The landscape context of cereal aphid-parasitoid interactions   总被引:10,自引:0,他引:10  
Analyses at multiple spatial scales may show how important ecosystem services such as biological control are determined by processes acting on the landscape scale. We examined cereal aphid-parasitoid interactions in wheat fields in agricultural landscapes differing in structural complexity (32-100% arable land). Complex landscapes were associated with increased aphid mortality resulting from parasitism, but also with higher aphid colonization, thereby counterbalancing possible biological control by parasitoids and lastly resulting in similar aphid densities across landscapes. Thus, undisturbed perennial habitats appeared to enhance both pests and natural enemies. Analyses at multiple spatial scales (landscape sectors of 0.5-6 km diameter) showed that correlations between parasitism and percentage of arable land were significant at scales of 0.5-2 km, whereas aphid densities responded to percentage of arable land at scales of 1-6 km diameter. Hence, the higher trophic level populations appeared to be determined by smaller landscape sectors owing to dispersal limitation, showing the 'functional spatial scale' for species-specific landscape management.  相似文献

The IL-4-inducing principle from Schistosoma mansoni eggs (IPSE/α-1), the major secretory product of eggs from the parasitic worm S. mansoni, efficiently triggers basophils to release the immunomodulatory key cytokine interleukin-4. Activation by IPSE/α-1 requires the presence of IgE on the basophils, but the detailed molecular mechanism underlying activation is unknown. NMR and crystallographic analysis of IPSEΔNLS, a monomeric IPSE/α-1 mutant, revealed that IPSE/α-1 is a new member of the βγ-crystallin superfamily. We demonstrate that this molecule is a general immunoglobulin-binding factor with highest affinity for IgE. NMR binding studies of IPSEΔNLS with the 180-kDa molecule IgE identified a large positively charged binding surface that includes a flexible loop, which is unique to the IPSE/α-1 crystallin fold. Mutational analysis of amino acids in the binding interface showed that residues contributing to IgE binding are important for IgE-dependent activation of basophils. As IPSE/α-1 is unable to cross-link IgE, we propose that this molecule, by taking advantage of its unique IgE-binding crystallin fold, activates basophils by a novel, cross-linking-independent mechanism.  相似文献

European agri-environment schemes encourage farmers to establish sown field margin strips to protect and enhance wild plant diversity. However, plant diversity in such wild plant sowings based on seed mixtures is often low due to the high competitiveness of few, common species. Here we analysed whether intraspecific aggregation could enhance the performance of less competitive species, and how plant performance is influenced by the number of species in a mixture. We focused on inter- and intraspecific competition between six agricultural wild plant species (Centaurea cyanus, Calendula arvensis, Melilotus officinalis, Poa annua, Bromus mollis, Medicago lupulina), and tested (i) two different seeding patterns (intraspecifically aggregated vs. randomly dispersed) and (ii) three different species mixtures (monocultures, three-species, and six-species mixtures). Plant performance was measured in terms of number of individuals, biomass per individual, and biomass per m2. Intraspecific aggregation resulted in higher numbers of individuals of all species, while mixtures generated lower numbers of individuals. The performance of plant species differed depending on their position in the competitive hierarchy. Competitively weak species suffered much less from intraspecific than interspecific competition in terms of biomass, and the competitively weakest species became even excluded in the most species rich and randomly dispersed sowings with high interspecific competition. In conclusion, the performance of wild plant species was influenced by both seeding pattern and number of species in a mixture. Intraspecific aggregation enabled the coexistence of competitively weak species by reducing interspecific competitive exclusion processes. Consequently, agri-environmental schemes designed to preserve and enhance biodiversity should consider small-scale processes influencing the distribution and abundance of plants, and develop new agricultural sowing technologies to cultivate competitively weak and endangered wild plant species.  相似文献

Survey samplings on late summer phytoplankton communities were carried out on 48 high mountain lakes located on the Austrian and Italian slopes of the Eastern Alps. The lakes of North Tyrol (A) and South Tyrol (I) were sampled in 2000 as part of the EU project EMERGE (EVK1-CT-1999- 00032). The lakes of Trentino (I) were investigated in other research projects during 1996 and 1997 (Adamello mountain range) and 2000 (catchment of the River Avisio), respectively. The objectives of this paper are: (1) to study taxonomy and biodiversity of Chrysophyceae, Dinophyceae and Cryptophyceae in high altitude lakes of the Eastern Alps; (2) to identify functional flagellate groups characterising lakes with similar habitat properties, (3) to identify the environmental variables driving abundance and distribution of the three selected algal groups, thus contributing to the selection of sensitive bioindicator taxa. The lakes investigated show rather wide morphological, chemical and trophic state gradients. Flagellate algae account for a median relative abundance (R.A.) of 68%. Chrysophyceae are the most important group in terms of biodiversity and R.A. Special flagellate associations could be related to lake features, like catchment geology, mineralization level and nutrient concentrations. However, the distribution of flagellate algae did not allow a complete geographical separation of the lakes studied in the different districts. Multivariate canonical analyses indicate that the distribution of Chrysophyceae is mainly driven by NO3-N concentration and thermal conditions, while Dinophyceae are driven by a combination of alkalinity, altitude, thermal condition and, less importantly, nutrient concentration. Physical properties of the lakes, such as thermal condition and lake depth, represent the principal driving variables for Cryptophyceae. The responses to the different environmental variables suggest that the three flagellate groups analysed might be used as indicators for environmental changes in high mountain lakes of the Eastern Alps.  相似文献

Many external signals influence growth cone motility, pathfinding, and the formation of synapses that lead to the final map formation of the retinotectal system. Chick temporal retinal ganglion cell axons (RGCs) collapse and retract after encountering posterior tectal cells in vitro. During this process lateral extensions appear along the RGC axonal shaft. Lateral extensions appear as nascent interstitial axonal branches and also as defasciculating growth cones that are trailing along the pioneer axon. RGC branching controlled by repellent tectal cues has recently been shown to be the critical event in retinotectal map development. The intracellular mechanism underlying this phenomenon, however, is not understood. Inhibiting RhoA with either C3 toxin or inhibiting p160Rock kinase, an effector of RhoA, with Y27632 inhibited collapse, retraction, and the number of axons that showed lateral extensions. Lateral extension length increased significantly. Inhibiting Rac1A and cdc42 with cell permeable peptide inhibitors did not inhibit collapse of growth cones, but did inhibit axon retraction. In addition, the number of axons that showed lateral extensions and lateral extension length were significantly reduced. A dynamic cytoskeleton is necessary to react to incoming guidance information. This study addresses the problems of how growth cone motility and branching or defasciculation are affected by Rho‐GTPases as extracellular signals are transmitted to the cytoskeleton. © 2002 Wiley Periodicals, Inc. To understand and predict the efficacy and/or toxicity of liposomal drugs in vivo, it is essential to have rapid, reliable methods of separating and quantitating both the free and the liposomal forms of the drug. A method using solid-phase extraction chromatography columns was developed to separate and quantitate unencapsulated doxorubicin and liposome-associated doxorubicin in plasma following the intravenous injection of liposomal doxorubicin. The method facilitated the recovery and quantitation of free and liposomal drug. The separation and recovery of doxorubicin were linear across the entire range of possible mixtures (0 to 100%) of the two forms of the drug in plasma. Free drug and liposomal drug were readily separated for liposomal doxorubicin systems varying in size (0.1-1.0 microns) and lipid composition (egg yolk phosphatidylcholine/cholesterol and distearylphosphatidylcholine/cholesterol). The method is rapid and allows for multiple samples to be processed simultaneously.  相似文献

The non-covalent homodimer formed by the C-terminal domains of the IgG1 heavy chains (C(H)3) is the simplest naturally occurring model system for studying immunoglobulin folding and assembly. In the native state, the intrachain disulfide bridge, which connects a three-stranded and a four-stranded beta-sheet is buried in the hydrophobic core of the protein. Here, we show that the disulfide bridge is not required for folding and association, since the reduced C(H)3 domain folds to a dimer with defined secondary and tertiary structure. However, the thermodynamic stability of the reduced C(H)3 dimer is much lower than that of the oxidized state. This allows the formation of disulfide bonds either concomitant with folding (starting from the reduced, denatured state) or after folding (starting from the reduced dimer). The analysis of the two processes revealed that, under all conditions investigated, one of the cysteine residues, Cys 86, reacts preferentially with oxidized glutathione to a mixed disulfide that subsequently interacts with the less-reactive second thiol group of the intra-molecular disulfide bond. For folded C(H)3, the second step in the oxidation process is slow. In contrast, starting from the unfolded and reduced protein, the oxidation reaction is faster. However, the overall folding reaction of C(H)3 during oxidative folding is a slow process. Especially, dimerization is slow, compared to the association starting from the denatured oxidized state. This deceleration may be due to misfolded conformations trapped by the disulfide bridge.  相似文献

Gene drives offer the possibility of altering and even suppressing wild populations of countless plant and animal species, and CRISPR technology now provides the technical feasibility of engineering them. However, population-suppression gene drives are prone to select resistance, should it arise. Here, we develop mathematical and computational models to identify conditions under which suppression drives will evade resistance, even if resistance is present initially. Previous models assumed resistance is allelic to the drive. We relax this assumption and show that linkage between the resistance and drive loci is critical to the evolution of resistance and that evolution of resistance requires (negative) linkage disequilibrium between the two loci. When the two loci are unlinked or only partially so, a suppression drive that causes limited inviability can evolve to fixation while causing only a minor increase in resistance frequency. Once fixed, the drive allele no longer selects resistance. Our analyses suggest that among gene drives that cause moderate suppression, toxin-antidote systems are less apt to select for resistance than homing drives. Single drives of moderate effect might cause only moderate population suppression, but multiple drives (perhaps delivered sequentially) would allow arbitrary levels of suppression. The most favorable case for evolution of resistance appears to be with suppression homing drives in which resistance is dominant and fully suppresses transmission distortion; partial suppression by resistance heterozygotes or recessive resistance are less prone to resistance evolution. Given that it is now possible to engineer CRISPR-based gene drives capable of circumventing allelic resistance, this design may allow for the engineering of suppression gene drives that are effectively resistance-proof.  相似文献

In acute ischemic stroke, time from symptom onset to intervention is a decisive prognostic factor. In order to reduce this time, prehospital thrombolysis at the emergency site would be preferable. However, apart from neurological expertise and laboratory investigations a computed tomography (CT) scan is necessary to exclude hemorrhagic stroke prior to thrombolysis. Therefore, a specialized ambulance equipped with a CT scanner and point-of-care laboratory was designed and constructed. Further, a new stroke identifying interview algorithm was developed and implemented in the Berlin emergency medical services. Since February 2011 the identification of suspected stroke in the dispatch center of the Berlin Fire Brigade prompts the deployment of this ambulance, a stroke emergency mobile (STEMO). On arrival, a neurologist, experienced in stroke care and with additional training in emergency medicine, takes a neurological examination. If stroke is suspected a CT scan excludes intracranial hemorrhage. The CT-scans are telemetrically transmitted to the neuroradiologist on-call. If coagulation status of the patient is normal and patient''s medical history reveals no contraindication, prehospital thrombolysis is applied according to current guidelines (intravenous recombinant tissue plasminogen activator, iv rtPA, alteplase, Actilyse).Thereafter patients are transported to the nearest hospital with a certified stroke unit for further treatment and assessment of strokeaetiology. After a pilot-phase, weeks were randomized into blocks either with or without STEMO care. Primary end-point of this study is time from alarm to the initiation of thrombolysis. We hypothesized that alarm-to-treatment time can be reduced by at least 20 min compared to regular care.  相似文献
