Growth of chimpanzees reared at the Kumamoto Primates Park of Sanwa Kagaku Kenkyusho Co. Ltd. was studied cross-sectionally
from the viewpoints of somatic growth and reproductive maturation. Distance and velocity curves were expressed using spline
function method. Males showed adolescent growth acceleration in body weight, with a peak at 7.86 yrs of age, but not in trunk
length. Females showed continuous rapid growth from mid-juvenile to adolescent phase in both body weight and trunk length,
but no isolated adolescent spurt. The Sanwa chimpanzees matured at about 12.5 yrs of age for females and 15.0 yrs for males.
The mean adult weights and trunk lengths were 53.2 kg and 507.8 mm for males and 42.7 kg and 481.6 mm for females. The Sanwa
chimpanzees had similar growth patterns to those of the Yerkes chimpanzees, although they showed a slight delay in infancy,
and a higher growth rate from the early juvenile phase onwards. Growth patterns in these two laboratories may be regarded
as “normative” for laboratory-reared chimpanzees. They matured earlier than wild chimpanzees by more than two years. The major
reason for the retarded maturation in wild chimpanzees is the delay of growth from infant to the early juvenile phases (0–4
yrs of age), probably owing to a limited nutritional supply from the mother. Development of the testes comprised three phases:
slow growth from infant to juvenile (until 6.4 yrs); rapid growth around adolescence (until 9.2 yrs); and adult (mean testicular
volume, 187 cm3). Setting the nutritional standard at 2,000–2,600 Cal/day (= Kcal/day) per adult, calories were considered for captive chimpanzees

Benzyladenine-induced changes in the translatable mRNA population in excised cucumber cotyledons were studied. Poly (A)+ RNA was prepared from etiolated cotyledons incubated with or without benzyladenine (BA) for various periods in the dark. Using nonequilibrium pH gradient electrophoresis-SDS polyacrylamide gel electrophoresis and isoelectric focusing-SDS polyacrylamide gel electrophoresis, both basic and neutral proteins translated in vitro were separated. Mouse mammary tumor viruses (MMTVs) contain distinct membrane glycoproteins of 52,000 daltons (gp52) and 36,000 daltons (gp36). We report here the development of new radioimmunoassays for gp36, using gp36 purified by hydrophobic chromatography and gel filtration. These assays demonstrate that gp36 has both type-specific and group-specific antigenic determinants. The virus-coded nature of these determinants was shown by utilizing different MMTVs grown in the same feline cell line. Interspecies determinants on gp36 were demonstrated by the observations that (i) MC-MTV (a virus isolate from the Asian rodent Mus cervicolor, and morphologically identical to MMTVs) competed, with an altered slope, in the gp36 radioimmunoassay, and (ii) antisera raised against MC-MTV immunopreciptitated 125I-labeled gp36. The detection of gp36 in spontaneous mammary tumors of several strains of mice also facilitates further studies on the replication of MMTVs and the host's immune response to MMTV-mediated oncogenesis. 相似文献

Under the influence of maturation-inducing hormone (MIH) secreted from follicle cells, oocyte maturation is finally triggered by maturation-promoting factor (MPF), which consists of a homolog of the cdc2+ gene product of fission yeast (p34cdc2) and cyclin B. Two species of cyclin B clones were isolated from a cDNA library constructed from mature goldfish oocytes. Sequence comparisons revealed that these two clones are highly homologous (95%) and were found to be similar to Xenopus cyclin B1. Using monoclonal antibodies against Escherichia coli-produced goldfish cyclin B and the PSTAIR sequence of p34cdc2, we examined the levels of cyclin B and p34cdc2 proteins during goldfish oocyte maturation induced in vitro by 17 alpha, 20 beta-dihydroxy-4-pregnen-3-one (17 alpha, 20 beta-DP), a natural MIH in fish. Protein p34cdc2 was found in immature oocyte extracts and did not remarkably change during oocyte maturation. Cyclin B was not detected in immature oocyte extracts and appeared when oocytes underwent germinal vesicle breakdown. Cyclin B that appeared during oocyte maturation was labelled with [35S]methionine, indicating its de novo synthesis. Introduction of E. coli-produced cyclin B into immature oocyte extracts induced p34cdc2 (MPF) activation. Although the possibility that immature goldfish oocytes contain an insoluble cyclin B is not completely excluded, these results strongly suggest that 17 alpha, 20 beta-DP induces oocytes to synthesize cyclin B, which in turn activates preexisting p34cdc2, forming active MPF. 相似文献

Maturation-promoting factor and a homolog of fission yeast cdc2+ gene product (p34cdc2) were investigated during the final 24 hr of maturation of quail oocytes. Kinase activity of p34cdc2 in the oocyte germinal disk (GD) increased 15 times at maturation. Two bands, at 32 and 34 kDa, were detected in immature oocytes by immunoblotting of SDS-PAGE with anti-p34cdc2 monoclonal antibody. A new band, which is close to the 32-kDa band but with a slightly faster mobility, appeared during maturation. No p34cdc2 could be detected outside the GD. When polyethyleneimine (PEI), a water-soluble cationic polymer, was added to solutions of glucose dehydrogenase (GDH) from Bacillus megaterium IWG3 at a molar ratio of PEI to GDH greater than 10, the thermal stability of GDH markedly increased. By addition of PEI, the rate of GDH-catalysed oxidation of -d-glucose increased in a low concentration range of NAD+ and NADP+ and the Michaelis constants and inhibition constants for both NAD+ and NADP+ decreased. These results suggested that negatively charged GDH interacts with cationic water-soluble polymers to form conjugates by electrostatic attraction, and also that negatively charged coenzymes are adsorbed by the polymers, resulting in enrichment of the coenzymes in the vicinity of GDH. Eleven urea compounds were administered individually as a single oral dose to rats either on day 12 or 14 and to mice on day 10 of pregnancy. 1-Methylurea and 1-ethylurea were not teratogenic in either of the animal species. Administration of 1-methylthiourea and 1-ethylthiourea, however, resulted in high incidences of malformations in the rat but not in the mouse fetuses. The types of malformation were similar to those produced by the known teratogen ethylenethiourea. Methylated ureas such as 1,3-dimethylurea, 1,3-dimethylthiourea, 1,1,3,3-tetramethylurea, and 1,1,3,3-tetramethylthiourea were fetotoxic and produced malformations in the tail, palate, or extremities of the surviving rat and/or mouse fetuses. These findings suggest that the teratogenic properties of N-alkylureas can be categorized into two groups, i.e., mono-alkylated thioureas and methylated ureas or thioureas. 相似文献

The effects of nucleoid proteins Fis and Dps of Escherichia coli on the higher order structure of a giant DNA were studied, in which Fis and Dps are known to be expressed mainly in the exponential growth phase and stationary phase, respectively. Fis causes loose shrinking of the higher order structure of a genome-sized DNA, T4 DNA (166 kbp), in a cooperative manner, that is, the DNA conformational transition proceeds through the appearance of a bimodal size distribution or the coexistence of elongated coil and shrunken globular states. The effective volume of the loosely shrunken state induced by Fis is 30–60 times larger than that of the compact state induced by spermidine, suggesting that cellular enzymes can access for DNA with the shrunken state but cannot for the compact state. Interestingly, Dps tends to inhibit the Fis-induced shrinkage of DNA, but promotes DNA compaction in the presence of spermidine. These characteristic effects of nucleotide proteins on a giant DNA are discussed by adopting a simple theoretical model with a mean-field approximation. 相似文献
