We present structure, photoexcited triplet-state property, and singlet oxygen ((1)O 2) generation capability of human serum albumin complexed with a tris(dicarboxymethylene)[60]fullerene C3-isomer (HSA-CF). Small angle X-ray scattering measurements showed that a globular size-dimension and surface charge distribution of HSA were unaltered by monomolecular complexation of CF into the hydrophobic cavity. Laser flash photolysis to the HSA-CF solution yielded a photoexcited triplet state of the CF chromophore ((3)CF*) with lifetime of 46 mus. In the presence of O2, energy transfer occurred from HSA-(3)CF* to O2 to generate (1)O2; the quenching rate constant [k q(O2)] was determined to be 2.2 x 10 (8) M (-1) s (-1). The HSA-CF showed strong photoresistence relative to HSA complexed with a protoporphyrin IX. The quantum yield of (1)O2 production for this artificial protein was compared to those of other photosensitizing agent. The HSA-CF did not show a dark cytotoxicity, but induced cell death efficiently under visible light irradiation.  相似文献

Seed dormancy is strongly related to the physiological conditions,especially as they relate to responsiveness to ABA, of embryocells during maturation of seeds. In this study, seeds of Triticumaestivum L. cv. Chihoku, which showed nondormancy at harvest,and line Kitakei-l354 (referred to as Kitakei), which showedpost-harvest dormancy, were collected 30 days after anthesis(DPA 30) and at the mature stage (DPA 60). Poly(A)+RNA was extractedfrom the embryos of the seeds and translated in a wheat germsystem. The majority of products of translation from the twogenotypes migrated to the same positions in two-dimensionalgels. Levels of six (for polypeptides h, i, k, m, n, and o)out of 14 Chihoku-specific mRNAs decreased dramatically duringseed maturation, concurrently with the loss of dormancy. Bycontrast, levels of 3 (for polypeptides c, e and f) out of 6Kitakei specific mRNAs were maintained during maturation andduring a 48-h imbibition of the dormant seeds but decreasedat germination. Polypeptides n of Chihoku and e of Kitakei hadthe same molecular size and slightly different pI values. Thesetwo polypeptides may be encoded by the same gene and may playsome role in the maintenance of seed dormancy. Levels of mRNAsfor 10 polypeptides, found in both Chihoku and Kitakei embryosat DPA 30, changed to different extents during maturation. Outof the 10 mRNAs, the relative abundance of 4 mRNAs of Kitakeidid not change dramatically during seed maturation, while inChihoku these mRNAs decreased in level or disappeared duringthe same maturation period. In addition, levels of 2 of these4 mRNAs did not decrease significantly during imbibition ofthe dormant Kitakei seeds but disappeared upon treatment forbreaking of dormancy. The maintenance of these mRNAs in thedormant seeds during maturation and imbibition suggests thatthe respective gene products are involved in the maintenanceof dormancy in wheat seeds. The production of microbial cell substances from hydrocarbons has become the object of commercial attention. We have tested hydrocarbon-utilizing bacterial strains for cell production, and found out one strain of high cell yield, which is very similar to Pseudomonas aeruginosa. The effect of medium composition on cell yield and the utilization of individual hydrocarbons by this strain were investigated. Sixty percent of the added kerosene was converted into cell materials in the following medium of optimum composition: kerosene 2.5%, urea 0.13%, dipotassium phosphate 0.25%, magnesium sulfate 7 aq. 0.1% and tap water. Aliphatic series lower than C10, aromatic and naphthenic hydrocarbons were not or very slightly assimilated. However, aliphatic members higher than C12 were utilized with increasing case. Especially, n-docosane and octadecene-1 were utilized very effectively, and 70% of them were converted into cell substances.  相似文献

Heat evolution during the microbial degradation of glucose in soil was measured with a multiplex heat-conduction calorimeter in the presence of mercury, cadmium, selenium and iodoacetic acid as pollutants. Effects of the pollutant concentration on the degradation thermograms were compared and the results implied that each pollutant had a different type of inhibitory effect on the degradation process of glucose in soil. The dependence of peak times on the thermograms on the pollutant concentration was quantitatively analyzed with a simple mathematical model and possible parameters describing the inhibitory effect of each substance were presented.  相似文献

Objective

To compare the effect of tocilizumab (TCZ) with other biologic therapies in improving anemia of rheumatoid arthritis (RA) patients.

Methods

We compared the change of hemoglobin (Hb) levels in a cohort of 147 consecutive RA patients who were treated with biologics for more than 12 weeks. Twenty eight patients were treated with TCZ, and 119 patients were treated with biologics other than TCZ (87 with TNF inhibitors and 32 with abatacept). The change of Hb levels from baseline to week 12 was compared between the TCZ and the non-TCZ groups. We performed univariate and multivariate analyses with adjustment of potential confounders such as baseline characteristics, concomitant treatment, and the clinical response to treatment.

Results

Hb levels generally increased after biologic therapies both in the TCZ and the non-TCZ groups. The increase of Hb levels was greater in the TCZ group than in the non-TCZ groups (1.1 g/dL in the TCZ group vs 0.3 g/dL in the non-TCZ group, p = 0.009). Univariate analysis revealed that increase of Hb levels was also significantly associated with lower Hb, higher Low Hemoglobin Density, and higher CRP levels at baseline and greater reduction in the clinical disease activity index. TCZ therapy was significantly associated with the increase of Hb levels even after adjustment for these factors by multivariate analysis (p<0.001, effect size 0.08–0.12).

Conclusion

TCZ therapy is an independent factor associated with the increase of Hb level after biologic therapies in RA patients. It will help in selecting appropriate biologics for RA patients with anemia.  相似文献

Epithelial ovarian cancer is classified into four major histological subtypes: serous, clear cell, endometrioid and mucinous. Ovarian clear cell carcinoma (OCCC) responds poorly to conventional chemotherapies and shows poor prognosis. Thus, there is a need to develop new drugs for the treatment of OCCC. In this study, we performed CRISPR/Cas9 screens against OCCC cell lines and identified candidate genes important for their proliferation. We found that quite different genes are required for the growth of ARID1A and PIK3CA mutant and wild-type OCCC cell lines, respectively. Furthermore, we found that the epigenetic regulator KDM2A and the translation regulator PAIP1 may play important roles in the growth of ARID1A and PIK3CA mutant, but not wild-type, OCCC cells. The results of our CRISPR/Cas9 screening may be useful in elucidating the molecular mechanism of OCCC tumorigenesis and in developing OCCC-targeted drugs.  相似文献

We previously cloned a cDNA for sorbitol dehydrogenase (SDH1) from Bombyx mori. In the present study we cloned two additional cDNAs encoding SDHs (designated as SDH2a and SDH2b). The amino acid sequences of SDH2ab were almost the same and had higher similarity to the SDHs of other organisms than to B. mori SDH1. The SDH2ab and SDH1genes were located in tandem within about 40 kbp on chromosome 21. SDH2ab mRNAs increased after exposing diapause eggs to 5 °C for 40 days, beginning at 2 days post-oviposition, to break diapause. However, they were at very low levels in diapausing eggs incubated at 25 °C continuously from oviposition. These changes in expression pattern of SDH2ab mRNA were almost the same as that of SDH1 mRNA. To understand whether SDH1 and SDH2 were responsible for the SDH activity seen in diapause eggs exposed to 5 °C for more than 60 days, we expressed a His-tagged SDH2a fusion protein in Escherichia coli and examined its enzymatic parameters. The maximum activity of SDH2a observed at pH 8.4∼9.0, and the Km value for sorbitol was 12.6 mM, similar to the kinetic properties of other SDHs. Due to the significantly higher similarity between SDH2a and b, they were thought to have similar kinetic properties. Therefore, we purified SDH from B. mori diapause-terminated eggs exposed to 5 °C for 300 days which showed higher SDH activity using two-step affinity chromatography. The highly purified SDH showed a higher Km value (125 mM) for sorbitol, being similar to the value (136 mM) determined previously from Eadie-Hofstee plots using egg crude extract as an enzyme source; additionally, the plots showed one slope indicating one Km value. Moreover, in silico analysis indicated that no SDH genes other than SDH1 and 2ab are present in B. mori genomic DNA. These results suggest that SDH1 activity may be responsible for the majority of the increased SDH activity seen in diapause eggs after acclimation to 5 °C rather than SDH2ab. Further, the relative sequence divergence among these genes is consistent with the idea/hypothesis that the original SDH gene was first duplicated into SDH1 and SDH2, and then SDH2 was duplicated into the SDH2a and SDH2b genes.  相似文献
