Research on the impact of Maillard reaction products (MRPs) on microorganisms has been reported in the literature for the last 60 years. In the current study, the impact of an MRP-rich medium on the growth of three strains of Escherichia coli was measured by comparing two classic methods for studying the growth of bacteria (plate counting and optical density at 600 nm) and by tracing MRP utilisation. Early stage and advanced MRPs in the culture media were assessed by quantifying furosine and N ε -carboxymethyllysine (CML) levels, respectively, using chromatographic methods. These measures were performed prior to and during bacterial growth to estimate the potential use of these MRPs by Escherichia coli CIP 54.8. Glucose and lysine, the two MRP precursors used in the MRP-rich medium, were also quantified by chromatographic means. Compared to control media, increased lag phases and decreased growth rates were observed in the MRP-rich medium for two out of the three Escherichia coli strains tested. In contrast, one strain isolated from the faeces of a piglet fed on a MRP-rich diet was not influenced by the presence of MRPs in the medium. Overall, CML as well as the products obtained by the thermal degradation of glucose and lysine, regardless of the Maillard reaction, did not affect the growth of the three strains tested. In addition, no degradation of fructoselysine or CML was found in the presence of Escherichia coli CIP 54.8.  相似文献

The discovery of monoclonal antibodies (mAbs) that bind to a particular molecular target is now regarded a routine exercise. However, the successful development of mAbs that (1) express well, (2) elicit a desirable biological effect upon binding, and (3) remain soluble and display low viscosity at high concentrations is often far more challenging. Therefore, high throughput screening assays that assess self-association and aggregation early in the selection process are likely to yield mAbs with superior biophysical properties. Here, we report an improved version of affinity-capture self-interaction nanoparticle spectroscopy (AC-SINS) that is capable of screening large panels of antibodies for their propensity to self-associate. AC-SINS is based on concentrating mAbs from dilute solutions around gold nanoparticles pre-coated with polyclonal capture (e.g., anti-Fc) antibodies. Interactions between immobilized mAbs lead to reduced inter-particle distances and increased plasmon wavelengths (wavelengths of maximum absorbance), which can be readily measured by optical means. This method is attractive because it is compatible with dilute and unpurified mAb solutions that are typical during early antibody discovery. In addition, we have improved multiple aspects of this assay for increased throughput and reproducibility. A data set comprising over 400 mAbs suggests that our modified assay yields self-interaction measurements that are well-correlated with other lower throughput assays such as cross-interaction chromatography. We expect that the simplicity and throughput of our improved AC-SINS method will lead to improved selection of mAbs with excellent biophysical properties during early antibody discovery.  相似文献

Four tributaries of Lake St-Jean (Québec, Canada) are used for spawning and juvenile habitat by land-locked Atlantic salmon. Spawning runs have drastically declined since the mid-1980s, and consequently, a supportive-breeding programme was undertaken in 1990. In this study, we analysed seven microsatellite loci and mtDNA, and empirically estimated effective population sizes to test the hypotheses that (i) fish spawning in different tributaries form genetically distinct populations and (ii) the supportive breeding programme causes genetic perturbations on wild populations. Allele frequency distribution, molecular variance and genetic distance estimates all supported the hypothesis of genetic differentiation among salmon from different tributaries. Gene flow among some populations was much more restricted than previously reported for anadromous populations despite the small geographical scale (40 km) involved. Both mtDNA and microsatellites revealed a more pronounced differentiation between populations from two tributaries of a single river compared with their differentiation with a population from a neighbouring river. The comparison of wild and F1-hatchery fish (produced from breeders originating from the same river) indicated significant changes in allele frequencies and losses of low-frequency alleles but no reduction in heterozygosity. Estimates of variance and inbreeding population size indicated that susceptibility to genetic drift and inbreeding in one population increased by twofold after only one generation of supplementation.  相似文献

Rat uterine stromal cells (UIII) express pancreatic type PLA2 (PLA2-I) receptor and internalize the enzyme bound to receptors. Here, we investigate the proliferating effect and alterations in binding of PLA2-I. There is a dramatic decline in PLA2-I binding in UIII cells as they progress from a nonconfluent proliferating state (40,000 sites/cell) to a confluent state (1300 sites/cell). Intracellular concentration of PLA2-I changed with the alteration in binding, suggesting that regulation in the PLA2 binding capacity may have important implications in growth control mechanisms.  相似文献

An important environmental factor determining both phytoplankton and zooplankton community composition is lake depth and thermal stratification. However, there is little information on how the interaction between zooplankton grazers and their phytoplankton food changes along an environmental gradient of lake depth. We contrasted resource availability for daphniid zooplankton populations living in two shallow, unstratified lakes and in two deep, stratified lakes using a novel growth bioassay. Stratified lakes had consistently lower resource richness than shallow unstratified lakes. To test whether resources were important in explaining differences in daphniid composition of shallow and deep lakes, we performed reciprocal transplant experiments. We raised daphniids typical of shallow (Ceriodaphnia reticulata) and deep (Daphnia dentifera) lakes in the resources from replicate shallow and deep lakes and monitored survival and reproduction. The two species exhibited a performance trade-off, measured by life table r and R 0, across a gradient in natural resource richness. D. dentifera had higher relative fitness than C. reticulata when raised in the poorest resource environment from a deep lake. However, under richer resource conditions typical of shallow lakes, C. reticulata outperformed D. dentifera. We further created a gradient in natural resource quantity (by dilution) to test whether this trade-off in species relative fitness involved aspects of resource quality. No trade-off in species performance was evident across the dilution gradient, indicating that resource quality was important to the trade-off. We conclude that shifts in daphniid species composition along a gradient of lake depth involve an adaptive trade-off in ability to exploit rich versus poor resource quality. 1. Thin-tipped micro-electrodes were used to measure oxygen concentrations in the burrows of two common aquatic insects, the mayfly Hexagenia limbata and the alderfly Sialis velata . Both species maintain their surroundings oxygenated by drawing water from above the sediment surface into their tubes. 2. The temporal pattern of oxygen in the burrows differed between the species. The constant high oxygen concentration (>75% of air saturation) measured in the tubes of the mayfly suggest that this animal pumps water almost continuously, which is consistent with its high oxygen requirements. In contrast, oxygen concentration in burrows of the alderfly fluctuated widely over time, suggesting that this animal irrigates only irregularly, probably because it can tolerate short periods of low oxygen concentration in its burrow. 3. The interval between pumping episodes by the alderfly decreased with increasing temperature, a result of increased oxygen consumption by the animal and by sediment at high temperature. 4. Based on the tube dimensions, oxygen penetration depth and animal density in lakes, we estimate that Hexagenia could create an oxic micro-environment equivalent to 3–35% of the volume of the surface oxidized sediment layer created by molecular diffusion. The mosaic of oxic micro-environments created by the burrowing and irrigation of freshwater animals could influence chemical and biological processes in sediments, the fluxes of materials between the sediment and the overlying water column, and the exposure of benthic animals to sedimentary contaminants.  相似文献

Abstract. The effects of disturbance and microtopography on the organization and dynamics of plant communities were studied in a European salt marsh located in the Bay of Mont St. Michel, France. The existence of seed trapping mechanisms was also tested. The study took place in the lower and middle marsh plant communities dominated by the perennials Puccinellia maritima and Halimione portulacoides, respectively and associated with the annual Suaeda maritima. Three treatments were used in series of plots placed in each community: (1) vegetation removal and root destruction to a depth of 10 cm and refilling, (2) non‐remnant herbicide treatment without vegetation removal and (3) creation of depressions (20 cm deep). These treatments were compared with adjacent control plots. The first year of the experiment showed that the perennials facilitated the establishment of Suaeda by trapping its seeds. Estimation of cover, density and biomass over 5 yr following the disturbances showed that in the first 2 yr Suaeda dominated the disturbed plots. Thereafter Suaeda was gradually eliminated by competitive exclusion after ca. 3 yr in the zone originally dominated by Puccinellia maritima and after 4 yr in the zone occupied by Halimione portulacoides. Depressions constituted refuge habitats for Suaeda by limiting competition with the perennials but also led to a high risk of mortality with temporal fluctuations in density. Despite a period of investigation limited to 5 yr, our study demonstrated that natural disturbances of various types occurred and influenced the dynamics of Suaeda, Halimione and Puccinellia. We deduced that natural disturbances and microtopography are responsible for the maintenance of the habitat in a state of non‐equilibrium by favouring the establishment of both spatial and temporal environmental heterogeneity. These conditions appear to be particularly favourable for the maintenance of annual species such as Suaeda maritima.  相似文献

Summary A study was conducted in 29 households with asthmatic patients in order to analyse the indoor mould concentration; an important contamination was found in both the air and the dust. Although it is known that the outdoor mould concentration in the air decreases during winter, we found that the indoor one did not vary throughout the year. Moreover, the indoor mould concentration increased during the winter in recently built houses, probably because of the poor ventilation. The immunological study showed a lack of correlation between the moulds collected and the patients sensitization.  相似文献
