, natively -helical states to a small extent. Association occurs through an intermediate state in the periphery of the S100B binding pocket, stabilized by nonnative interactions which are either hydrophobic or electrostatic in nature. Our results highlight the importance of overall physical properties of IDP segments, such as net charge or presence of strongly hydrophobic amino acids, for molecular recognition via coupled folding-binding.  相似文献

选用华南地区广泛应用的8个杂交稻不育系与10个恢复系,按NCII设计配制两套不完全双列杂交组合61个,通过在湛江的两年种植试验,结合62对与产量性状QTL连锁SSR标记,对18个亲本的产量配合力进行综合评价和标记位点鉴定。结果表明:供试亲本间一般配合力存在较大差异,其中不育系珍汕97A、万金A和恢复系直龙、广恢3550的一般配合力相对较高;两套组合共检测出36个与亲本产量配合力显著相关的标记位点,其中RM1、RM216、RM231和RM542为两套材料所共有;检测出的杂合标记位点中,23个为增效标记位点,13个为减效标记位点,其中杂合标记位点RM231、RM209可使F1的单株粒重增加13.50 %和16.43 %。该结果可为华南地区杂交稻育种的亲本改良及选配提供了理论指导。  相似文献
