Nitric oxide (NO) may affect the genomes of various pathogens, and this mutagenesis is of particular interest for viral pathogenesis and evolution. Here, we investigated the effect of NO on viral replication and mutation. Exogenous or endogenous NO had no apparent antiviral effect on influenza A virus and Sendai virus. The mutagenic potential of NO was analyzed with Sendai virus fused to a green fluorescent protein (GFP) gene (GFP-SeV). GFP-SeV was cultured in SW480 cells transfected with a vector expressing inducible NO synthase (iNOS). The mutation frequency of GFP-SeV was examined by measuring loss of GFP fluorescence of the viral plaques. GFP-SeV mutation frequency in iNOS-SW480 cells was much higher than that in parent SW480 cells and was reduced to the level of mutation frequency in the parent cells by treatment with an NO synthase (NOS) inhibitor. Immunocytochemistry showed generation of more 8-nitroguanosine in iNOS-SW480 cells than in SW480 cells without iNOS transfection. Authentic 8-nitroguanosine added exogenously to GFP-SeV-infected CV-1 cells increased the viral mutation frequency. Profiles of the GFP gene mutations induced by 8-nitroguanosine appeared to resemble those of mutations occurring in mouse lungs in vivo. A base substitution that was characteristic of both mutants (those induced by 8-nitroguanosine and those occurring in vivo) was a C-to-U transition. NO-dependent oxidative stress in iNOS-SW480 cells was also evident. Together, the results indicate unambiguously that NO has mutagenic potential for RNA viruses such as Sendai virus without affecting viral replication, possibly via 8-nitroguanosine formation and cellular oxidative stress.  相似文献

A water-soluble polysaccharide-peptidoglycan complex (PSPG) was prepared from heat-killedLactobacillus casei by digesting the bacteria with N-acetylmuramidase. The molecular weight of PSPG was over 30,000, and the polysaccharide portion of PSPG, its main component was composed of rhamnose, glucose, galactose, glucosamine and galactosamine. Mice pretreated intraperitoneally with PSPG survived after a lethal infection withListeria monocytogenes orPseudomonas aeruginosa. The growth of infecting bacteria (L. monocytogenes, P. aeruginosa, Salmonella typhimurium, Escherichia coli) in both the peritoneal cavity and the liver was inhibited markedly in the mice that had been treated with PSPG. It was suggested that macrophages may be the main effector for the anti-infectious effect of PSPG since treatment of mice with carrageenan, a selective macrophage blocker, markedly reduced the anti-infectious effect of PSPG.  相似文献

Vanadate, which is known to inhibit the plasma membrane H$-ATPaseof Neurospora, was applied intracellularly to internodal cellsof Nitellopsis by use of the intracellular perfusion technique.It inhibited electrogenesis and H$-extrusion, evidence thatthe electrogenic pump of the Characeae plasmalemma is the H$-extrudingone. The concentration of vanadate for the half-maximal inhibitionof the activity of the pump was 5 µM. The membrane potentialand H$-extrusion occasionally recovered from vanadate inhibitionsfor reasons that are unknown. Membrane excitability, which isdependent on Mg?ATP, was not inhibited by vanadate, which suggeststhat the ATPase involved with membrane excitability differsfrom that of the H$ pump. Cytoplasmic streaming took place evenwhen the cell was perfused with the medium containing 1 mM vanadate,which indicates that the vanadate-insensitive actomyosin systemis concerned with the motive force generation of the streaming. A trichloroacetic acid (TCA)-soluble fraction, extracted using cold TCA, was derived from the cell wall of Lactobacillus casei strain S-1. It not only inhibited the adsorption of phage J-l but also desorbed these phages, in their active form, which had previously been adsorbed onto the cell walls. l-Rhamnose, one of the components of this TCA-soluble fraction, had an identical activity to this TCA-soluble fraction, on phage adsorption. This suggested that l-rhamnose is a part of phage receptor material in the cell wall of L. casei strain S-l; and the binding of the phage to the cell wall is reversible, even at 37 C.  相似文献

The transient absorption change for the photoreduction of protochlorophyllide(PChlide) was studied with a solution of extracted PChlide holochromeand with etiolated leaves. The following results were obtained:(1) A short-lived intermediate with an absorption maximum at690 nm was found before formation of chlorophyllide a (Chlide).(2) The intermediate appeared instantly at the onset of theactinic laser, and it decayed very fast with a rate constantof 3.4 x 105 sec–1. Modified techniques for the histochemical demonstration of phosphorylase of value in the diagnosis of cancer cells of human gastro-intestinal origin are described. Preservation of the enzyme activity is especially considered; this is facilitated by the use of thoroughly dried cryostat sections. Routine use of these techniques in the clinical laboratory is suggested. Using these procedures, carcinoma ceils of human stomach and intestines stained in 102 of 117 cases; normal control sections of human gastric epithelial cells are regularly unstained.  相似文献
