Translation initiation has been described as a key step for the control of growth and differentiation of several protozoan parasites in response to environmental changes. This occurs by the activation of protein kinases that phosphorylate the alpha subunit of the translation initiation factor 2 (eIF2α), which decreases translation, and in higher eukaryotes favors the expression of stress remedial response genes. However, very little is known about the signals that activate eIF2α kinases in protozoan parasites. Here, we characterized an eIF2α kinase of Trypanosoma cruzi (TcK2), the agent of Chagas’ disease, as a transmembrane protein located in organelles that accumulate nutrients in proliferating parasite forms. We found that heme binds specifically to the catalytic domain of the kinase, inhibiting its activity. In the absence of heme, TcK2 is activated, arresting cell growth and inducing differentiation of proliferative into infective and non-proliferative forms. Parasites lacking TcK2 lose this differentiation capacity and heme is not stored in reserve organelles, remaining in the cytosol. TcK2 null cells display growth deficiencies, accumulating hydrogen peroxide that drives the generation of reactive oxygen species. The augmented level of hydrogen peroxide occurs as a consequence of increased superoxide dismutase activity and decreased peroxide activity. These phenotypes could be reverted by the re-expression of the wild type but not of a TcK2 dead mutant. These findings indicate that heme is a key factor for the growth control and differentiation through regulation of an unusual type of eIF2α kinase in T. cruzi.  相似文献

Understanding the factors that control uptake rates and allocation of chemical elements among plant organs is a fundamental prerequisite to improve phytostabilization techniques of hazardous elements in contaminated areas. The present study shows evidence that different substrate textures (coarse and fine laterite) do not significantly change the partitioning of root and shoot dry biomass and with few exceptions, do not significantly affect the final average concentration of elements in Eremanthus erythropappus, but change the root:shoot allocation of both essential nutrients and elements potentially toxic to biota. Growth on coarse laterite resulted in significant higher K (30%), Mg (34%), P (25%), S (32%), Cu (58%), and Na (43%) concentrations in roots and lower Cd concentration (29%). In shoots, coarse laterite led to reduction in K, Fe, Al, and Cr and increase in Na and Sr concentrations. Changes in element allocation could be, in part, a result of differences in the water availability of substrates. Matric potential in coarse laterite was significantly lower in at least 47% of the days analyzed throughout the year. Changes in element phytoextraction or phytostabilization potential could influence the efficiency of rehabilitation projects in areas degraded by mining activities.  相似文献

The trade in live animals between India and Brazil dates from the late nineteenth century when European travellers traded animals of Indian origin for display in zoos. Considering the origin of coffee and sugar cane, as well as the expertise related to mineral evaluation, we need to consider that India was involved in important economic cycles of Brazil, even indirectly. This virtuous flow of trade has been maintained and intensified throughout modern history, especially after these two nations gained political independence from their colonisers, thereby becoming independent in mercantile affairs. This paper addresses the main points related to the use of animals of Indian origin in Brazil. We revisit some of the historical aspects of the process of colonisation of Brazil, as well as the importation of animals from India. The restrictions imposed on this process due to the occurrence of diseases in cattle and buffalo in India will be examined. At the end of the text, emphasis will be given to the risks of introducing exotic diseases into Brazil.  相似文献

This study evaluated three different methods for the formation of an inclusion complex between alpha- and beta-cyclodextrin (α- and β-CD) and limonene (LIM) with the goal of improving the physicochemical properties of limonene. The study samples were prepared through physical mixing (PM), paste complexation (PC), and slurry complexation (SC) methods in the molar ratio of 1:1 (cyclodextrin:limonene). The complexes prepared were evaluated with thermogravimetry/derivate thermogravimetry, infrared spectroscopy, X-ray diffraction, complexation efficiency through gas chromatography/mass spectrometry analyses, molecular modeling, and nuclear magnetic resonance. The results showed that the physical mixing procedure did not produce complexation, but the paste and slurry methods produced inclusion complexes, which demonstrated interactions outside of the cavity of the CDs. However, the paste obtained with β-cyclodextrin did not demonstrate complexation in the gas chromatographic technique because, after extraction, most of the limonene was either surface-adsorbed by β-cyclodextrin or volatilized during the procedure. We conclude that paste complexation and slurry complexation are effective and economic methods to improve the physicochemical character of limonene and could have important applications in pharmacological activities in terms of an increase in solubility.  相似文献

Major histocompatibility complex class one (MHC-I) antigen-presenting molecules participate in central nervous system (CNS) synaptic plasticity, as does the paired immunoglobulin-like receptor B (PirB), an MHC-I ligand that can inhibit immune-cells and bind to myelin axon growth inhibitors. Based on the dual roles of both molecules in the immune and nervous systems, we evaluated their expression in the central and peripheral nervous system (PNS) following sciatic nerve injury in mice. Increased PirB and MHC-I protein and gene expression is present in the spinal cord one week after nerve transection, PirB being mostly expressed in the neuropile region. In the crushed nerve, MHC-I protein levels increased 2 weeks after lesion (wal) and progressively decreased over the next eight weeks. The same kinetics were observed for infiltrating cytotoxic T lymphocytes (CTLs) but not for PirB expression, which continuously increased. Both MHC-I and PirB were found in macrophages and Schwann cells but rarely in axons. Interestingly, at 8 wal, PirB was mainly restricted to the myelin sheath. Our findings reinforce the participation of MHC-I and PirB in CNS plasticity events. In contrast, opposing expression levels of these molecules were found in the PNS, so that MHC-I and PirB seem to be mostly implicated in antigen presentation to CTLs and axon myelination, respectively.  相似文献
