Polyunsaturated fatty acids (PUFAs) occur in relatively high amounts in phospholipids of the synapses. PUFAs may thus determine the fluidity of the synaptosomal membrane and, hereby, they may regulate the neuronal transmission. It was therefore tempting to suggest a system in the brain, that inhibits autooxidation of PUFAs. In order to trace such a protection system, Wistar rats were equally loaded with 4500 kBq of 75-Se either as selenite or as L-Se-methionine. By means of gradient ultracentrifugation, particulate fractions of the brains were isolated, and the radioactivity as well as the glutathione-transferase and -peroxidase activities were estimated. The distribution of the two selenium components among the particulate fractions was different. Thus, selenite gave higher radioactivity in myelin, then followed by the light synaptosomal and the vesicular fraction. L-Se-methionine was more equally incorporated in all particulate fractions, although highest activity was found in the mitochondrial fraction. Myelin and synaptic vesicles were devoid of transferase activity. On the other hand, the synaptosomal fraction showed highest specific transferase activity. The glutathione peroxidase activity was highest in the myelin fraction, followed by the vesicular and the synaptosomal fractions. The data obtained thus support the idea that the PUFAs of the synaptic compartment are protected against peroxidation, at least in part, by the selenium containing glutathione peroxidase.  相似文献

Utilizing a two-beam technique in the frequency domain, the pumped absorption of PS II membrane fragments from spinach and of acetonic chlorophyll-a solutions was measured at room temperature. In a very narrow wavelength region (0.2 nm around the pump pulse wavelength) the relative test beam transmission exhibited either a decrease or an increase, respectively, dependent on the intensity of a strong pump beam. In contrast, the transmission changes of chl-a solutions were not affected by the wavelength mistuning between pump and test beam. The data obtained for PS II membrane fragments were interpreted in terms of excited state absorption of pigment-protein clusters within the light-harvesting complex of PS II. The interpretation of the small absorption band as a homogeneously broadened line led to a transversal relaxation time for chlorophyll in vivo of about 1 ps.Abbreviations PS I photosystem I of green plants - PS II photosystem II of green plants - P700 primary donor of PS I - P680 primary donor of PS II  相似文献

Summary The structural determination of biological molecules in solution by NMR relies on the determination of a set of interatomic distances obtained by measurement of intramolecular nuclear Overhauser effects (NOE). It is shown in this paper that it is possible to obtain the accurate relaxation rate (and hence the interatomic distance) from the direct measurement of a single NOE signal. The precise analysis of a NOESY peak evolution with respect to the mixing time allows the evaluation of the relaxation parameters for the pair of spins under consideration. This is done without any assumption on the relaxation of unmeasured spins, or on the movement of the molecule. The theoretical basis of this method is presented. In order to evaluate the proposed method, a simulated case on the protein BPTI is studied, which shows that the method performs very well even in the case of noisy data sets.  相似文献

Aplanospores ofHaematococcus pluvialis MUR 145 contained 0.7% carotenoids (dry wt. basis) consisting of β,β-carotene (5% of total carotenoid), echinenone (4%), canthaxanthin (4%), (3S,3′S)-astaxanthin diester (34%), (3S,3′S)-astaxanthin monoester (46%), (3S,3′S)-astaxanthin (1%) and (3R,3′R,6′R)-lutein (6%). The astaxanthin esters were examined by TLC and HPLC and VIS,1H NMR and mass spectra recorded. Their chirality was determined by the camphanate method (Vecchi & Müller, 1979) after anaerobic hydrolysis. The tough cell wall of the aplanospores required enzymatic treatment prior to pigment extraction. The potential use of this microalga as a feed ingredient in aquaculture is discussed briefly.  相似文献

Protonephridia and Metanephridia - their relation within the Bilateria   总被引:3,自引:0,他引:3  
Two different kinds of nephridia occur within the Bilateria, protonephridia closed up by a terminal cell and metanephridia opening into the coelomic cavity. Both initially filter and subsequently modify intercellular fluids. Whereas metanephridia are strictly correlated to a coelom, proto-nephria occur in acoelomate as well as in coelomate organisms. Protonephridia of different bilaterian taxa correspond to each other in several structural features. Therefore, it is hypothesized that protonephridia are homologous organs throughout the Bilateria. They must have evolved once as one pair of monociliated organs orinatinng from the ectoderm and consistin of one terminal, one duct and one nephropore cell In the ground pattern of the Bilateria the cilium of the terminal cell has only one rootlet and is surrounded by resumably eight strengthened and elongated microvilli. Cilium and microvilli extend into the hollow cyinder of the terminal cell, which is oriented distally and is attached to the adjacent duct cell by desmosomes. This cylinder is perforated by clefts and represents the supporting structure of the filtration barrier consisting of extracellular matrix. In the Annelida and Phoronida, the metanehridia at the postlarval stages are ontogenetically preceded by protonephridia in the larva, but far reaching structural and developmental differ ences exist between the metanephridia of both. In horonids the rotonephrdial duct of the larva is retained in the postlarva and acquires a coelothelially derived funnel, whereas in annelids the metanephridia are uniform organs orihating from a solid anlage, which is a repetition of the protonehridial anlage of the larva. The differences contradict a homology of the metanephridia in Annegda and Phoronida. We therefore have to conclude that metanephridia must have evolved indeendently, at least two times. The comparative analysis of nephridia in the Bilateria allows the following hyothesis: Pro tonephridia were evolved in a monohasic acoelomate organism in the stem fineage of the Bilateria. During the evolution of biphasic life cycles consisting of an acoelomate larva and a coelomate adult, the information about the differentiation of protonephridia has been preserved in the early acoelomate developmental (larval) stages. During postlarval development and the formation of a coelom the protonephridia have either been retained or modified into meta nephridia. Accordin to the differences between the metanehridia of phoronids and annelids, we emphasize that. tiere is no possibility to trace back all bilaterian taxa with a coelom to a common stem species.  相似文献

1.  Physiological adaptation to hypothermia were studied in newly hatched great snipe chicks (Gallinago media) by measuring oxygen uptake (VO2), heart rate (HR), respiratory frequency (RF), and body temperature (Tb) at different ambient temperatures (Ta).
2.  Tb of 1-day-old chicks at Ta of 35°C stabilized at about 40°C. At Ta between 20 and 30°C the chicks maintained a Tb about 8°C above Ta. Hatchlings maintained a higher gradient when active than when resting. Below 20°C they were unable to maintain a stable Tb.
3.  In resting hatchlings VO2 was similar at Ta between 35 and 20°C (Tb 40–30°C), VO2 range 1.7–2.5 ml·g-1·h-1. Below 20°C, VO2 declined with time.
4.  The HR of 1-day-old chicks fell linearly with Tb during cooling. The Q10 of the HR was 1.7 at Tb 38°C and increased to 3.0 at 29°C. The RF showed a slight tendency to decrease with decreasing Tb.
5.  It is concluded that the ability to maintain normal dexterity at low Tb is an important aspect of snipe survival strategy. Maintaining a temperature gradient rather than a constant high Tb presumably saves energy. It is suggested that the mechanisms whereby VO2 is maintained at a low Tb may involve isoenzymes and adaptations of the nervous system. However, such adaptations would not seem to affect the pacemaker mechanism as evidenced by the high Q10 of the HR.

Summary A series of experiments has established the molecular defect in the medium-chain acyl-coenzyme A (CoA) dehydrogenase (MCAD) gene in a family with MCAD deficiency. Demonstration of intra-mitochondrial mature MCAD indistinguishable in size (42.5-kDa) from control MCAD, and of mRNA with the correct size of 2.4 kb, indicated a point-mutation in the coding region of the MCAD gene to be disease-causing. Consequently, cloning and DNA sequencing of polymerase chain reaction (PCR) amplified complementary DNA (cDNA) from messenger RNA of fibroblasts from the patient and family members were performed. All clones sequenced from the patient exhibited a single base substitution from adenine (A) to guanine (G) at position 985 in the MCAD cDNA as the only consistent base-variation compared with control cDNA. In contrast, the parents contained cDNA with the normal and the mutated sequence, revealing their obligate carrier status. Allelic homozygosity in the patient and heterozygosity for the mutation in the parents were established by a modified PCR reaction, introducing a cleavage site for the restriction endonuclease NcoI into amplified genomic DNA containing G985. The same assay consistently revealed A985 in genomic DNA from 26 control individuals. The A to G mutation was introduced into an E. coli expression vector producing mutant MCAD, which was demonstrated to be inactive, probably because of the inability to form active tetrameric MCAD. All the experiments are consistent with the contention that the G985 mutation, resulting in a lysine to glutamate shift at position 329 in the MCAD polypeptide chain, is the genetic cause of MCAD deficiency in this family. We found the same mutation in homozygous form in 11 out of 12 other patients with verified MCAD deficiency.  相似文献

Population densities and total phosphorus concentrations in samples from different lakes of south-eastern Norway were determined. In addition some transplant experiments with dilute phytoplankton populations were carried out. A laboratory batch culture method was used.The diatoms studied may be divided into three ecological groups based on their cell densities and total phosphorus concentrations in the samples. This classification was supported by the experimental results. Cyclotella spp., Asterionella formosa and Tabellaria fenestrata did not grow or had low growth rates above pH 9. Synedra cf. acus and Fragilaria crontonensis had often high growth rates within the pH 9–10 range, but were not able to grow at pH values above 10. High pH-values had no effect on the growth rate of Oscillatoria. Oscillatoria, Synedra and Stephanodiscus were severely growth-limited in filtered water from oligotrophic lakes. Maximum growth rates of all the populations studied were often obtained after addition of phosphate and chelated iron (FeEDTA) in combination to filtered water samples from oligotrophic/mesotrophic lakes.  相似文献
