Abstract. Gas exchange, leaf-nitrogen concentration and water potential were measured in early and late spring in early successional herbaceous plants occurring after cutting and after fire, and in mature woody species from the Mediterranean climax community Quercetum ilicis in central Italy. Net photosynthesis peaked in early spring in all species studied when values for temperature and light were lower but leaf-nitrogen content was higher as compared to late spring, suggesting that nitrogen more than energy input controlled photosynt-hetic rates. Herbaceous pioneer species occurring after cutting showed higher field photo synthetic capacity than evergreen climax trees and shrubs. By contrast, net photosynthesis of herbaceous species occurring in a persistent stage after fire, was in the same range as that of climax trees. This evidence suggests that carbon-gaining appears to be partly related to the dynamic stage of succession and not solely to the growth form. 相似文献

Eco R124I, Eco DXXI and Eco prrI are the known members of the type IC family of DNA restriction and modification systems. The first three are carried on large, conjugative plasmids, while Eco prrI is chromosomally encoded. The enzymes are coded by three genes, hsdR , hsdM and hsdS . Analysis of the DNA sequences upstream and downstream of the type IC hsd loci shows that all are highly homologous to each other and also to sequences present in the bacteriophage P1 genome. The upstream sequences include functional phd and doc genes, which encode an addiction system that stabilizes the P1 prophage state, and extend to and beyond pac , the site at which phage DNA packaging begins. Downstream of the hsd loci, P1 DNA sequences begin at exactly the same place for all of the systems. For Eco DXXI and Eco prrI the P1 homology extends for thousands of base pairs while for Eco R124I an IS 1 insertion and an associated deletion have removed most of the P1-homologous sequences. Immobilization of lipoxygenase in an alginate-silicate solgel matrix: formation of fatty acid hydroperoxides 总被引:3,自引:0,他引:3
A method for the immobilization of lipoxygenase (LOX) in an alginate-silicate gel matrix was developed. In this method, a mixture of calcium alginate beads and LOX in borate buffer are dispersed into a hexane solution of tetramethoxy-ortho-silicate (TMOS). Hydrolysis of the TMOS gives products that permeate and co-polymerize with the alginate gel to form a colloid within the beads that entraps the LOX. Optimum reaction conditions for sol-gel entrapment of LOX are at pH 9.0 in 0.2M borate buffer. The composite gel, after isolation and vacuum drying, had excellent protein retention that has good enzyme activity and stability at room temperature. The activity of the entrapped LOX was less than the activity of the free enzyme. However, the activity of the immobilized LOX can be restored by the addition of borate buffer and glycerol, or borate buffer saturated with an organic solvent. In contrast to the free enzyme in solution, which loses its activity in less than one day, sol-gel entrapped LOX retains its activity at ambient temperature for at least 25 days and can be recycled. This report demonstrates that the sol-gel entrapment method for immobilizing LOX can be useful in developing a process for the oxidation of polyunsaturated fatty acids. 相似文献

Methylobacterium rhodesianum MB 126 was cultivated using extended cultures without outflow. The feeding regime was based on the pH-regulated synchronous dosages of ammonia, methanol, phosphatc and trace elements according to supposed stoichiometric relations. The acidity of the culture medium was kept constant at pH 6.8, whereas the dissolved oxygen concentration was adjusted at 80% of saturation by autoregulation of the stirrer speed. However, besides testing technical conditions, two types of fermentations were discovered which are described in this paper. Firstly, although at the beginning of the bioprocesses the impeller speed increased up to 2,000 rpm, a decrease of dissolved oxygen down to zero was unavoidable. Secondly, methanol was accumulated temporarily up to 44 g/l and 26 g/l at 23 h of fermentation time and without inhibition of growth at least up to 30 g/l or PHB production. During this accumulation of the carbon substrate, exponential growth phases were detected showing growth rates of μ = 0.20/h and 0.21/h. But then, phases of retarded growth followed, whereas the methanol disappeared either continuously or after a steady level. In the course of a 54-h fermentation period, the synthesized PHB amounted to a content of above 50% of cell dry mass. From this data, a volumetric productivity of 0.4 g PHB/lxh was estimated. Moreover, the growth related yield coefficients were calculated to YX/MeOH = 0.21 and YX/MeOH = 0.14, whereas the product related yield coefficients amounted to YPHB/MeOH = 0.12 and YPHB/MeOH = 0,09. Since the shift down of growth rates as well as the production of PHB agreed in time with partial oxygen limitation (40% oxygen saturation), the competition observed between the tricarboxylic acid cycle and PHB synthesis was discussed. Summarizing the results, it was concluded that the frequently described inhibitory effect of methanol of above 2 g/l seems to be rather an effect of experimentally chosen conditions than of a general physiological phenomenon. Therefore, it could be demonstrated that the toxicity of methanol could be overcome if it was not dosed at different times but simultaneously with other medium components. 相似文献

Techniques are described for capturing and marking redtail monkeys (Cercopithecus ascanius). An air-powered darting rifle and syringe darts loaded with a Ketamine-Rompun mixture were used for capture. Ketamine was used for maintaining anesthesia. Monkeys were darted 48 times and captured 27 times. In 24 of the 27 captures, the monkeys were released unharmed. Adult males were marked with radiotransmitters attached to collars of nylon webbing.
