The purpose of the current experiment was to investigate the impacts of grinding and pelleting procedures applied to wheat in a wheat–rapeseed meal diet on the coefficients of standardized ileal digestibility, i.e., apparent digestibility corrected for basal endogenous losses (CSID), and true ileal digestibility, i.e., apparent digestibility corrected for total endogenous losses (CTID), of nitrogen (N) and amino acids (AA) in pigs. Ileal digestibility was measured by collecting digesta from pigs fitted with ileorectal anastomoses. Four diets, involving four technological treatments applied to wheat, were compared in vivo according to a 4 × 4 Latin square design (four pigs each fed four diets during four successive periods of 1 week). The technological treatments of wheat were two grinding procedures and two pelleting processes. Wheat was ground to obtain mean flour particle sizes of 1000 and 500 μm, leading after mixing with rapeseed meal and minerals-vitamins premix to the first and second diets named “coarse” and “fine”, respectively. Part of the 500 μm wheat flour was pelleted through dies of same screen diameter (4 mm) but different thicknesses, 16 and 20 mm, inducing a low and high compression ratio, leading after mixing with rapeseed meal and premix to the third and fourth diets named “LCR” and “HCR”, respectively. Basal endogenous losses were determined by feeding a protein-free diet during the 5th week of the experiment. Total endogenous losses were measured by way of the isotopic dilution method using 15N-labeled wheat and rapeseed meal. Decreasing wheat particle size from 1000 to 500 μm improved (P<0.05) the coefficient of ileal digestibility of dietary energy (0.707 versus 0.665), organic matter (0.718 versus 0.677) and dry matter (0.681 versus 0.645), but neither AA CSID nor N retention. The pelleting processes did not further increase (P>0.10) energy or organic matter digestibility but improved (P<0.05) N and AA CSID (0.785 versus 0.759 for N and 0.725 versus 0.679 for lysine, with HCR versus fine diet, respectively). Pelleting wheat flour at higher compression ratio (HCR versus LCR diet) was more efficient to improve dietary N and AA digestibility values due to a significant decrease in ileal specific, i.e., total minus basal, N and AA endogenous losses (P<0.05) associated with an increase in CTID. Q fever is a zoonosis caused by Coxiella burnetii, a bacterium largely carried by ruminants and shed into milk, vaginal mucus, and feces. The main potential hazard to humans and animals is due to shedding of bacteria that can then persist in the environment and be aerosolized. The purpose of this study was to evaluate shedding after an outbreak of Q fever abortion in goat herds and to assess the relationship with the occurrence of abortions and antibody responses. Aborting and nonaborting goats were monitored by PCR for C. burnetii shedding 15 and 30 days after the abortion episodes. PCR analysis of all samples showed that 70% (n = 50) of the aborting and 53% (n = 70) of the nonaborting goats were positive. C. burnetii was shed into vaginal mucus, feces, and milk of 44%, 21%, and 38%, respectively, of goats that aborted and 27%, 20%, and 31%, respectively, of goats that delivered normally. Statistical comparison of these shedding results did not reveal any difference between these two groups. PCR results obtained for the vaginal and fecal routes were concordant in 81% of cases, whereas those for milk correlated with only 49% of cases with either vaginal or fecal shedding status. Serological analysis, using enzyme-linked immunosorbent assay (ELISA), indirect immunofluorescence assay (IFA), and complement fixation tests, showed that at least 24% of the seronegative goats shed bacteria. Positive vaginal and fecal shedding, unlike positive milk shedding, was observed more often in animals that were weakly positive or negative by ELISA or IFA. Two opposite shedding trends were thus apparent for the milk and vaginal-fecal routes. Sulfation and phosphorylation are post-translational modifications imparting an isobaric 80-Da addition on the side chain of serine, threonine, or tyrosine residues. These two post-translational modifications are often difficult to distinguish because of their similar MS fragmentation patterns. Targeted MS identification of these modifications in specific proteins commonly relies on their prior separation using gel electrophoresis and silver staining. In the present investigation, we report a potential pitfall in the interpretation of these modifications from silver-stained gels due to artifactual sulfation of serine, threonine, and tyrosine residues by sodium thiosulfate, a commonly used reagent that catalyzes the formation of metallic silver deposits onto proteins. Detailed MS analyses of gel-separated protein standards and Escherichia coli cell extracts indicated that several serine, threonine, and tyrosine residues were sulfated using silver staining protocols but not following Coomassie Blue staining. Sodium thiosulfate was identified as the reagent leading to this unexpected side reaction, and the degree of sulfation was correlated with increasing concentrations of thiosulfate up to 0.02%, which is typically used for silver staining. The significance of this artifact is discussed in the broader context of sulfation and phosphorylation site identification from in vivo and in vitro experiments. 相似文献

The relationship between different levels of integration is a key feature for understanding the genotype-phenotype map. Here, we describe a novel method of integrated data analysis that incorporates protein abundance data into constraint-based modeling to elucidate the biological mechanisms underlying phenotypic variation. Specifically, we studied yeast genetic diversity at three levels of phenotypic complexity in a population of yeast obtained by pairwise crosses of eleven strains belonging to two species, Saccharomyces cerevisiae and S. uvarum. The data included protein abundances, integrated traits (life-history/fermentation) and computational estimates of metabolic fluxes. Results highlighted that the negative correlation between production traits such as population carrying capacity (K) and traits associated with growth and fermentation rates (Jmax) is explained by a differential usage of energy production pathways: a high K was associated with high TCA fluxes, while a high Jmax was associated with high glycolytic fluxes. Enrichment analysis of protein sets confirmed our results.This powerful approach allowed us to identify the molecular and metabolic bases of integrated trait variation, and therefore has a broad applicability domain. 相似文献

Chelant-enhanced phytoextraction has given variable and often unexplained experimental results. This work was carried out to better understand the mechanisms of Cd plant uptake in the presence of EDTA and to evaluate the contributions of Cd-EDTA complexes to the uptake.Method
A 1-D mechanistic model was implemented, which described the free Cd2+ root absorption, the dissociation and the direct absorption of the Cd-EDTA complexes. It was used to explain Cd uptake by maize in hydroponics and in soil.Results
In hydroponics, the addition of EDTA caused a decrease in Cd uptake by maize, particularly when the ratio of total EDTA ([EDTA] T ) to total Cd ([Cd] T ) was greater than 1. At [Cd] T = 1 μM, when [EDTA] T /[Cd] T < 1, the model indicated that Cd uptake was predominantly due to the absorption of free Cd2+, whose pool was replenished by the dissociation of Cd-EDTA. When [EDTA] T /[Cd] T > 1, the low Cd uptake was mostly due to Cd-EDTA absorption. In soil spiked with 5 mg Cd kg?1, Cd uptake was not affected by the various EDTA additions, because of the buffering capacity of the soil solid phase.Conclusions
Addition of EDTA to soil increases Cd solubility but dissociation of Cd-EDTA limits the availability of the free Cd2+ at the root surface, which finally reduces the plant uptake of the metal. 相似文献149.

Alternatively activated macrophages have been implicated in the therapeutic activity of biodegradable chitosan on wound healing, however, the mechanisms of phenotypic differentiation are still unclear.In vitro, macrophages stimulated with high doses of chitosan (?500 μg/mL) were reported to produce low-level markers associated with alternative activation (arginase-1) as well as classical activation (nitric oxide), and to undergo apoptosis. In this study, we tested the hypothesis that 40 kDa biodegradable chitosan (5–500 μg/mL) is sufficient to polarize mouse bone marrow-derived macrophages (BMDM) in vitro to an alternatively activated phenotype. Control cultures were stimulated with IL-4 (alternative activation), IFN-γ/LPS (classical activation), 1 μm diameter latex beads (phagocytosis), or left untreated. After 48 h of in vitro exposure, BMDM phagocytosed fluorescent chitosan particles or latex beads, and remained viable and metabolically active, although some cells detached with increasing chitosan and latex bead dosage. Arginase-1 was over 100-fold more strongly induced by IL-4 than by chitosan, which induced only sporadic and weak arginase-1 activity over untreated BMDM, and no nitric oxide. IFN-γ/LPS stimulated nitric oxide production and arginase-1 activity and high concentrations of inflammatory cytokines (IL-6, IL-1β, TNF-α, MIP-1α/MIP-1β), while latex beads stimulated nitric oxide and not arginase-1 activity. Chitosan or latex bead exposure, but not IL-4, tended to promote the release of several chemokines (MIP-1α/β, GM-CSF, RANTES, IL-1β), while all treatments promoted MCP-1 release. These data show that chitosan phagocytosis is not sufficient to polarize BMDM to the alternative or the classical pathway, suggesting that biodegradable chitosan elicits alternatively activated macrophages in vivo through indirect mechanisms. 相似文献

The secretory pathway maintains multiple quality control checkpoints. Initially, endoplasmic reticulum-associated degradation pathways monitor protein folding to retain and eliminate aberrant products. Despite its broad client range, some molecules escape detection and traffic to Golgi membranes. There, a poorly understood mechanism termed Golgi quality control routes aberrant proteins for lysosomal/vacuolar degradation. To better understand Golgi quality control, we examined the processing of the obligate substrate Wsc1p. Misfolded Wsc1p does not use routes of typical vacuolar membrane proteins. Instead, it partitions into intralumenal vesicles of the multivesicular body (MVB) pathway, mediated by the E3 ubiquitin ligase Rsp5p. Its subsequent transport to the vacuolar lumen is essential for complete molecule breakdown. Surprisingly, the transport mode plays a second crucial function in neutralizing potential substrate toxicity. Eliminating the MVB sorting signal diverted molecules to the vacuolar limiting membrane, resulting in the generation of toxic by-products. These data demonstrate a new role of the MVB pathway in protein quality control. 相似文献
