The effects of herbivory and the season of disturbance on species composition and algal succession were experimentally tested at a tropical intertidal shore, Phuket Island, Thailand. Dead coral patches were cleared of all organisms during both the dry and rainy seasons in order to study the effects of season on algal succession and cages were set up to exclude fish herbivory. Algal succession in this intertidal habitat showed a simple pattern and took a year from the early Ulva paradoxa C. Agardh stage to the late Polysiphonia sphaerocarpa Børgesen stage. The abundance of algae during succession was under the influence of seasonal change. U. paradoxa reproduced and recruited throughout the year. Caging effects did not apparently influence algal abundance, perhaps because resident herbivorous damselfishes excluded other herbivores from their territories and maintained their algal “farms”. Unexpectedly, the percent cover of Ulva in the caged plots was lower than in uncaged plots. This pattern may indicate that caging excluded damselfishes only, but allowed small herbivores that consumed substantial amounts of soft filamentous algae in the cages.  相似文献

The effects of nutrient enrichment and herbivory on resource allocation patterns among morphology, reproduction, and chemical content of the brown alga, Turbinaria conoides (J. Agardh) Kützing were tested in the shallow subtidal zone of the Gulf of Thailand. The field experimental design comprised 36 plots (50 × 50 cm2) with and without herbivores, and two nutrient levels. Cages (uncaged and fully caged plots) were used to exclude herbivorous fishes and two nutrients levels were achieved by experimental enrichment above ambient nutrient concentrations. For morphology of Turbinaria, the maximum length (holdfast to frond apex), the width of the base of the stipe, number of blades, holdfast and stipe diameter were measured. Biomass, reproductive output, and tissue nutrient (carbon: nitrogen: phosphorus) content of T. conoides were examined. Phlorotannin concentrations were examined using a modified Folin‐Ciocalteu method. The results showed that herbivory had no effect on morphology, reproduction, or phlorotannin concentrations. This could be due to the structural and morphological deterrents of alga, which might minimize grazing effects from herbivorous fishes. Nutrient enrichment had no effect on morphology and reproduction of T. conoides, possibly due to low nutrient demand in Turbinaria. However, nutrient enrichment did affect phlorotannin concentrations, as phlorotannins in the enriched plots were lower than the ambient plots. These results support, in part, the carbon–nutrient balance hypothesis that states algae will allocate fewer resources to production of anti‐herbivore chemical compounds when enriched with potential growth‐limiting nutrients.  相似文献

Artificial membrane‐feeding techniques have replaced direct feeding on animals for the maintenance of malaria and arbovirus vectors in many laboratories. Membrane feeding facilitates controlled experimentation of pathogen transmission during mosquito feeding. Sheep blood is commonly used due to its availability and low cost. We evaluated the impact of blood source (human, guinea pig, sheep, and hamster via direct feeding) on feeding rates, adult survival, fecundity, hatching rates, and developmental times for five species of laboratory‐colonized mosquitoes (Anopheles dirus, An. cracens, An. minimus, An. sawadwongporni, and Ae. aegypti). We found that feeding rates differ among blood sources within mosquito species. Survival, fecundity, and hatching rates were lower in all Anopheles species and Ae. aegypti after membrane feeding on sheep blood. Survival rates seven days post‐feeding on sheep blood were significantly lower (P<0.05) for An. dirus (84.2%), An. minimus (67.2%), An. sawadwongporni (51.5%), and An. cracens (35.5%) relative to other blood sources. An. minimus and An. sawadwongporni laid no eggs by seven days post‐feeding with sheep blood, while An. dirus and An. cracens produced significantly fewer numbers of eggs and demonstrated significantly lower hatching rates relative to what was observed with the other blood sources. These findings support the conclusion that sheep blood is not a suitable blood source for laboratory rearing of Anopheles spp.  相似文献

We investigated changes in serotonin (5-HT) and dopamine (DA) levels and in their distribution patterns in the central nervous system (CNS) and ovary during the ovarian maturation cycle in the Pacific white shrimp, Litopenaeus vannamei. The concentrations of these two neurotransmitters were determined by using high performance liquid chromatography with electrochemical detection. The 5-HT concentration exhibited a gradual increase in the brain and thoracic ganglia during early ovarian stages I, II, and III, reaching a maximum at the mature ovarian stage IV, whereas DA showed its highest concentration at ovarian stage II in the brain and thoracic ganglia and then declined to its lowest concentration at ovarian stage IV. In the ovaries, 5-HT was lowest at ovarian stage I and gradually increased to a peak at ovarian stage IV. Conversely, the concentration of DA was highest at ovarian stages I and II and lowest at ovarian stage IV. In the brain, 5-HT immunoreactivity (−ir) from stage IV and DA-ir from stage II were distributed extensively in neurons of clusters 6, 11, and 17, in fibers, and in the anterior and posterior medial protocerebral, olfactory, antenna II, and tegumentary neuropils. In the circumesophageal, subesophageal, thoracic, and abdominal ganglia, both 5-HT-ir and DA-ir were detected in neuropils and surrounding neurons and fibers. 5-HT-ir and DA-ir were more intense in the thoracic ganglia than in other parts of the CNS. In the ovary, 5-HT-ir exhibited high intensity in late oocytes, whereas DA-ir was more intense in early oocytes. Thus, opposing changes occur in the levels of these two neurotransmitters and in their specific localizations in the CNS and ovary during ovarian maturation, indicating their important involvement in female reproduction.  相似文献

The normal lymphoid organ of Penaeus monodon (which tested negative for WSSV and YHV) was composed of two parts: lymphoid tubules and interstitial spaces, which were permeated with haemal sinuses filled with large numbers of haemocytes. There were three permanent types of cells present in the wall of lymphoid tubules: endothelial, stromal and capsular cells. Haemocytes penetrated the endothelium of the lymphoid tubule's wall to reside among the fixed cells. The outermost layer of the lymphoid tubule was covered by a network of fibers embedded in a PAS-positive extracellular matrix, which corresponded to a basket-like network that covered all the lymphoid tubules as visualized by a scanning electron microscope (SEM). Argyrophilic reticular fibers surrounded haemal sinuses and lymphoid tubules. Together they formed the scaffold that supported the lymphoid tubule. Using vascular cast and SEM, the three dimensional structure of the subgastric artery that supplies each lobe of the lymphoid organ was reconstructed. This artery branched into highly convoluted and blind-ending terminal capillaries, each forming the lumen of a lymphoid tubule around which haemocytes and other cells aggregated to form a cuff-like wall. Stromal cells which form part of the tubular scaffold were immunostained for vimentin. Examination of the whole-mounted lymphoid organ, immunostained for vimentin, by confocal microscopy exhibited the highly branching and convoluted lymphoid tubules matching the pattern of the vascular cast observed in SEM.  相似文献

The unique beauty of spherical aggregation forming algae has attracted much attention from both the scientific and lay communities. Several aegagropilous seaweeds have been identified to date, including the plants of genus Cladophora and Chaetomorpha. However, this phenomenon remains poorly understood. In July 2013, a mass occurrence of spherical Cladophora aggregations was observed in a salt field reservoir in Central Thailand. The aims of the present study were to describe the habitat of the spherical aggregations and confirm the species. We performed a field survey, internal and external morphological observations, pyrenoid ultrastructure observations, and molecular sequence analysis. Floating spherical Cladophora aggregations (1–8 cm in diameter) were observed in an area ~560 m2, on the downwind side of the reservoir where there was water movement. Individual filaments in the aggregations were entangled in each other; consequently, branches growing in different directions were observed within a clump. We suggest that water movement and morphological characteristics promote the formation of spherical aggregations in this species. The molecular sequencing results revealed that the study species was highly homologous to both C. socialis and C. coelothrix. However, the diameter of the apical cells in the study species was less than that of C. coelothrix. The pyrenoid ultrastructure was more consistent with that of C. socialis. We conclude that the study species is C. socialis. This first record of spherical aggregations in this species advances our understanding of these formations. However, further detailed physical measurements are required to fully elucidate the mechanism behind these spherical formations.  相似文献

The tsunami that hit the coast of Thailand along the Andaman Sea on 26 December 2004 caused great damage to human life, property and coastal resources. Here we report the effect of the tsunami on the seaweed community, and its recovery, at Talibong Island, one of the affected areas in Trang province. We made surveys at three sites around the island from April 2004 to January 2006. Fifteen 0.5 m × 0.5 m permanent plots were set up on the west coast of the island to monitor changes in the seaweed community. Eighteen species were found. Sargassum stolonifolium and Laurencia composita were the most abundant, covering 90% and 39%, respectively, of the rocky substrate. Thirteen species varied among sites and seasons. Eleven species, however, were strongly affected by the tsunami. L. composita and Padina sanctae-crucis, for example, were initially washed up onto the shore by the strong wave action, which clearly resulted in a decrease in percentage cover. Also, many permanent plots were covered by sediment causing anoxic conditions, an indirect impact of the tsunami. A difference in species composition revealed a change in overall diversity. Particular morphologies and life forms were more sensitive to the extremes of the tsunami than others. Therefore, the recovery ability of populations of the affected species varied. We did not find any seasonal pattern to the recovery of seaweed after the tsunami within the 13 months of this study.  相似文献

A protease was isolated and purified from Artocarpus heterophyllus (jackfruit) latex and designated as a 48-kDa antimicrobial protease (AMP48) in a previous publication. In this work, the enzyme was characterized for more biochemical and medicinal properties. Enzyme activity of AMP48 was strongly inhibited by phenylmethanesulfonyl fluoride and soybean trypsin inhibitor, indicating that the enzyme was a plant serine protease. The N-terminal amino acid sequences (A-Q-E-G-G-K-D-D-D-G-G) of AMP48 had no sequence similarity matches with any sequence databases of BLAST search and other plant serine protease. The secondary structure of this enzyme was composed of high α-helix (51%) and low β-sheet (9%). AMP48 had fibrinogenolytic activity with maximal activity between 55 and 60°C at pH 8. The enzyme efficiently hydrolyzed α followed by partially hydrolyzed β and γ subunits of human fibrinogen. In addition, the fibrinolytic activity was observed through the degradation products by SDS-PAGE and emphasized its activity by monitoring the alteration of secondary structure of fibrin clot after enzyme digestion using ATR-FTIR spectroscopy. This study presented the potential role to use AMP48 as antithrombotic for treatment thromboembolic disorders such as strokes, pulmonary emboli and deep vein thrombosis.  相似文献

In total, 435 specimens of the Southeast Asian freshwater leech species within the Hirudinidae family were collected from 17 locations of various types of aquatic habitats in northeastern Thailand. They were all morphologically placed within the genus Hirudinaria Whitman, 1886 and there were three distinct species: the common Hirudinaria manillensis, 78.2% of all collected specimens and at all 17 locations, Hirudinaria javanica at 20.3% of collected samples and from five locations and a rarer unidentified morphospecies (Hirudinaria sp.) with six samples from only two locations. The karyotypes of these three species were examined across their range in this study area for 38, 11 and 6 adult specimens of Hirudinaria manillensis, Hirudinaria javanica and Hirudinaria sp., respectively. This revealed different chromosome numbers among all three species, with Hirudinaria javanica having n = 13, 2n = 26, Hirudinaria manillensis lacked one small chromosome pair with n = 12, 2n = 24, and the unknown Hirudinaria sp. differed from any known Hirudinaria karyotypes in exhibiting a higher chromosome number (n = 14, 2n = 28) and a gradual change in size from large to small chromosomes. This suggests that the unknown Hirudinaria sp. is a new biological species. However, phylogenetic analysis based upon a 658 bp fragment of the cytochrome oxidase subunit I gene placed this unknown morphospecies within the Hirudinaria manillensis clade, perhaps then suggesting a recent sympatric speciation, although this requires further confirmation. Regardless, the chromosomes of all three species were asymmetric, most with telocentric elements. A distinct bi-armed chromosome marker was present on the first chromosome pair in Hirudinaria javanica, whilst it was on pairs 1, 2, 3 and 5 in Hirudinaria manillensis, and on pairs 3 and 5 for the unknown Hirudinaria sp.  相似文献
