Rosette-forming glioneuronal tumor (RGNT) of the IV ventricle is a rare and recently recognized brain tumor entity. It is histologically composed by two distinct features: a glial component, resembling pilocytic astrocytoma, and a component forming neurocytic rosettes and/or perivascular rosettes. Herein, we describe a 33-year-old man with RGNT arising in the spinal cord. Following an immunohistochemistry validation, we further performed an extensive genomic analysis, using array-CGH (aCGH), whole exome and cancer-related hotspot sequencing, in order to better understand its underlying biology. We observed the loss of 1p and gain of 1q, as well as gain of the whole chromosomes 7, 9 and 16. Local amplifications in 9q34.2 and 19p13.3 (encompassing the gene SBNO2) were identified. Moreover, we observed focal gains/losses in several chromosomes. Additionally, on chromosome 7, we identified the presence of the KIAA1549:BRAF gene fusion, which was further validated by RT-PCR and FISH. Across all mutational analyses, we detected and validated the somatic mutations of the genes MLL2, CNNM3, PCDHGC4 and SCN1A. Our comprehensive molecular profiling of this RGNT suggests that MAPK pathway and methylome changes, driven by KIAA1549:BRAF fusion and MLL2 mutation, respectively, could be associated with the development of this rare tumor entity.  相似文献

Spot blotch caused by Bipolaris sorokiniana is a destructive disease of wheat in warm and humid wheat-growing regions of the world. This fungus shows a high genetic diversity and morphological and physiologic variability. In this study, 19 polysporic and 57 monosporic isolates of B. sorokiniana were characterized using universal rice primers—URP-PCR. The results obtained when the dendrogram was constructed with all the data produced with the amplification products showed very distinct clusters. However, the similarity among the isolates was low where 37 and 26.3 % of the monosporic and polysporic isolates, respectively, showed similarity above 70 %. All primers amplified multiple DNA fragments of polysporic as well as the monosporic isolates. Isolates fingerprints were constructed based on binary characters revealed by the three primers. An amplified fragment of approximately 750 bp was observed among 40 % of the isolates, when primer URP-1F was used. When primers URP-4R and URP-2R were used, a fragment of 450 and 400 bp was present in 31.5 and 29 % of the isolates, respectively. It was expected a higher similarity among the isolates since the monosporic cultures were originated from the polysporic. The dendrogram did not enable the separation of B. sorokiniana isolates by their geographic origin. This low correlation suggests that gene transfer may have occurred by parasexual combination in this fungus population. However, in spite of the research efforts for that end, it has not been possible to establish patterns that characterize the profile of B. sorokiniana.  相似文献

The present study analyzed, the influence of the treatment with juvenile hormone on the ultrastructure of Apis mellifera L. workers' venom glands. Newly emerged workers received topical application of 1 microl of juvenile hormone diluted in hexane, in the concentration of 2 microg/pl. Two controls were used; one control received no treatment (group C1) and other received topical application of 1 microl of hexane (group C2). The aspect of the glandular cells, in not treated newly emerged workers, showed that they are not yet secreting actively. Cellular modifications happened according to the worker age and to the glandular area considered. The most active phase of the gland happened from the emergence to the 14th day. At the 25th day the cells had already lost their secretory characteristic, being the distal area the first to suffer degeneration. The treatment with juvenile hormone and hexane altered the temporal sequence of the glandular cycle, forwarding the secretory cycle and degeneration of the venom gland.  相似文献

This study evaluated the antioxidant activities in the leaves and stem bark fractions of Scutia buxifolia. Cerebral lipid peroxidation (TBARS) was induced by Fe(II) and radical-scavenging activity was determined by DPPH method. Folin–Ciocalteu was used to determine phenolic contents. Quercetin, quercitrin, isoquercitrin and rutin were isolated from leaf ethyl acetate fraction and their levels were measured by high performance liquid chromatography.-photodiode array detector.IC50 (DPPH) varied from 4.35 ± 1.30 to 29.55 ± 0.54 μg/mL for stem bark and from 6.50 ± 0.40 to 30.54 ± 1.14 in the leaves. Ethyl acetate and butanolic fractions caused a sharp fall in TBARS production with IC50 from 2.93 ± 2.17 to 40.46 ± 2.51 μg/mL for the leaves and 0.66 ± 0.17 to 27.3 ± 1.23 for the stem bark. Results obtained indicated that S. buxifolia has a great potential to prevent disease caused by the overproduction of free radicals and also it might be used as a potential source of natural antioxidant agents.  相似文献

In 1959, G.E. Hutchinson provided a general explanation for the diversity of species in his paper “Homage to Santa Rosalia or why are there so many kinds of animals?” To assess the contribution of the ideas Hutchinson introduced in “The Homage” to aquatic ecology research, we performed a bibliometric evaluation of all the articles that cited this paper between 1960 and 2009. The articles were retrieved using the database from Thomson Reuters (ISI Web of Knowledge) in March 2009. For each paper, we first identified the studied environment (terrestrial, marine or freshwater) and whether the study was theoretical or empirical. For marine and freshwater studies, we recorded the journal where the article was published, the year of publication, the number of citations, the taxonomic group (e.g. fish, phytoplankton, zooplankton, macroinvertebrates or macrophytes), the habitat (e.g. wetlands, lakes, rivers, streams or ocean) and the main ideas addressed. A total of 1345 articles cited “The Homage”, and the number of citations increased significantly with time. Most of the articles that cited “The Homage” described research carried out in a terrestrial environment or were theoretical, and a lesser number of citations came from freshwater and marine papers, which used mainly fish and invertebrates as model organisms. In the aquatic sciences, most of the papers discussed the influences of competition and energy (productivity) on diversity. Our results indicate that “The Homage” can still be considered a “citation classic” and a breakthrough contribution, and that it is still having a great impact on different fields of ecology, including limnology and marine ecology.  相似文献

The aim of this study was to determine the influence of two ethylene glycol-based vitrification solutions on in vitro and in vivo survival after in-straw cryoprotectant dilution of vitrified in vitro-produced bovine embryos. Day-7 expanded blastocysts were selected according to diameter (> or = 180 microm) and osmotic characteristics and randomly assigned to one of three groups (i) VSa: vitrification in 40% EG+17.1% SUC+0.1% PVA; (ii) VSb: vitrification in 20% EG+20% DMSO; (iii) control: non-vitrified embryos. Vitrification was performed in hand-pulled glass micropipettes (GMP) and cryoprotectant dilution in 0.25 ml straws after warming in a plastic tube. Embryo viability was assessed by re-expansion and hatching rates after 72 h of IVC and by pregnancy rates after direct transfer of vitrified embryos. No differences in re-expansion rates were observed between vitrified groups after 24 h in culture (VSa=84.5%; VSb=94.8%). However, fewer VSa embryos (55.2%, P<0.05) hatched after 72 h than the VSb (75.8%) and control embryos (80.0%). To evaluate in vivo viability, vitrified embryos (VSa=20; VSb=21) were warmed under field conditions and individually transferred to synchronous recipients. Pregnancy rates (day 60) were similar between groups (VSa=20%; VSb=19%). Greater hatching rates occurred after 72 h of IVC for EG+DMSO than EG+SUC+PVA vitrification solutions. However, using a GMP vitrification container and in-tube warming, both solutions provided similar pregnancy rates after the in-straw cryoprotectant dilution and direct embryo transfer.  相似文献

Colonies of the honey bee, Apis mellifera, consist of males and two female castes: workers and queens. The castes and males from A. mellifera have a distinct morphology, physiology and behaviour that correlate with their roles in the society and are characterized by some brain polymorphisms. Compound eyes are one of the characteristics that differ among the castes and sexes. A. mellifera is a holometabolous insect; therefore, the development of adult organs during metamorphosis, which will produce these differences, requires the precise coordination of three main programmed cellular processes: proliferation, differentiation and death. These processes take place simultaneously during pupation. Our purpose was to investigate cell division and death in the optic lobes (OL) of workers, queens and males during pupation to identify how the differences in the compound eyes in adults of these classes are achieved. The results showed that OL differentiation follows a similar pattern in the three classes of individuals studied, without structural differences in their development. The main non-structural differences involve cell division, mortality rates and timing. The results suggest a modelling of the brain during differentiation, which contributes to the specific functions of each individual class.  相似文献

Size-related and seasonal evaluation of the dietary composition of fat snook ( Centropomus parallelus Poey 1860) in the upper sector of an estuary of the southeastern coast of Brazil were carried out based on stomach analyses of specimens ranging from 40 to 170 mm standard length. Results reveal that C. parallelus is a carnivorous species feeding mainly on benthic crustaceans. Relatively high stomach replenishment suggests that this environment is an important feeding ground for fat snook juveniles. Multivariate analyses indicated that predator size effect is significantly more important than seasonal variation in determining dietary composition. Predator length was associated with increased consumption of palaemonid shrimps ( Macrobrachium spp.) and grapsid crabs, and decreased foraging on tanaids ( Kalliapseudes schubarti ), thus showing a preference shift from smaller to larger prey. Predator length was also positively associated with an increase in the stomach repletion index. Additionally, allometric growth of both gape and head were consistently correlated with this ontogenetic dietary transition, suggesting that such changes might be related to an individual's ability to capture and consume larger, more elusive prey. The digestive tube is short and grows isometrically, which is in accordance with the carnivorous habit of this estuarine fish and its maintenance through ontogeny.  相似文献
