A phylogenetic hypothesis for the Asian newts of the Tylototriton asperrimus group was generated using data from two mitochondrial fragments including COI and the ND1-ND2 regions. Four distinct clades (A, B, C, D) were resolved with high nodal support within this monophyletic group. Clade A included T. asperrimus, T. hainanensis, T. notialis, "T. vietnamensis", and two unnamed salamander populations from Vietnam. Clade A, constituted the sister group of clades B + C. Newly identified clade C likely represents a new cryptic species. Clade C was the sister group of T. wenxianensis. The true T. vietnamensis exclusively constituted clade D. Our results bring into question some previous taxonomic decisions, and a revision is required. This study illustrates the necessity to include samples from type localities in taxonomic studies, and highlights the importance of fine-grained geographical sampling.  相似文献

Respiratory syncytial virus (RSV) is an important cause of severe upper and lower respiratory disease in infants and in the elderly. There are 2 main RSV subtypes A and B. A recombinant vaccine was designed based on the central domain of the RSV-A attachment G protein which we had previously named G2Na (aa130-230). Here we evaluated immunogenicity, persistence of antibody (Ab) response and protective efficacy induced in rodents by: (i) G2Na fused to DT (Diphtheria toxin) fragments in cotton rats. DT fusion did not potentiate neutralizing Ab responses against RSV-A or cross-reactivity to RSV-B. (ii) G2Nb (aa130-230 of the RSV-B G protein) either fused to, or admixed with G2Na. G2Nb did not induce RSV-B-reactive Ab responses. (iii) G2Na at low doses. Two injections of 3 μg G2Na in Alum were sufficient to induce protective immune responses in mouse lungs, preventing RSV-A and greatly reducing RSV-B infections. In cotton rats, G2Na-induced RSV-reactive Ab and protective immunity against RSV-A challenge that persisted for at least 24 weeks. (iv) injecting RSV primed mice with a single dose of G2Na/Alum or G2Na/PLGA [poly(D,L-lactide-co-glycolide]. Despite the presence of pre-existing RSV-specific Abs, these formulations effectively boosted anti-RSV Ab titres and increased Ab titres persisted for at least 21 weeks. Affinity maturation of these Abs increased from day 28 to day 148. These data indicate that G2Na has potential as a component of an RSV vaccine formulation.  相似文献

Background

Dust accumulation on surfaces of critical instruments has been a major concern during lunar and Mars missions. Operation of instruments such as solar panels, chromatic calibration targets, as well as Extra Vehicular Activity (EVA) suits has been severely compromised in the past as a result of dust accumulation and adhesion. Wind storms with wind speeds of up to 70 mph have not been effective in removing significant amounts of the deposited dust. This is indeed an indication of the strength of the adhesion force(s) involved between the dust particles and the surface(s) that they have adhered to. Complications associated with dust accumulation are more severe for non-conducting surfaces and have been the focus of this work.

Methodology

Argon plasma treatment was investigated as a mechanism for lowering dust accumulation on non-conducting polymeric surfaces. Polymers chosen for this study include a popular variety of silicones routinely used for space and terrestrial applications namely RTV 655, RTV 615, and Sylgard 184. Surface properties including wettability, surface potential, and surface charge density were compared before and after plasma treatment and under different storage conditions. Effect of ultraviolet radiation on RTV 655 was also investigated and compared with the effect of Ar plasma treatment.

Conclusion/Significance

Gravimetric measurements proved Ar plasma treatment to be an effective method for eliminating dust adhesion to all three polymers after short periods of exposure. No physical damage was detected on any of the polymer surfaces after Ar plasma treatment. The surface potential of all three polymers remained zero up to three months post plasma exposure. Ultraviolet radiation however was not effective in reducing surface and caused damage and significant discoloration to RTV 655. Therefore, Ar plasma treatment can be an effective and non-destructive method for treating insulating polymeric surfaces in order to eliminate dust adhesion and accumulation.  相似文献

The emergence of the H1N1 swine flu pandemic has the possibility to develop the occurrence of disaster- or drug-resistant viruses by additional reassortments in novel influenza A virus. In the course of an anti-influenza screening program for natural products, 10 xanthone derivatives (1-10) were isolated by bioassay-guided fractionation from the EtOAc-soluble extract of Polygala karensium. Compounds 1, 3, 5, 7, and 9 with a hydroxy group at C-1 showed strong inhibitory effects on neuraminidases from various influenza viral strains, H1N1, H9N2, novel H1N1 (WT), and oseltamivir-resistant novel H1N1 (H274Y) expressed in 293T cells. In addition, these compounds reduced the cytopathic effect of H1N1 swine influenza virus in MDCK cells. Our results suggest that xanthones from P. karensium may be useful in the prevention and treatment of disease by influenza viruses.  相似文献

A MeOH extract of the dry root of Lithospermum erythrorhizon showed strong increasing effect on serine palmitoyltransferase (SPT) in normal human keratinocyte cells (HaCaT cells). Bioassay-guided separation on this extract using repeated chromatography resulted in the isolation of lithospermic acid (1) and two derivative esters, 9″-methyl lithospermate (2) and 9′-methyl lithospermate (3). Compounds 13 significantly increased SPT expressions in the relative quantity (%) of SPT1 mRNA as well as SPT2 mRNA. These constituents also raised the level of SPT protein in HaCaT cells in a dose-dependent manner, with the increased level of SPT protein in HaCaT cells of 55%, 23%, and 81% at the concentration of 100 μg/ml, respectively. This finding suggests that lithospermic acid and its derivatives from L. erythrorhizon might improve the permeability barrier by stimulating the protein level of SPT.  相似文献

The extent of mitochondrial and peroxisomal contribution to beta-oxidation of 18-, 20- and 24-carbon n-3 and n-6 polyunsaturated fatty acids (PUFAs) in intact rat hepatocytes is not fully clear. In this study, we analyzed radiolabeled acid soluble oxidation products by HPLC to identify mitochondrial and peroxisomal oxidation of 24:5n-3, 18- and 20-carbon n-3 and n-6 PUFAs. Mitochondrial fatty acid oxidation produced high levels of ketone bodies, tricarboxylic acid cycle intermediates and CO(2), while peroxisomal beta-oxidation released acetate. Inhibition of mitochondrial fatty acid oxidation with 2-tetradecylglycidic acid (TDGA), high amounts of [14C]acetate from oxidation of 24:5n-3, 18- and 20-carbon PUFAs were observed. In the absence of TDGA, high amounts of [14C]-labeled mitochondrial oxidation products were formed from oxidation of 24:5n-3, 18- and 20-carbon PUFAs. With 18:1n-9, high amounts of mitochondrial oxidation products were formed in the absence of TDGA, and TDGA strongly suppressed the oxidation of this fatty acid. Data of this study indicated that a shift in the partitioning from mitochondrial to peroxisomal oxidation differed for each individual fatty acid and is a specific property of 24:5n-3, 18- and 20-carbon n-3 and n-6 PUFAs.[14C]22:6n-3 was detected with [3-14C]24:5n-3, but not with [1-14C]24:5n-3 as the substrate, while [14C]16:0 was detected with [1-14C]24:5n-3, but not with [3-14C]24:5n-3 as the substrate. Furthermore, the amounts of 14CO(2) were similar when cells were incubated with [3-14C]24:5n-3 versus [1-14C]24:5n-3. These findings indicated that the proportion of 24:5n-3 oxidized in mitochondria was high, and that 24:5n-3 and 24:6n-3 were mostly beta-oxidized only one cycle in peroxisomes.  相似文献

The role of mosquitoes and moths as pollinators of Habenaria obtusata was studied in northern Wisconsin during the summer of 1969. In each population studied, meter-square quadrants were marked and the number of plants, flowers per inflorescence, rate and time of pollinia removed, and capsule set recorded. Carbon dioxide and modified New Jersey Light Traps were set during the flowering period to capture pollinia-bearing insects. The study showed that female mosquitoes of the genus Aedes (particularly A. communis) and two species of Geometrid moths of the genus Xanthorhoe are important pollinators of H. obtusata. Pollination occurred during the last few days of flowering with an average of 18.0% of the pollinia removed and with 14.2% capsule set.  相似文献

BACKGROUND AND AIMS: The mutualistic interaction between insects and flowers is considered to be a major factor in the early evolution of flowering plants. The Schisandraceae were, until now, the only family in the ANITA group lacking information on pollination biology in natural ecosystems. Thus, the objective of this research was to document the pollination biology and breeding system of Schisandra henryi. METHODS: Field observations were conducted in three populations of S. henryi and the floral phenology, floral characters and insect activities were recorded. Floral fragrances were sampled in the field and analysed using TCT-GC-MS. Floral thermogenesis was measured with a TR-71U Thermo Recorder. Pollen loads and location of pollen grains on insect bodies (including the gut) were checked with a scanning electron microscope and under a light microscope. KEY RESULTS: Schisandra henryi is strictly dioecious. Male flowers are similar to female flowers in colour, shape, and size, but more abundant than female flowers. The distance between tepals and the androecium or gynoecium is narrow. Neither male nor female flowers are fragrant or thermogenic. Schisandra henryi is pollinated only by adult female Megommata sp. (Cecidomyiidae, Diptera) that eat the pollen grains as extra nutrition for ovary maturation and ovipositing. Both male and female flowers attract the pollinators using similar visual cues and thus the female flowers use deceit as they offer no food. CONCLUSIONS: Schisandra henryi exhibits a specialized pollination system, which differs from the generalized pollination system documented in other ANITA members. Pollen is the sole food resource for Megommata sp. and the female flowers of S. henryi attract pollinators by deceit. This is the first report of predacious gall midges utilizing pollen grains as a food source. The lack of floral thermogenesis and floral odours further enforces the visual cues by reducing attractants for other potential pollinators.  相似文献

Sleep deprivation (SD) leads to impairments in cognitive function. Here, we tested the hypothesis that cognitive changes in the sleep-deprived brain can be explained by information processing within and between large-scale cortical networks. We acquired functional magnetic resonance imaging (fMRI) scans of 20 healthy volunteers during attention and executive tasks following a regular night of sleep, a night of SD, and a recovery nap containing nonrapid eye movement (NREM) sleep. Overall, SD was associated with increased cortex-wide functional integration, driven by a rise of integration within cortical networks. The ratio of within versus between network integration in the cortex increased further in the recovery nap, suggesting that prolonged wakefulness drives the cortex towards a state resembling sleep. This balance of integration and segregation in the sleep-deprived state was tightly associated with deficits in cognitive performance. This was a distinct and better marker of cognitive impairment than conventional indicators of homeostatic sleep pressure, as well as the pronounced thalamocortical connectivity changes that occurs towards falling asleep. Importantly, restoration of the balance between segregation and integration of cortical activity was also related to performance recovery after the nap, demonstrating a bidirectional effect. These results demonstrate that intra- and interindividual differences in cortical network integration and segregation during task performance may play a critical role in vulnerability to cognitive impairment in the sleep-deprived state.
