Coiled-coil motifs provide simple systems for studying molecular self-assembly. We designed two 28-residue peptides to assemble into an extended coiled-coil fiber. Complementary interactions in the core and flanking ion-pairs were used to direct staggered heterodimers. These had "sticky-ends" to promote the formation of long fibers. For comparison, we also synthesized a permuted version of one peptide to associate with the other peptide and form canonical heterodimers with "blunt-ends" that could not associate longitudinally. The assembly of both pairs was monitored in solution using circular dichroism spectroscopy. In each case, mixing the peptides led to increased and concentration-dependent circular dichroism signals at 222 nm, consistent with the desired alpha-helical structures. For the designed fiber-producing peptide mixture, we also observed a linear dichroism effect during flow orientation, indicative of the presence of long fibrous structures. X-ray fiber diffraction of partially aligned samples gave patterns indicative of coiled-coil structure. Furthermore, we used electron microscopy to visualize fiber formation directly. Interestingly, the fibers observed were at least several hundred micrometers long and 20 times thicker than expected for the dimeric coiled-coil design. This additional thickness implied lateral association of the designed structures. We propose that complementary features present in repeating structures of the type we describe promote lateral assembly, and that a similar mechanism may underlie fibrillogenesis in certain natural systems.  相似文献

It is concluded that the permeability of the soybean nodule to gases is not linked to the supply of solutes or water via the phloem to the nodule. Nodule respiration and nitrogenase activity were less affected by diel variation and shading treatments than partitioning to the nodule, as assessed using a non-invasive 11C-based technique. Thus C import to the nodule was not matched to C requirement by the nodule. Transit times of tracer to, and within, the nodulated root increased under conditions of reduced photosynthetic rate. The increase in transit time was interpreted as a reduction in the flux of phloem sap. Thus the fluxes of both water and C to the nodule decreased following a reduction in photosynthetic rate. The change in partitioning of recent photosynthate to soybean roots and nodules in response to changes in photoassimilate availability was also used to assess the 'priority' of these sinks. Partitioning from the leaf to the root system was greatly decreased when photoassimilate availability was limited, indicating that root system priority is lower than that of the shoot, as reported for other systems. However, partitioning of tracer arriving in the root system between the nodulated and non-nodulated zones of the root was not affected by changes in photoassimilate availability, as caused by diel change, shading, or steaming of branch roots. Thus although nodules are sinks of high sink 'activity', they have 'priority' equal to that of other root sinks. It is suggested that there are similar phloem unloading kinetics, despite the very different metabolic destiny of the carbohydrate within the two organs.  相似文献

This paper describes an experimental study of the effects of food supply, growth rates and social interactions on homing by juvenile Atlantic salmon Salmo salar in response to displacement. Groups of five fish were housed in a section of an artificial stream and given either rations allowing maximum growth (the rich condition) or 0·1 of this amount (the poor condition); daily specific growth rates were significantly higher in the rich condition. After a 6-day settlement period, the fish were captured, displaced downstream and their movements recorded over the next 3 h. Prior to displacement, the fish showed a high degree of site fidelity and high levels of aggression. Dominant fish and those with stronger site attachment grew faster prior to displacement, these effects being independent. Following displacement, 24% of all fish returned to their previously favoured site and stayed there, 23% returned home initially, but subsequently moved on, 5% settled in a new site and 49% failed to move. The distribution of responses was identical for the rich and poor conditions, but fish that homed were dominant and had grown faster during the pre-displacement period.  相似文献

Multivariate statistical methods were used to elucidate which environmental factors influence the activity patterns of free-living Malayan pit vipers, Calloselasma rhodostoma. Fourteen adult snakes were implanted with miniature radiotransmitters and located a total of 887 times in 5 months. The pit vipers usually remained coiled on the ground for several consecutive days before moving at night to a new site. Partial correlation tests revealed that the frequency and distance of movements to new sites by tagged snakes were highly positively correlated with ambient relative humidity, but not with rainfall, ambient temperature or the lunar cycle. This finding was corroborated by the frequency with which active non-tagged C. rhodostoma were encountered at night, In each site, the proportion of the snakes' bodies exposed to view was positively correlated with ambient humidity, and the snakes retreated to areas with deeper undergrowth when ambient humidity was low. Overt thermoregulatory behaviour was not observed, and implanted thermosensitive transmitters revealed that the snakes were passive thermoconformers.
These findings seem lo contradict much of the current literature which shows temperature to be the dominant abiotic factor affecting reptilian activity, but most herpetologists have considered only temperate forms. Ambient temperature in our tropical study site was warm and relatively constant throughout the year (mean daily range = 24- 33°C), so the pit vipers could passively maintain body temperature within a fairly narrow range, with a daytime mean of 29.4°C. Ambient relative humidity, on the other hand, was very variable, and confining exposure and activity to periods of high ambient humidity may be necessary to avoid dehydration  相似文献

Articular cartilage has a poor intrinsic capacity for self-repair. The advent of autologous chondrocyte implantation has provided a feasible method to treat cartilage defects. However, the associated drawbacks with the isolation and expansion of chondrocytes from autologous tissue has prompted research into alternative cell sources such as mesenchymal stem cells (MSCs) which have been found to exist in the bone marrow as well as other joint tissues such as the infrapatellar fat pad (IFP), synovium and within the synovial fluid itself. In this work we assessed the chondrogenic potential of IFP-derived porcine cells over a 6 week period in agarose hydrogel culture in terms of mechanical properties, biochemical content and histology. It was found that IFP cells underwent robust chondrogenesis as assessed by glycosaminoglycan (1.47±0.22% w/w) and collagen (1.44±0.22% w/w) accumulation after 42 days of culture. The 1 Hz dynamic modulus of the engineered tissue at this time point was 272.8 kPa (±46.8). The removal of TGF-β3 from culture after 21 days was shown to have a significant effect on both the mechanical properties and biochemical content of IFP constructs after 42 days, with minimal increases occurring from day 21 to day 42 without continued supplementation of TGF-β3. These findings further strengthen the case that the IFP may be a promising cell source for putative cartilage repair strategies.  相似文献

Analytical treatment interruptions (ATIs) of antiretroviral therapy (ART) play a central role in evaluating the efficacy of HIV-1 treatment strategies targeting virus that persists despite ART. However, it remains unclear if ATIs alter the rebound-competent viral reservoir (RCVR), the virus population that persists during ART and from which viral recrudescence originates after ART discontinuation. To assess the impact of ATIs on the RCVR, we used a barcode sequence tagged SIV to track individual viral lineages through a series of ATIs in Rhesus macaques. We demonstrate that transient replication of individual rebounding lineages during an ATI can lead to their enrichment in the RCVR, increasing their probability of reactivating again after treatment discontinuation. These data establish that the RCVR can be altered by uncontrolled replication during ATI.  相似文献

For a long time, tracer profiles observed at various positionsalong the phloem transport pathway have been used to infer detailsabout both the mechanisms and the amount of flow through thispathway within whole plants. But the mechanisms still eludeus. This paper investigates why this is so and shows that althoughthe amount of mechanistic information available from tracerprofiles is rather limited, they do provide reliable quantitativemeasurements of the amount of flow. Detailed quantitative analysis of tracer profiles in barleyleaves is used to illustrate techniques of mechanistic interpretationfrom input-output analysis and indicates the need of two compartmentsto describe carbon flow through a leaf from fixation to phloemloading. Although obesity is a risk factor for development of type 2 diabetes and chemical modification of proteins by advanced glycoxidation and lipoxidation end products is implicated in the development of diabetic complications, little is known about the chemical modification of proteins in adipocytes or adipose tissue. In this study we show that S-(2-succinyl)cysteine (2SC), the product of chemical modification of proteins by the Krebs cycle intermediate, fumarate, is significantly increased during maturation of 3T3-L1 fibroblasts to adipocytes. Fumarate concentration increased > or =5-fold during adipogenesis in medium containing 30 mm glucose, producing a > or =10-fold increase in 2SC-proteins in adipocytes compared with undifferentiated fibroblasts grown in the same high glucose medium. The elevated glucose concentration in the medium during adipocyte maturation correlated with the increase in 2SC, whereas the concentration of the advanced glycoxidation and lipoxidation end products, N(epsilon)-(carboxymethyl)lysine and N(epsilon)-(carboxyethyl)lysine, was unchanged under these conditions. Adipocyte proteins were separated by one- and two-dimensional electrophoresis and approximately 60 2SC-proteins were detected using an anti-2SC polyclonal antibody. Several of the prominent and well resolved proteins were identified by matrix-assisted laser desorption ionization time-of-flight/time-of-flight mass spectrometry. These include cytoskeletal proteins, enzymes, heat shock and chaperone proteins, regulatory proteins, and a fatty acid-binding protein. We propose that the increase in fumarate and 2SC is the result of mitochondrial stress in the adipocyte during adipogenesis and that 2SC may be a useful biomarker of mitochondrial stress in obesity, insulin resistance, and diabetes.  相似文献

1 Mating disruption is the primary tactic used to reduce rates of gypsy moth population spread in the United States Department of Agriculture’s Slow‐the‐Spread of the gypsy moth programme (STS). Because STS targets very low‐density gypsy moth populations within which it is extremely difficult to collect females or egg masses, mating success in native populations cannot be determined. Therefore, the evaluation of mating disruption treatments in field experiments such as those designed to test new formulations and application methods requires deploying and recovering laboratory‐reared female moths to determine mating success. 2 Five methods of deploying females were evaluated for cost, rates of female and egg mass recovery, and female mating success. The deployment methods tested were: modified delta trap, square barrier, single and double trunk bands, and tethered females. 3 Deployment of tethered females had the highest cost and mating success rate, but it did not yield the highest rates of female and egg mass recovery. Deployment of females in delta traps produced the lowest cost and mating success rate, but yielded the highest recovery rate. Neither of these deployment methods is recommended because of unacceptably high cost (tethered female) or low mating success (delta trap). 4 There were no significant differences in cost or mating success among the other three deployment methods. 5 The differences among the square barrier, single trunk band, and double trunk band methods in cost, female and egg mass recovery, and mating success are too small to recommend any one over the others.  相似文献
