Evidence for a stem cell hierarchy in the adult human breast   总被引:14,自引:0,他引:14       下载免费PDF全文
Cellular pathways that contribute to adult human mammary gland architecture and lineages have not been previously described. In this study, we identify a candidate stem cell niche in ducts and zones containing progenitor cells in lobules. Putative stem cells residing in ducts were essentially quiescent, whereas the progenitor cells in the lobules were more likely to be actively dividing. Cells from ducts and lobules collected under the microscope were functionally characterized by colony formation on tissue culture plastic, mammosphere formation in suspension culture, and morphogenesis in laminin-rich extracellular matrix gels. Staining for the lineage markers keratins K14 and K19 further revealed multipotent cells in the stem cell zone and three lineage-restricted cell types outside this zone. Multiparameter cell sorting and functional characterization with reference to anatomical sites in situ confirmed this pattern. The proposal that the four cell types are indeed constituents of an as of yet undescribed stem cell hierarchy was assessed in long-term cultures in which senescence was bypassed. These findings identify an adult human breast ductal stem cell activity and its earliest descendants.  相似文献

Aims

To examine the long-term risk of hyperthyroidism in patients admitted to hospital with new-onset AF. Hyperthyroidism is a well-known risk factor for atrial fibrillation (AF), but it is unknown whether new-onset AF predicts later-occurring hyperthyroidism.

Methods and Results

All patients admitted with new-onset AF in Denmark from 1997–2009, and their present and subsequent use of anti-thyroid medication was identified by individual-level linkage of nationwide registries. Patients with previous thyroid diagnosis or thyroid medication use were excluded. Development of hyperthyroidism was assessed as initiation of methimazole or propylthiouracil up to a 13-year period. Risk of hyperthyroidism was analysed by Poisson regression models adjusted for important confounders such as amiodarone treatment. Non-AF individuals from the general population served as reference. A total of 145,623 patients with new-onset AF were included (mean age 66.4 years [SD ±13.2] and 55.3% males) of whom 3% (4,620 events; 62.2% women) developed hyperthyroidism in the post-hospitalization period compared to 1% (48,609 events; 82% women) in the general population (n = 3,866,889). In both women and men we found a significantly increased risk of hyperthyroidism associated with new-onset AF compared to individuals in the general population. The highest risk was found in middle-aged men and was consistently increased throughout the 13-year period of observation. The results were confirmed in a substudy analysis of 527,352 patients who had thyroid screening done.

Conclusion

New-onset AF seems to be a predictor of hyperthyroidism. Increased focus on subsequent risk of hyperthyroidism in patients with new-onset AF is warranted.  相似文献

An epithelial cell line, referred to as A163, was established from breast carcinoma derived from a patient with a strong family history of breast cancer but no known breast cancer susceptibility mutation. A163 was propagated in a serum-free culture medium including the epidermal growth factor. Immunophenotypic characterization demonstrated a mixed luminal and basal-like phenotype. When epidermal growth factor was excluded from the culture medium, A163 entered a quiescent period followed by a period of increased cell proliferation in a subpopulation of the cells. The epidermal growth factor-independent subpopulation retained the basal-like phenotype of the parental cell line. Karyotype and fluorescent in situ hybridization analysis showed an amplification of epidermal growth factor receptor on 7q in A163-S1 only, resulting in high expression of total and phosphorylated epidermal growth factor receptor. The A163-S1 sub-line piles up in culture, indicating a loss of contact inhibition. When grown on transwell filters, A163 shows basal expression of P63 and cytokeratin 14, whereas A163-S1 expresses P63 ubiquitously, and has lost the basal specific expression of cytokeratin 14, indicating a loss of polarity. Furthermore, when cultured in reconstituted basement membrane matrix, A163 form polarized normal like acini. In contrast, A163-S1 form large disorganized structures with lack of polarity. These cell lines may prove useful to understand molecular changes in breast cancer progression, in particular basal-like breast cancer subtype with bad prognosis and no current treatment options.  相似文献

Background

Non steroidal anti-inflammatory drugs (NSAIDs) increase mortality and morbidity after myocardial infarction (MI). We examined cause-specific mortality and morbidity associated with NSAIDs in a nationwide cohort of MI patients.

Methods and Results

By individual-level linkage of nationwide registries of hospitalization and drug dispensing from pharmacies in Denmark, patients aged >30 years admitted with first-time MI during 1997–2009 and their subsequent NSAID use were identified. The risk of three cardiovascular specific endpoints: cardiovascular death, the composite of coronary death and nonfatal MI, and the composite of fatal and nonfatal stroke, associated with NSAID use was analyzed by Cox proportional hazard analyses. Of 97,698 patients included 44.0% received NSAIDs during follow-up. Overall use of NSAIDs was associated with an increased risk of cardiovascular death (hazard ratio [HR] 1.42, 95% confidence interval [CI] 1.36–1.49). In particular use of the nonselective NSAID diclofenac and the selective cyclooxygenase-2 inhibitor rofecoxib was associated with increased risk of cardiovascular death (HR 1.96 [1.79–2.15] and HR1.66 [1.44–1.91], respectively) with a dose dependent increase in risk. Use of ibuprofen was associated with increased risk of cardiovascular death (HR 1.34[1.26–1.44]), whereas naproxen was associated with the lowest risk of (e.g., HR 1.27[1.01–1.59].

Conclusion

Use of individual NSAIDs is associated with different cause-specific cardiovascular risk and in particular rofecoxib and diclofenac were associated with increased cardiovascular morbidity and mortality. These results support caution with use of all NSAIDs in patients with prior MI.  相似文献

Thermophilic viruses represent a novel source of genetic material and enzymes with great potential for use in biotechnology. We have isolated a number of thermophilic viruses from geothermal areas in Iceland, and by combining high throughput genome sequencing and state of the art bioinformatics we have identified a number of genes with potential use in biotechnology. We have also demonstrated the existence of thermostable counterparts of previously known bacteriophage enzymes. Here we describe a thermostable RNA ligase 1 from the thermophilic bacteriophage RM378 that infects the thermophilic eubacterium Rhodothermus marinus. The RM378 RNA ligase 1 has a temperature optimum of 60–64°C and it ligates both RNA and single-stranded DNA. Its thermostability and ability to work under conditions of high temperature where nucleic acid secondary structures are removed makes it an ideal enzyme for RNA ligase-mediated rapid amplification of cDNA ends (RLM-RACE), and other RNA and DNA ligation applications.  相似文献

The seasonal variations in biomass, abundance and species composition of zooplankton in relation to hydrography and chlorophyll a were studied in the subarctic waters north of Iceland. The sampling was carried out at approximately monthly intervals from February 1993 to February 1994 at eight stations arranged along a transect extending from 66°16′N–18°50′W to 68°00′N–18°50′W. The mean temperature at 50 m depth showed a clear seasonal pattern, with lowest water temperatures in February (∼1.1°C) and the highest in July (∼5.4°C). The spring growth of the phytoplankton began in late March and culminated during mid-April (∼7.0 mg Chl a m−3). Both the biomass and the abundance of total zooplankton were low during the winter and peaked once during the summer in late May (∼4 g m−2 and ∼38,000 individuals m−2). A total of 42 species and taxonomic groups were identified in the samples. Eight taxa contributed ∼90% of the total zooplankton number. Of these Calanus finmarchicus was by far the most abundant species (∼60% of the total zooplankton). Less important groups were ophiuroid larvae (∼9%), Pseudocalanus spp. (∼8%), Metridia longa (∼4%), C. hyperboreus (∼3%), Acartia longiremis (∼2%), chaetognaths (∼2%) and euphausiid larvae (∼2%). The dominant copepods showed two main patterns in seasonal abundance: C. finmarchicus, C. hyperboreus and C. glacialis had one annual peak in numbers in late May, while Pseudocalanus spp., M. longa and A. longiremis showed two maxima during the summer (July) and autumn (October/November). Ophiuroid larvae and chaetognaths (mainly Sagitta elegans) peaked during the middle of July, while the number of euphausiid eggs and larvae was greatest from May to July. The succession in population structure of C. finmarchicus indicated its main spawning to be in April and May, coincident with the phytoplankton spring bloom. A minor spawning was also observed sometime between August and October. The seasonal variation in abundance and development of Calanusfinmarchicus, Calanus hyperboreus and Calanus glacialis in relationto hydrography and chlorophyll (Chl) a was studied in the Arcticwaters of the East Icelandic Current to the north-east of Icelandfrom March 1995 to February 1996. The sampling was carried outat approximately monthly intervals on a transect of five stationsextending from 67°00'N, 13°55'W to 68°00'N, 12°40'W.In April, May and June, vertical distribution was also investigated.Spring warming of the surface waters began in May, with maximumtemperatures recorded in August (~5°C, mean for uppermost50 m). Below 75 m, temperature remained at <0°C throughoutthe year. The spring bloom of phytoplankton started in earlyMay and the highest Chl a concentrations were measured duringlate May to early June (~1 mg Chl a m-3). Calanus finmarchicusdominated in terms of numbers (~75%), while C. hyperboreus dominatedbiomass (~76%). Calanus glacialis occurred only in low numbers(~1%) and was only a small portion of biomass (~0.7%). The abundanceof all species was low during the winter and peaked once duringsummer: C. finmarchicus in July (~16 000 ind. m-2) and C. glacialisand C. hyperboreus in June (~370 and ~7700 ind. m-2, respectively).The biomass of C. finmarchicus had two maxima, in April (~1.9g m-2) and July (~1.5 g m-2), while C. hyperboreus peaked inJune (~12.3 g m-2). Calanus finmarchicus was estimated to spawnin early May at about the start of the spring bloom, while C.hyperboreus spawned prior to the spring bloom, in late Februaryto early March. On the basis of copepod stage distribution,C. finmarchicus was considered to have a 1-year life cycle andC. hyperboreus at least a 2-year life cycle.  相似文献

The reproductive strategy of most fishes is to produce a large number of tiny eggs, leading to a huge difference between egg size and asymptotic body size. The viability of this strategy is examined by calculating the life-time reproductive success R0 as a function of the asymptotic body size. A simple criterion for the optimality of producing small eggs is found, depending on the rate of predation relative to the specific rate of consumption. Secondly it is shown that the success of the reproductive strategy is increasing with asymptotic body size. Finally the existence of both upper and lower limits on the allowed asymptotic sizes is demonstrated. A metabolic upper limit to asymptotic body size for all higher animals is derived.  相似文献
