Animal models of cardiovascular diseases allow to investigate relevant pathogenetic mechanisms in detail. In the present study, the mutations Asp175Asn and Glu180Gly in alpha-tropomyosin (TPM1), known cause familiar hypertrophic cardiomyopathy (FHC) were studied for changes in hemodynamic parameters and spontaneous baroreflex regulation in transgenic rats in comparison to transgenic and non-transgenic controls by telemetry. Heart rate variability (HRV) and blood pressure variability (BPV) were analyzed using time- and frequency domain, as well as non-linear measures. The dual sequence method was used for the estimation of the baroreflex regulation. In transgenic rats harboring mutated TPM1, changes in HRV were detected during exercise, but not at rest. Both mutations, Asp175Asn and Glu180Gly, caused increased low frequency power. In addition, in animals with mutation Asp175Asn a reduced total HRV was observed. BPV did not show any differences between all transgenic animal lines. During exercise, a strong increase in the number of bradycardic and tachycardic fluctuations accompanied with decreased baroreflex sensitivity (BRS) was detected in animals with either TPM1 mutation, Asp175Asn or Glu180Gly. These data suggest, that the analysis of cardiac autonomic control, particularly of baroreflex regulation, represents a powerful non-invasive approach to investigate the effects of subtle changes in sarcomeric architecture on cardiac physiology in vivo. In case of mutations Asp175Asn or Glu180Gly in TPM1, early detection of alterations in autonomic cardiac control could help to prevent sudden cardiac death in affected persons.  相似文献

Rabbit antiserum against B10.AQR mouse spleen and lymph node cells (RAQR), after appropriate absorption, reacted with Iak-positive spleen and lymph node cells in cytotoxic and complement-fixing indicator systems. It reacted neither with Iak-positive thymocytes nor Iak-negative thymocytes, spleen, and lymph node cells. Cryostat sections of tissue from Iak-positive and Iak-negative mice were incubated with RAQR and either rabbit anti-mouse Ig or rabbit anti-T cell globulin. With the unlabeled antibody enzyme method, RAQR-stained lymphocytes were concentrated in the B-cell regions of spleen and lymph nodes of Iak-positive CBA mice. The tissues of mice bearingI-region haplotypes different fromk were negative. Reticular cells of the T cell-supporting network were also positive in Iak mice, but liver, gall bladder, and testicular cells were not. Macrophages of both Iak-positive and -negative mice were stained by RAQR and also by heat-aggregated, peroxidase-labeled Ig. Iak-positive reticular cells survived 900 R total body irradiation and persisted after grafting with Iak-negative bone marrow. Background  

The RNA interference (RNAi) pathway acts as an innate antiviral immune response in Aedes aegypti, modulating arbovirus infection of mosquitoes. Sindbis virus (SINV; family: Togaviridae, genus: Alphavirus) is an arbovirus that infects Ae. aegypti in the laboratory. SINV strain TR339 encounters a midgut escape barrier (MEB) during infection of Ae. aegypti. The nature of this barrier is not well understood. To investigate the role of the midgut as the central organ determining vector competence for arboviruses, we generated transgenic mosquitoes in which the RNAi pathway was impaired in midgut tissue of bloodfed females. We used these mosquitoes to reveal effects of RNAi impairment in the midgut on SINV replication, midgut infection and dissemination efficiencies, and mosquito longevity.  相似文献

The subarctic environment of northernmost Sweden has changed over the past century, particularly elements of climate and cryosphere. This paper presents a unique geo-referenced record of environmental and ecosystem observations from the area since 1913. Abiotic changes have been substantial. Vegetation changes include not only increases in growth and range extension but also counterintuitive decreases, and stability: all three possible responses. Changes in species composition within the major plant communities have ranged between almost no changes to almost a 50 per cent increase in the number of species. Changes in plant species abundance also vary with particularly large increases in trees and shrubs (up to 600%). There has been an increase in abundance of aspen and large changes in other plant communities responding to wetland area increases resulting from permafrost thaw. Populations of herbivores have responded to varying management practices and climate regimes, particularly changing snow conditions. While it is difficult to generalize and scale-up the site-specific changes in ecosystems, this very site-specificity, combined with projections of change, is of immediate relevance to local stakeholders who need to adapt to new opportunities and to respond to challenges. Furthermore, the relatively small area and its unique datasets are a microcosm of the complexity of Arctic landscapes in transition that remains to be documented.  相似文献

应用典范对应分析(CCA)对长白山北坡静水水体12个样点中28种水甲虫与环境关系的研究表明,长白山5种环境因子中水底腐殖化程度和海拔对水甲虫的分布起主要作用,与排序轴的相关系灵敏分别高达0.8371和0.7206,而水温和植被密度也有较大的影响,在环境因子的影响下不同生境中水甲虫分布的种群不同,深刻斜凹龙虱,端钩切眼龙虱,布朗沟牙甲,沼梭科水甲虫等主要分布在深水区,与水温没有关系。而异毛龙虱和舟型牙甲等与海拔和水泡子的腐殖化程度呈正相关。  相似文献

In this paper,we propose a new system for understanding the biodiversity in different conservation areas.It includes three aspects:the capacity,the connectivity and the quality.The capacity refers to the numbers of biodiversity,including absolute and relative richness of the vegetation types Nv and Dv = (Nv-1)/lnA,species numbers S and richness of species dGI = (S- 1)/lnA,and germ plasm resources within a nature reserve,and also the potential biological living space offered by the natural resource.It comprises the total biological resources in a nature reserve.The connectivity refers to the flux of biodiversity,including similarity and connected status of the vegetation types SILi = 2z/(x + y) and species numbers SIc = 2z/(x + y) among different nature reserves.The quality refers to the stability of biodiversity,including relative species richness index RSLi = d/dmax,relative vegetation richness index RVLi =Dv/Dmaxv,fastness to invasion species fLi = 1-Si/St,weighted values,representativeness and vulnerability of special vegetations,special species,CITES species and rare species as the protected targets.  相似文献

Regulated degradation of circadian clock proteins is a crucial step for rhythm generation per se but also for establishing a normal circadian period. Here, the authors show that the F-box protein beta-transducin repeat containing protein 1 (beta-TrCP1) as part of the E3 ubiquitin ligase complex is an essential component of the mammalian circadian oscillator. Down-regulation of endogenous beta-TrCP1 as well as expression of a dominant-negative form both result in lengthening of the circadian period in oscillating fibroblasts. These phenotypes are due to an impaired degradation of PERIOD (PER) proteins, since expression of beta-TrCP interaction-deficient PER2 variants--but not wild-type PER2--results in a dramatic stabilization of PER2 protein as well as in the disruption of circadian rhythmicity. Mathematical modeling conceptualizes the authors' findings and suggests that loss of sustained rhythmicity in cells with eliminated beta-TrCP-mediated PER2 degradation is due to excessive nuclear repression, a prediction they verified experimentally.  相似文献
