The alcohol dehydrogenase (Adh) region from five planitibia subgroup
species of Hawaiian picture-wing Drosophila has been cloned. A total of 15
kb of DNA in and around the Adh gene has been compared among the five
species. Genetic distances were calculated to determine evolutionary
relationships. These distances agree with previous distances determined by
protein polymorphism and DNA hybridization techniques and can be
interpreted in terms of specific island colonization and speciation
(founder) events over the past 5 Myr. Examination of the restriction maps
of the cloned Adh region from the five species shows many instances of
small deletions, insertion of a transposable element in D. heteroneura, and
the existence of a highly variable region on the 3' side of the Adh gene.
Clustering relationships and rates of DNA change are calculated and
compared with the relationship found for other species of Drosophila.

Anthracobunidae is an Eocene family of large mammals from south Asia that is commonly considered to be part of the radiation that gave rise to elephants (proboscideans) and sea cows (sirenians). We describe a new collection of anthracobunid fossils from Middle Eocene rocks of Indo-Pakistan that more than doubles the number of known anthracobunid fossils and challenges their putative relationships, instead implying that they are stem perissodactyls. Cranial, dental, and postcranial elements allow a revision of species and the recognition of a new anthracobunid genus. Analyses of stable isotopes and long bone geometry together suggest that most anthracobunids fed on land, but spent a considerable amount of time near water. This new evidence expands our understanding of stem perissodactyl diversity and sheds new light on perissodactyl origins. 相似文献

Anti-neutrophil cytoplasmic antibody (ANCA) - associated vasculitis (AAV) is a life-threatening autoimmune disease characterized
by an antibody-mediated glomerulonephritis and necrotizing vasculitis. Apart from antibodies, T cells are also involved in
disease pathogenesis. This review stresses the hallmarks of T cell-mediated pathology in AAV and highlights the characteristics
of lesional and circulating T cells in the immune response in AAV. Circulating effector T-cell populations are expanded and
are in a persistent state of activation. Circulating regulatory T-cell subsets are less well characterized but seem to be
impaired in function. Lesional effector T cells are present in granulomas, vasculitic lesions, and nephritis. Lesional T cells
usually show pro-inflammatory properties and promote granuloma formation. Apart from T cells, dendritic cells are abundantly
present at the sites of inflammation and locally orchestrate the immune response. Targeting the above-mentioned T cell-mediated
disease mechanisms will potentially provide powerful therapeutic tools for AAV. 相似文献

Natural killer T (NKT) cells have been implicated in the regulatory immune mechanisms that control autoimmunity. However,
their precise role in the pathogenesis of rheumatoid arthritis (RA) remains unclear. The frequency, cytokine profile and heterogeneity
of NKT cells were studied in peripheral blood mononuclear cells (PBMCs) from 23 RA patients and 22 healthy control individuals,
including paired PBMC–synovial fluid samples from seven and paired PBMC–synovial tissue samples from four RA patients. Flow
cytometry revealed a decreased frequency of NKT cells in PBMCs from RA patients. NKT cells were present in paired synovial
fluid and synovial tissue samples. Based on the reactivity of PBMC-derived NKT cells toward α-galactosylceramide, RA patients
could be divided into responders (53.8%) and nonresponders (46.2%). However, NKT cells isolated from synovial fluid from both
responders and nonresponders expanded upon stimulation with α-galactosylceramide. Analysis of the cytokine profile of CD4+ and CD4- PBMC derived NKT cell lines from RA patients revealed a significantly reduced number of IL-4 producing cells. In contrast,
synovial fluid derived NKT cell lines exhibited a Th0-like phenotype, which was comparable to that in healthy control individuals.
This suggests that synovial fluid NKT cells are functional, even in patients with nonresponding NKT cells in their blood.
We conclude that, because the number of Vα24+Vβ11+CD3+ NKT cells is decreased and the cytokine profile of blood-derived NKT cells is biased toward a Th1-like phenotype in RA patients,
NKT cells might be functionally related to resistance or progression of RA. Providing a local boost to the regulatory potential
of NKT cells might represent a useful candidate therapy for RA. 相似文献

In 1996 the Minister of Public Health, Welfare and Sports in The Netherlands published a 'Planning Decree Special Interventions in the Heart'. She requested from the professional organizations guidelines for the indications for interventions in the heart. A working group was formed with representatives from the Dutch professional organizations for cardiology and thoracic surgery, to address this issue for patients with coronary artery disease. The working group confirmed the need to discuss all patients who are considered for either elective or emergency revascularization during a multidisciplinary consultation in (or with) one of the specialized Dutch hospitals. During this meeting of the 'heart team', at least one interventional cardiologist and one thoracic surgeon should be present. There are three possible outcomes of the heart team's consultations for each patient: drug therapy only ('conservative management'), coronary surgery or catheter intervention. For each case, the team should indicate the expected benefit, the risk of the intervention, the urgency and the estimated waiting time. The guidelines presented in this paper address these issues for three patient categories: stable angina pectoris, unstable angina pectoris and acute myocardial infarction. 相似文献

A nearly complete cranium of Ignacius graybullianus provides increased understanding of the cranial anatomy of Plesiadapiformes. In nearly all details of cranial anatomy, Ignacius differs markedly from primates. USNM 421608 exhibits a long tapering snout, small widely spaced orbits, and a complete lack of postorbital process or bar. Large olfactory bulbs are inferred from the wide interorbital space. The marked flare of the zygomatic arches suggests that Ignacius possessed large and powerful temporal muscles. The basicranial region is particularly well preserved and reveals a distinct suture between the petrosal bone and an entotympanic bulla. This suture is visible on both the left and right sides of the skull and dispels the hypothesis that Ignacius and, by inference, other Plesiadapiformes share the primate synapomorphy of a petrosal bulla. To test the phylogenetic position of Ignacius, cranial characters were identified and scored for Ignacius, Plesiadapis, Cynocephalus, and a number of primates, bats, and scandentians. Two erinaceomorph insectivores were also included to allow the assessment of archontan monophyly. These characters were incorporated into a maximum-parsimony analysis to determine the phylogenetic position of Plesiadapiformes. There are several important phylogenetic conclusions that can be inferred from this analysis: 1) Ignacius and Plesiadapis make up a monophyletic clade; 2) Plesiadapiformes may be the sister group of Dermoptera; 3) Scandentia, not Plesiadapiformes, is the sister group of Primates; and 4) Primates, plesiadapiforms, bats, colugos, and scandentians may not form a monophyletic clade Archonta. Tympanic bullae and baleen plates from bowhead whales of the Western Arctic population were examined. Growth layer groups (GLGs) in the involucrum of the tympanic bone were used to estimate age of the whales, and compared to stable isotope signatures along transects of baleen plates and the involucrum. The involucrum of the tympanic bone consists of three regions that form in utero, during nursing in the first year, and during the first decades of life, respectively. Life history events, such as annual migration, are recorded in the bowhead tympanic bulla. It is likely that bone growth in the bowhead tympanic occurs during periods of high food intake, while slow or arrested growth occurs during periods of low food intake. Comparisons between numbers of GLGs in the tympanic, number of isotopic oscillations in a baleen plate, length of the baleen plate, and total whale length show correlation coefficients as high as 0.97. The tympanic GLG method is particularly useful for estimating the age of whales up to 20 yr old. 相似文献

An essential mechanism for repairing DNA double‐strand breaks is homologous recombination (HR). One of its core catalysts is human RAD51 (hRAD51), which assembles as a helical nucleoprotein filament on single‐stranded DNA, promoting DNA‐strand exchange. Here, we study the interaction of hRAD51 with single‐stranded DNA using a single‐molecule approach. We show that ATP‐bound hRAD51 filaments can exist in two different states with different contour lengths and with a free‐energy difference of ~4 kBT per hRAD51 monomer. Upon ATP hydrolysis, the filaments convert into a disassembly‐competent ADP‐bound configuration. In agreement with the single‐molecule analysis, we demonstrate the presence of two distinct protomer interfaces in the crystal structure of a hRAD51‐ATP filament, providing a structural basis for the two conformational states of the filament. Together, our findings provide evidence that hRAD51‐ATP filaments can exist in two interconvertible conformational states, which might be functionally relevant for DNA homology recognition and strand exchange. 相似文献
