Manipulation of rhizobia microflora for improving legume productivity and soil fertility: A critical assessment 总被引:11,自引:0,他引:11
Inputs of biologically fixed nitrogen derived from the symbiotic relationship between legumes and their root-nodule bacteria into terrestrial ecosystems amount to at least 70 million metric tons per year. It is obvious that this enormous quantity will need to be augmented as the world's population increases and as the natural resources that supply fertilizer nitrogen diminish. This objective will be achieved through the development of superior legume varieties, improvement in agronomic practice, and increased efficiency of the nitrogen fixation process itself by better management of the symbiotic relationship between plant and bacteria. This paper considers ways and means by which populations of root-nodule bacteria, established and introduced, can be manipulated ecologically, agronomically, edaphically and genetically to improve legume productivity and, as a consequence, soil fertility. 相似文献

Thiophene S-oxides have been prepared and their photoreactivity has been studied. In many cases, the sulfur moiety of the thiophene S-oxides is deoxygenated. Here, the corresponding thiophenes or hydroxylated thiophenes are isolated. In the case of the photoirradiation of tert-butyl substituted thiophene S-oxides, the oxygen of the sulfoxy unit is incorporated into the heterocyclic ring system and the corresponding furans have been isolated. Also, structures with two thiophene cores have been oxidized to the respective thiophene S-oxides. These molecules undergo photodeoxygenation, a reaction, which is catalysed by (non-oxidized) thiophenes. 相似文献

To determine features of the steady state [Ca2+]-tension relationship in intact heart, we measured steady force and intracellular [Ca2+] ([Ca2+]i) in tetanized ferret papillary muscles. [Ca2+]i was estimated from the luminescence emitted by muscles that had been microinjected with aequorin, a Ca2+-sensitive, bioluminescent protein. We found that by raising extracellular [Ca2+] and/or by exposing muscles to the Ca2+ channel agonist Bay K 8644, tension development could be varied from rest to an apparently saturating level, at which increases in [Ca2+]i produced no further rise in force. 95% of maximal Ca2+-activated force was reached at a [Ca2+]i of 0.85 +/- 0.06 microM (mean +/- SEM; n = 7), which suggests that the sensitivity of the myofilaments to [Ca2+]i is far greater than anticipated from studies of skinned heart preparations (or from previous studies using Ca2+-sensitive microelectrodes in intact heart). Our finding that maximal force was reached by approximately 1 microM also allowed us to calculate that the steady state [Ca2+]i-tension relationship, as it might be observed in intact muscle, should be steep (Hill coefficient of greater than 4), which is consistent with the Hill coefficient estimated from the entire [Ca2+]i-tension relationship derived from families of variably activated tetani (6.08 +/- 0.68; n = 7). Finally, with regard to whether steady state measurements can be applied directly toward understanding physiological contractions, we found that the relation between steady force and [Ca2+]i obtained during tetani was steeper than that between peak force and peak [Ca2+]i observed during physiological twitches. 相似文献

Temperature modulated effects of nutrients on phytoplankton changes in a mountain lake 总被引:2,自引:0,他引:2
Piburger See, a dimictic mountain lake in Austria, experienced moderate cultural eutrophication in the 1950s. Lake restoration led to a re-oligotrophication in the 1990s with a decrease in seasonal phytoplankton biovolume until the late 1990s, but a reversed trend from the early 2000s onwards. We hypothesize that recent changes in phytoplankton biomass and functional structure are triggered by changes in lake nitrogen and silica concentrations, and we expect climate-related factors to modulate the trophic status of Piburger See. Phytoplankton data were analyzed by non-metric multidimensional scaling (NMDS) applied on biovolume of morpho-functional groups, combined with correlation analyses of environmental variables. Since the 2000s, short-term changes in phytoplankton of Piburger See were explained by varying concentrations and ratios of nitrogen and silica, while the inter-annual variability in phytoplankton species composition was rather attributed to superimposed rising water temperature and lake thermal stability. Our results underline the co-dominant role of phosphorus and nitrogen as phytoplankton drivers in lakes that experience periods of nitrogen limitation. The combined impact of nutrients and climate on phytoplankton development can thus mimic short-term increases in the trophic level of less productive lakes. 相似文献

Epithelial to mesenchymal transition (EMT) and pulmonary fibrogenesis require epithelial integrin α3β1-mediated cross-talk between TGFβ1 and Wnt signaling pathways. One hallmark of this cross-talk is pY654-β-catenin accumulation, but whether pY654-β-catenin is a biomarker of fibrogenesis or functionally important is unknown. To clarify further the role of β-catenin in fibrosis, we explored pY654-β-catenin generation and function. α3β1 was required for TGFβ1-mediated activation of Src family kinases, and Src inhibition blocked both pY654 and EMT in primary alveolar epithelial cells (AECs). TGFβ1 stimulated β-catenin/Lef1-dependent promoter activity comparably in immortalized AECs stably expressing WT β-catenin as well as Y654E or Y654F β-catenin point mutants. But EMT was abrogated in the Tyr to Phe mutant. pY654-β-catenin was sensitive to the axin β-catenin turnover pathway as inhibition of tankyrase 1 led to high AEC axin levels, loss of pY654-β-catenin, and inhibition of EMT ex vivo. Mice given a tankyrase inhibitor (50 mg/kg orally) daily for 7 days beginning 10 days after intratracheal bleomycin had improved survival over controls. Treated mice developed raised axin levels in the lung that abrogated pY654-β-catenin and attenuated lung Snail1, Twist1, α-smooth muscle actin, and type I collagen accumulation. Total β-catenin levels were unaltered. These findings identify Src kinase(s) as a mediator of TGFβ1-induced pY654-β-catenin, provide evidence that pY654-β-catenin levels are a critical determinant of EMT and fibrogenesis, and suggest regulation of axin levels as a novel therapeutic approach to fibrotic disorders. 相似文献

Localized induction of bone formation is essential during orthopedic procedures that involve skeletal repair, such as surgical treatment of non-union bone fractures and degenerative disk disease. Herein we disclose the synthesis and biological evaluation of novel oxysterol derivatives designed as anabolic bone growth agents. Structure-activity relationship studies of oxysterol 4 have identified analogues such as 18, 21 and 30. These new analogues are characterized by higher potency in an osteoblast differentiation assay and/or by increased metabolic stability in human liver microsomes. Prey from the decomposer subsystem may help sustain predator populations in arable fields. Adding organic residues to agricultural
systems may therefore enhance pest control. We investigated whether resource addition (maize mulch) strengthens aboveground
trophic cascades in winter wheat fields. Evaluating the flux of the maize-borne carbon into the food web after 9 months via
stable isotope analysis allowed differentiating between prey in predator diets originating from the above- and belowground
subsystems. Furthermore, we recorded aphid populations in predator-reduced and control plots of no-mulch and mulch addition
treatments. All analyzed soil dwelling species incorporated maize-borne carbon. In contrast, only 2 out of 13 aboveground
predator species incorporated maize carbon, suggesting that these 2 predators forage on prey from the above- and belowground
systems. Supporting this conclusion, densities of these two predator species were increased in the mulch addition fields.
Nitrogen isotope signatures suggested that these generalist predators in part fed on Collembola thereby benefiting indirectly
from detrital resources. Increased density of these two predator species was associated by increased aphid control but the
identity of predators responsible for aphid control varied in space. One of the three wheat fields studied even lacked aphid
control despite of mulch-mediated increased density of generalist predators. The results suggest that detrital subsidies quickly
enter belowground food webs but only a few aboveground predator species include prey out of the decomposer system into their
diet. Variation in the identity of predator species benefiting from detrital resources between sites suggest that, depending
on locality, different predator species are subsidised by prey out of the decomposer system and that these predators contribute
to aphid control. Therefore, by engineering the decomposer subsystem via detrital subsidies, biological control by generalist
