Summary Mutations at the URA3 locus of Saccharomyces cerevisiae can be obtained by a positive selection. Wild-type strains of yeast (or ura3 mutant strains containing a plasmid-borne URA3 + gene) are unable to grow on medium containing the pyrimidine analog 5-fluoro-orotic acid, whereas ura3 mutants grow normally. This selection, based on the loss of orotidine-5-phosphate decarboxylase activity seems applicable to a variety of eucaryotic and procaryotic cells.  相似文献

Transmitter glutamate/aspartate synthesis is known to proceed along different metabolic pathways. In this light, the functional relevance of glutamate dehydrogenase in postnatally maturing glutamatergic/aspartatergic structures was studied by means of quantitative enzyme histochemistry. The basic requirements concerning the kinetics and calibration of the histochemical glutamate dehydrogenase reaction used were proved to be met in order to obtain valid quantitative data. The histochemically demonstrable activity of glutamate dehydrogenase (EC 1.4.1.3) in the hippocampal formation of the rat increased markedly during postnatal development. On day 30, the distribution pattern observed was similar to that in adult animals. While the enzyme activity rose within cell body layers from day 0 to day 30 by 240-285%, the increase in neuropil layers was found to be up to 830%. Maximum values were seen in the stratum lacunosum-moleculare of CA1 and CA3 and the stratum moleculare of the dentate fascia on day 30. Since the hippocampal neuropil is supposed to be copiously provided with glutamatergic (and aspartatergic?) structures which become functional in rats during the first weeks of postnatal life, the increase in enzyme activity is discussed to be primarily a consequence of maturing synaptic systems using glutamate and/or aspartate as transmitters.  相似文献

Summary We have traced the central projections of the receptor neurons associated with each of the eleven largest taste hairs on the labellum of the blowfly, Phormia regina (Meigen), by staining them with cobaltous lysine. The eleven hairs fall into three groups which reflect their peripheral locations and their branching patterns in the subesophageal ganglion. Group 1, consisting of the anterior hairs (numbers 1 and 2) and Group 3, consisting of the posterior hairs (numbers 9–11) project bilaterally, while Group 2, consisting of the middle hairs (numbers 3–8) projects primarily ipsilaterally. The central projections of the hairs within a single group are similar. Each hair houses four chemoreceptors, which have differing chemical sensitivities and behavioral roles, and one mechanoreceptor. In some cases, there were indications that the different cells within a single hair have different central branching patterns. For some hairs, however, it was clear that a single central branching region and pattern was shared by more than one receptor cell. We failed to find either a continuous somatotopic representation of a hair's position on the periphery, or an anatomical segregation of receptors coding for different modalities. Behavioral experiments indicate that the fly is informed both of the identity of the hair stimulated and of the chemical nature of the stimulus. Our results suggest that this information is not represented on a gross anatomical level.  相似文献

Summary Recordings were made from an identified auditory neuron, the omega neuron, in the cricketTeleogryllus oceanicus. Models of the conspecific calling song and of the song of another species were presented either singly or simultaneously, and the degree to which the temporal pattern of the conspecific model was encoded in the neuron's spike train was determined. When a single stimulus was presented alone, its temporal pattern was faithfully reflected by the cells's spiking activity, no matter what the azimuth of the broadcasting loudspeaker (Fig. 3). When two stimuli were presented simultaneously from opposite sides, encoding of the pattern ipsilateral to the recorded neuron was interfered with only slightly by the contralateral pattern, as long as the two loudspeakers were sufficiently separated (Figs. 2, 3, 4). When the loudspeakers were each 15° from the cricket's midline, however, the encoding of the temporal pattern of the ipsilateral song model was severely disrupted (Figs. 3, 4). Bilateral interactions are important in determining the response level of the neuron, but do not appear to contribute to the direction-selective encoding of the stimulus temporal pattern (Figs. 5, 6).Phonotactic steering movements of tethered, flying crickets were recorded under stimulus conditions similar to those used in the neurophysiological tests. Under one-stimulus conditions, crickets attempted to turn towards the conspecific model for all tested speaker locations. The heterospecific model elicited reliable steering behavior when it was broadcast from azimuths of 90° and 60°, but often failed to elicit consistent responses when the speaker was positioned closer to the cricket's midline (Figs. 7, 8A and 8B). Responses to the heterospecific pattern were smaller in amplitude than those to the conspecific song model (Figs. 7, 8B). Under two-stimulus conditions, the conspecific model was consistently preferred over the heterospecific song for all tested speaker locations in half the tested crickets. In the remaining animals, preference for the conspecific pattern was only evident for the larger loudspeaker azimuths (Figs. 7, 8C).These results demonstrate that simultaneouslypresented stimuli can be represented separately in the nervous system as a consequence of auditory directionality. It is postulated that the cricket's ability to choose between these stimuli may result from the interactions between two bilaterallypaired song recognizers, each of which may be driven primarily by sound stimuli from one side.  相似文献

Rat pheochromocytoma contains a protein kinase activity which remains associated with tyrosine hydroxylase (TH) during its purification. The incorporation of phosphate in TH is observed after incubation of TH with labelled ATP and magnesium without the need for an exogenous protein kinase. This Ca2+ and cAMP-independent kinase activity is different from previously described TH phosphorylating kinases from rat pheochromocytoma and other tissues.  相似文献

The rise time, of Signal IIf and the decay time of P-680+ have been measured kinetically as a function of pH by using EPR. The Photosystem II-enriched preparations which were used as samples were derived from spinach chloroplasts, and they evolved oxygen before Tris washing. The onset kinetics of Signal IIf are in agreement, within experimental error, with the fast component of the decay of an EPR signal attributable to P-680+. The signal IIf rise kinetics also show good agreement with published values of the pH dependence of the decay of P-680+ measured optically (Conjeaud, H. and Mathis, P. (1980) Biochim. Biophys. Acta 590, 353–359). These results are consistent with a model where the species Z (or D1) responsible for Signal IIf is the immediate electron donor to P-680+ in tris-washed Photosystem II fragments.  相似文献

Two hours after the addition of l-methionine-dl-sulfoximine to the cell suspension, glutamine synthetase activity was inhibited by more than 90% in air-grown Chlamydomonas reinhardii. Cells continued to take up NH3 from the medium provided that the concentration of dissolved CO2 was high (equilibrated with 4% CO2 in air). This NH3 uptake, about 30% of the control, is discussed in terms of glutamate dehydrogenase activity. Without CO2, or with a low CO2 level, a NH3 excretion was observed, the rate of which depended on the actual concentration of the dissolved CO2. Experiments with 15NH3 demonstrated that no NH3 uptake was masked by this excretion and inversely that no excretion occurred during the uptake in the conditions where it took place. Furthermore, the NH3 excretion observed in the absence of CO2 increased when O2 concentration rose to 15% and was inhibited when 10 millimolar isonicotinic acid hydrazide was supplied to the algal suspension. Thus, NH3 excretion in the presence of l-methionine-dl-sulfoximine seems to be related to a photorespiratory process inasmuch as it presents the same properties with regard to the O2 and the isonicotinic acid hydrazide effects. These results favor the hypothesis that NH3 produced in the medium originates from the glycine to serine reaction. On the other hand, partial inhibition (50%) of photosynthesis by l-methionine-dl-sulfoximine was attributed to uncoupling between electron transfer and photophosphorylation due to NH3 accumulation into the cell.  相似文献

In a masked, randomized, concurrently controlled clinical trial we compared the therapeutic response of two regimens in which the same topical corticoid was used to treat 52 men who had stable psoriasis. One regimen, called reduced dose, consisted of once-a-day application of a representative potent fluorinated topical steroid ointment, fluocinonide (Lidex), combined with three-times-a-day application of its vehicle. The other regimen, called traditional dose, consisted of four-times-a-day topical application of the same steroid. Patients were assigned to one of the two regimens and observed for six weeks. Confidence intervals for the difference in true mean response under these two regimens provide good evidence that for these patients the traditional dose was not clinically superior to the reduced dose.  相似文献

Core histones, (H2A,H2B,H3,H4)2, were reconstituted with the synthethic polynucleotides poly(dA-dT)·poly(dA-dT) and poly(dG-dC)·poly(dG-dC) to yield synthetic chromatins containing 200 basepairs per octamer. These synthetic chromatins displayed a 36% decrease in the circular dichroism (CD) peak ellipticity from the value of the polynucleotide free in solution; the poly(dA-dT)·poly(dA-dT)/chromatin showed an increase in the complexity of the thermal denaturation profile compared to that of the polynucleotide. Both the temperature of maximum dhdT for each transition (Tm) and the relative amount of poly(dA-dT)·poly(dA-dT) in the synthetic chromatin melting in each of the four thermal transitions is a function of the ionic strength over the 0–5 mM sodium phosphate range (0.25 mM EDTA, pH 7.0); a shift of material toward higher melting transitions was observed with increasing ionic strength. The CD peak ellipticity value for both synthetic chromatins was ionic strength-independent over the 0–5 mM sodium phosphate range. These results are in contrast to those observed with H1H5 stripped chicken erythrocyte chromatin (Fulmer, A. and Fasman, G.D. (1979) Biopolymers 18, 2875–2891), where an ionic strength dependence was found. Differences in the CD spectra between poly(dA-dT)·poly(dA-dT)/chromatin, poly(dG-dC)·poly(dG-dC)/chromatin and H1H5 stripped chicken erythrocyte chromatin suggest subtle differences in assembly. Finally, the temperature dependence of the CD spectra of poly(dA-dT)·poly(dA-dT)-containing synthetic chromatin, which is similar to that for the polynucleotide, suggests the core histone bound polynucleotide has a large degree of conformational flexibility allowing it to undergo the premelt transition.  相似文献
