We examined the effect of ovule position within the ovary on the probability of seed maturation, on seed weight, and on progeny performance in the outbreeding legume Phaseolus coccineus. Ovaries of P. coccineus possess six linearly arranged ovules (ovule position one = stylar end). We found that in both 1987 and 1988, ovule position had a significant effect on the probability of seed maturation under field conditions. In 1987, ovule positions one. two, and three had a higher probability of maturing seeds than the three most basal ovule positions. In 1988, the probability of producing a mature seed in ovule position one was more similar to the three most basal ovule positions than to ovule positions two and three. The position of the ovule in the ovary had no significant effect on seed weight in 1987, but it had a significant effect in 1988. Overall, seeds from ovule positions one, two, and three tended to produce heavier seeds than the three most basal ovule positions. The effects of ovule position on progeny performance were determined in a greenhouse and a field study. In the greenhouse study, we found no significant overall effect of the position of the ovule that produced the seed on progeny performance. In the field study, we did find a significant ovule position effect on several measures of reproductive performance as well as an overall effect on reproductive performance. In addition, we found a significant interaction between ovule position and number of seeds per fruit. Progeny from the stylar end of the fruit outperformed the progeny from the peduncular end in fruits containing many seeds, whereas there were no significant differences between progeny produced in the stylar and peduncular ends of fruits containing few seeds. Causes of position effects are unknown but hypotheses abound.  相似文献

A high-performance liquid chromatographic procedure was developed for the determination of caffeine in various biologic fluids and coffee. A reversed-phase column and UV detection at 254 nm were used to obtain a sensitivity of 0.1 μg/ml caffeine in serum and saliva using a sample volume of 0.1 ml. Caffeine metabolites and commonly ingested xanthines do not interfere with the assay. The within-day coefficients of variation were 9.8 and 9.9% at plasma caffeine concentrations of 2 and 10 μg/ml, respectively. The day-to-day coefficients of variation were 6.8 and 6.6% at plasma caffeine concentrations of 2 and 10 μg/ml, respectively. Serum and saliva caffeine concentrations were determined following a single oral dose of coffee and an intravenous infusion of caffeine in one subject. Computer estimates of caffeine pharmacokinetic parameters in one subject are in excellent agreement with previously published values.  相似文献

Two isolectins were isolated from the albumin fraction ofArtocarpus integrifolia L. seeds, by precipitation with ammonium sulfate, and DEAE-cellulose and SP-Sephadex C-50 chromatography. The isolectins, when passed through Sephadex G-100 at pH 7.4, had a molecular mass of 43 000 and when subjected to SDS electrophoresis in the presence of β-mercaptoethanol consisted of two subunits with molecular mass of 11 250 and 15 000. When they were tested with human erythrocytes of all the groups of the ABO system there was no blood specificity. The migration of 51Cr-labeled nylon-wool separated mouse thoracic duct T cells has been followed in order to determine whether there is a circulation of small (nondividing) T cells through the small intestine. Approximately 6% of the injected dose of T-TDL localized in the small intestine (minus Peyer's patches). Experiments revealed that this gut-localizing cell population consisted almost entirely, if not exclusively, of lymphoblasts present in mouse T-TDL. When lymphoblasts and small lymphocytes from mouse T-TDL were separated by velocity sedimentation, and the migration of separated fractions was studied, we found large cells (66% blasts) migrated well to the gut but poorly to the lymph nodes, whereas small cells (2% blasts) showed minimal migration to the gut but localized randomly in lymph nodes and spleen. The in vivo distribution of small cells from T-TDL was similar to that of T-PLN. Furthermore, the recirculatory patterns of both 51Cr-labeled T-TDL and T-PLN were found to be identical as accessed by their rate of recovery in the thoracic duct lymph of recipient mice. These results support the notion that the vast majority of T-TDL and T-PLN are part of a common pool of recirculating T cells which recirculate randomly through lymph nodes and spleen and not the small intestine.  相似文献

In vitro propagation of neem (Azadirachta indica A. Juss.) may offer an efficient alternative to seed propagation of this species. For optimization of in vitro propagation, different basal salt formulations, growth regulators, and culture container sealants (polytetrafluoroethylene hydrophobic membranes [PTFE]) were evaluated. Nodal segments cultured on Murashige and Skoog (MS) medium showed the highest shoot formation per explant (1.67). Explants cultured in flasks containing MS medium with 0.5 mg L−1 benzyladenine, 0.5 mg L−1 kinetin, and 0.05 mg L−1 naphthaleneacetic acid, and sealed with two PTFE membranes, produced the highest number of shoots (4.04). In contrast, explants cultured in flasks without membranes showed leaf chlorosis and senescence. For plant recovery, regenerants were acclimatized in a substrate of coconut fiber and eucalyptus bark (1:1) and showed 80% survival. Our results indicated that the use of flasks with vents was beneficial for in vitro propagation of this important plant.  相似文献

The potential impact of the soil seed bank of a given species and subsequent population persistence will depend on the robustness of the seeds against depletion. Invasiveness is frequently linked to this behavior and successful control of invasive species depends on reducing the size of their seed banks. We propose a hybrid model that simulates the seed bank dynamics in association with their physical dormancy (PD) based on experimental data on seed bank depletion of a Leucaena leucocephala population during the dispersal phase. The hybrid model uses cellular automata (CA) and Monte Carlo techniques. The sensitivity analysis revealed that the model delivers reproducible and reliable results. Then, we discuss the ability of the hybrid model to predict the effect of seasonal climate change. A 2.0 °C increase in temperature was enough to cause a considerable impact in the seed depletion curve, and after 1 year, ~?75% of the seeds had already disappeared from the seed bank. The proposed hybrid CA model can be used to study soil-stored seed banks with physical dormancy, and could be applied to other Fabaceae species with similar seed traits. We believe that this computational model contributes to a better understanding of seed bank dynamics and will aid in the control of invasive species and for testing different scenarios to reduce or eliminate seed banks that have already been formed.
