Regulation of orexin neurons by the monoaminergic and cholinergic systems   总被引:7,自引:0,他引:7  
Orexins are a pair of neuropeptides implicated in energy homeostasis and arousal. Here we characterize the electrophysiological properties of orexin neurons using slice preparations from transgenic mice in which orexin neurons specifically express green fluorescent protein. Orexin neurons showed high frequency firing with little adaptation by injecting a positive current. The hyperpolarization-activated current was observed in orexin neurons by a negative current injection. The neurotransmitters, which were implicated in sleep/wake regulation, affected the activity of orexin neurons; noradrenaline and serotonin hyperpolarized, while carbachol depolarized orexin neurons in either the presence or absence of tetrodotoxin. It has been reported that orexins directly or indirectly activate the nuclei that are the origin of the neurons containing these neurotransmitters. Our data suggest that orexin neurons have reciprocal neural circuitries between these nuclei for either a positive or negative feedback loop and orchestrate the activity of these neurons to regulate the vigilance states.  相似文献

Embryonic stem (ES) cells derived from the inner cell mass (ICM) of blastocysts grow infinitely while maintaining pluripotency. Leukemia inhibitory factor (LIF) can maintain self-renewal of mouse ES cells through activation of Stat3. However, LIF/Stat3 is dispensable for maintenance of ICM and human ES cells, suggesting that the pathway is not fundamental for pluripotency. In search of a critical factor(s) that underlies pluripotency in both ICM and ES cells, we performed in silico differential display and identified several genes specifically expressed in mouse ES cells and preimplantation embryos. We found that one of them, encoding the homeoprotein Nanog, was capable of maintaining ES cell self-renewal independently of LIF/Stat3. nanog-deficient ICM failed to generate epiblast and only produced parietal endoderm-like cells. nanog-deficient ES cells lost pluripotency and differentiated into extraembryonic endoderm lineage. These data demonstrate that Nanog is a critical factor underlying pluripotency in both ICM and ES cells.  相似文献

tda-1 XY sex reversal occurs when the Y chromosome of at least some populations of wild Mus musculus domesticus is placed on the C57BL/6J genomic background. Gross anatomical observations have previously revealed morphological similarities among fetal ovotestes of tda-1 and Tas-inherited XY sex reversals and BALB/cWt mosaic hermaphrodites. We studied the histology of tda-1 XY sex-reversed gonads, ranging in age from day 14 of gestation to adult. The obtained data revealed additional similarities with ovotestes of BALB/cWt mosaic hermaphrodites as well as with ovotestes of hermaphrodites found in XXSxr and XX/XY chimeras. It is proposed that ovotestes occurring in these various hermaphroditic conditions may be formed through a common pathway.  相似文献

Some properties of hemolysin produced by Vibrio vulnificus were investigated. The hemolysin was heat labile, and the hemolytic activity was inhibited by adding cholesterol or divalent cations. Cholesterol inhibited the temperature-independent hemolysin-binding step, suggesting that cholesterol made up the binding site of the cell membrane, whereas the divalent cations inhibited the temperature-dependent membrane-degradation step. However, the V. vulnificus hemolysin was stable to oxygen and sulfhydryl reagents and was not inactivated by antiserum against streptolysin O, suggesting that the V. vulnificus hemolysin differs from oxygen-labile hemolysins which bind to cholesterol. The V. vulnificus hemolysin seems to be one of the exceptional cholesterol-binding hemolysins.  相似文献

The littoral zone of lakes and lagoons is often used by fish for feeding or reproduction. However, the large changes in temperature that are typical of natural environments, including the littoral zone, represent a potential stressor for fish. Despite the importance of this habitat, little is known about the effect of daily temperature fluctuations on the stress responses of fish. We monitored daily temperature changes in the near-shore and offshore regions of a natural lagoon between May and July 2008–2010. We observed large temperature fluctuations more frequently in the near-shore zone than the offshore zone. We then exposed common carp (Cyprinus carpio) to a temperature regime similar to that observed in the near-shore zone and measured the levels of cortisol released into the water. The rate of cortisol release increased when carp were exposed to an increase in temperature of ~0.6°C/h over a 5-h period. Conversely, there was no change in the rate of release when temperatures decreased. Our results highlight the importance of maintaining high temporal resolution when evaluating the stress response to daily fluctuations temperature.  相似文献

The growth-inhibitory effects of Bacillus natto and Streptococcus faecalis on Canida albicans were investigated. When inoculated into the filtrate of a long-term culture of B. natto strain BN (BN), a stock culture of C. albicans RIMD 0301020 lost its viability completely, whereas C. albicans RIMD 0301011, a fresh isolate from a clinical source, did not. In continuous flow (CF) culture the growth of both strains of C. albicans was suppressed by mixed cultivation with BN. On the other hand, in classical batch culture BN did not suppress the growth of C. albicans. S. faecalis BIO-4R, a multi-drug resistant strain, was also antagonistic to C. albicans RIMD 0301011 but symbiotic with BN in CF culture. These findings suggest that BN in concert with S. faecalis BIO-4R may inhibit the growth of C. albicans in the intestinal tract.  相似文献

Interleukin-6 (IL-6) induces growth arrest and macrophage differentiation through its receptor in a murine myeloid leukaemic cell line, M1, although it is largely unknown how the IL-6 receptor generates these signals. By using chimeric receptors consisting of the extracellular domain of growth hormone receptor and the transmembrane and cytoplasmic domain of gp130 with progressive C-terminal truncations, we showed that the membrane-proximal 133, but not 108, amino acids of gp130 could generate the signals for growth arrest, macrophage differentiation, down-regulation of c-myc and c-myb, induction of junB and IRF1 and Stat3 activation. Mutational analysis of this region showed that the tyrosine residue with the YXXQ motif was critical not only for Stat3 activation but also for growth arrest and differentiation, accompanied by down-regulation of c-myc and c-myb and immediate early induction of junB and IRF1. The tight correlation between Stat3 activation and other IL-6 functions was further observed in the context of the full-length cytoplasmic region of gp130. The result suggest that Stat3 plays an essential role in the signals for growth arrest and differentiation.  相似文献

We have isolated suppressor mutants that suppress temperature-sensitive colony formation and anucleate cell production of a mukB mutation. A linkage group (smbB) of the suppressor mutations is located in the rne/ams/hmp gene encoding the processing endoribonuclease RNase E. All of the rne (smbB) mutants code for truncated RNase E polypeptides lacking a carboxyl-terminal half. The amount of MukB protein was higher in these rne mutants than that in the rne+ strain. These rne mutants grew nearly normally in the mukB+ genetic background. The copy number of plasmid pBR322 in these rne mutants was lower than that in the rne+ isogenic strain. The results suggest that these rne mutations increase the half-lives of mukB mRNA and RNAI of pBR322, the antisense RNA regulating ColE1-type plasmid replication. We have demonstrated that the wild-type RNase E protein bound to polynucleotide phosphorylase (PNPase) but a truncated RNase E polypeptide lacking the C-terminal half did not. We conclude that the C-terminal half of RNase E is not essential for viability but plays an important role for binding with PNPase. RNase E and PNPase of the multiprotein complex presumably cooperate for effective processing and turnover of specific substrates, such as mRNAs and other RNAs in vivo.  相似文献
