Using a bacterial expression system, large amounts of the catalytic core of an atrial natriuretic peptide receptor guanylyl cyclase were produced and purified. After refolding the protein from a buffer containing urea, the enzyme had positively cooperative kinetics with a Hill coefficient, nH = 1.42 +/- 0.08. Size exclusion chromatography and denaturing polyacrylamide gel electrophoresis demonstrated that the enzyme is composed of homodimers with interacting catalytic sites. 相似文献

Neuropeptides identified with a radioimmunoassay specific for the C-terminus of Met5-enkephalin-Arg6-Phe7 (YGGFMRF) have been extracted from nervous tissues of the blowfly Calliphora vomitoria and also from whole flies. Chromatographic characterisation, based on criteria of molecular weight, charge and hydrophobicity, reveals a complex multiplicity of immunoreactive peptides. Variations in the amounts and types of peptides found within different nervous tissues is evidence that the cellular precursor processing is selective. Physiological studies on the isolated blowfly salivary gland show that synthetic YGGFMRF is a potent secretagogue with a maximal rate of fluid secretion induced at a concentration of between 10(-13) and 10(-12) M. The tetrapeptide comprising the last four residues of the C-terminus of YGGFMRF, Phe-Met-Arg-Phe, is equally potent. However, the carboxyamidated variants, YGGFMRF-NH2 and the molluscan cardioacceleratory peptide FMRF-NH2, as well as the opioid peptides Met5- and Leu5-enkephalin, have no activity. Partially purified YGGFMRF-immunoreactive peptides from the blowfly have ED50 values in the bioassay approximating to 0.3 thoracic ganglion, 2.1 hypocerebral ganglion and 3.0 brain equivalents. 相似文献

Nocturnal downstrean migration of juvenile Atlantic salmon is usually interpreted as increased locomotor activity. The frequency of downstream passages of 0–1 + salmon in an endless stream channel was greater by night than by day in both smoking and non-smolting fish in autumn and spring. Movement increased at dusk, and decreased after dawn. Mature male 1 + fish moved slightly less than immatures in October, but significantly more in November. Total movement frequency was lower at full moon than at other moon phases, and movement was reduced when the moon was up. Under turbid conditions by day, the threshold water velocity inducing nett downstream movement was 8.2 cm s−1 , and the relative velocity of fish swimming downstream was never more than one third that of fish holding station at the normal maximal flow of 25–30 cm s−1 .
At the end of their first growing season in October, fish which had been offered food continuously through 24 h did not differ in size from those fed by day only, but the latter were significantly larger than those offered food only at night.
We conclude that: (1) the fish fed actively by day, and not by night; (2) station-holding represented activity, and downstream nocturnal movement represented relative inactivity (displacement) which occurred on loss of visual orientation, hence migration resulted from reduced activity; (3) lack of displacement in early autumn has adaptive value for maturing fish, but not for non-spawners. 相似文献

The effect of particle size of a commercial pelleted feed on the growth of Atlantic salmon from first feeding alevins to first and second year smolts was investigated using 20-day feeding experiments. Ten experiments were performed over a two year period, each comprising six groups of fish separately selected from stock populations. Each group was presented with one of six sizes of food particle ranging from larger than the respective mean mouth breadth (100% feed size), through 50%, 25%, 12·5%, 6·25% to 3·125%. Experiments were performed in six radial flow/circumferential drain tanks under ambient photoperiod and water temperature. Growth rate was found to be closely related to feed size. Maximum growth in each case was shown only on one size of particle; larger and smaller sizes resulted in reduced growth. The particle size for maximum growth increased in direct proportion to fish length. Fish from 4·2 to 20·3 cm in length showed maximum growth on particle diameters 0·022 to 0·026 × fish fork length (PFR). First feeding alevins were found to show comparable growth rate on particle diameters 0·0115 to 0·090 PFR. Some seasonal variation in growth response was indicated. The results are discussed in relation to developmental and seasonal effects. Possible factors affecting the energetics of prey size related differential growth are discussed. 相似文献
