Six 3H-spiro[benzofuran-2,1'-cyclohexane] derivatives were synthesized from naturally occurring filifolinol, and their classical complement pathway inhibitory activity was determined. IC(50) values of the most potent compounds were comparable to the activity of the natural complement inhibitor K76-COOH and some synthetic tricyclic analogs of it.  相似文献

In Mediterranean ecosystems the effect of aboveground and belowground environmental factors on soil microbial biomass and nutrient immobilization-release cycles may be conditioned by the distinctive seasonal pattern of the Mediterranean-type climates. We studied the effects of season, canopy cover and soil depth on microbial C, N and P in soils of two Mediterranean forests using the fumigation-extraction procedure. Average microbial values recorded were 820 μg C g?1, 115 μg N g?1 and 19 μg P g?1, which accounted for 2.7, 4.7 and 8.8% of the total pools in the surface soil, respectively. Microbial N and P pools were about 10 times higher than the inorganic N and P fractions available for plants. Microbial C values differed between forest sites but in each site they were similar across seasons. Both microbial and inorganic N and P showed maximum values in spring and minimum values in summer, which were positively correlated with soil moisture. Significant differences in soil microbial properties among canopy cover types were observed in the surface soil but only under favourable environmental conditions (spring) and not during summer. Soil depth affected microbial contents which decreased twofold from surface to subsurface soil. Microbial nutrient ratios (C/N, C/P and N/P) varied with seasons and soil depth. Soil moisture regime, which was intimately related to seasonality, emerged as a potential key factor for microbial biomass growth in the studied forests. Our research shows that under a Mediterranean-type climate the interaction among season, vegetation type and structure and soil properties affect microbial nutrient immobilization and thus could influence the biogeochemical cycles of C, N and P in Mediterranean forest ecosystems.  相似文献

Due to the spread of Visceral leishmaniasis in Brazilian territory, this research was undertaken with the objective to identify the sandfly fauna and aspects of the species behavior in forest and anthropic environments in Paraná State, Brazil. The sandfly captures were sporadically carried out in 37 municipalities of the Paraná state, during the period from March 2004 to November 2005. Each municipality was once visited, when Falc?o traps were installed during three consecutive nights in forested fragments and anthropic environments; captures with Shannon traps installed in forests and edge of forests, inspection of walls and electric aspiration in domiciles and forested areas were sporadically undertaken. The data analysis was based on estimates of frequency and abundance of the species, by five native landscapes. A total of 38,662 specimens belonging to 23 species were captured. Nyssomyia neivai (Pinto) (75.6%), Ny. whitmani (Antunes & Coutinho) (10.1%), Migonemyia migonei (Fran?a) (7.8%), Expapillata firmatoi (Barreto et al.) (2.1%) and Pintomyia fischeri (Pinto) (1.6%) together constituted 97.2% of the phlebotomine collected. High frequencies of sand flies were found in forests, henhouses, pig-pens and domiciles. The three most frequent species have been implicated in transmission of tegumentary leishmaniasis in Paraná and in the Brazilian Southeastern and Southern Regions. Lutzomyia longipalpis (Lutz & Neiva), the visceral leishmaniasis vector, was not found, however Lutzomyia gaminariai (Cordero et al.), female of which is similar to that of Lu. longipalpis was captured, raising the need to investigate its behavior, including as regard to the vectorial competence.  相似文献

OBJECTIVE: To provide updated, evidence-based recommendations for health care professionals on lifestyle changes to prevent and control hypertension in otherwise healthy adults (except pregnant women). OPTIONS: For people at risk for hypertension, there are a number of lifestyle options that may avert the condition--maintaining a healthy body weight, moderating consumption of alcohol, exercising, reducing sodium intake, altering intake of calcium, magnesium and potassium, and reducing stress. Following these options will maintain or reduce the risk of hypertension. For people who already have hypertension, the options for controlling the condition are lifestyle modification, antihypertensive medications or a combination of these options; with no treatment, these people remain at risk for the complications of hypertension. OUTCOMES: The health outcomes considered were changes in blood pressure and in morbidity and mortality rates. Because of insufficient evidence, no economic outcomes were considered. EVIDENCE: A MEDLINE search was conducted for the period January 1996 to September 1996 for each of the interventions studied. Reference lists were scanned, experts were polled, and the personal files of the authors were used to identify other studies. All relevant articles were reviewed, classified according to study design and graded according to level of evidence. VALUES: A high value was placed on the avoidance of cardiovascular morbidity and premature death caused by untreated hypertension. BENEFITS, HARMS AND COSTS: Lifestyle modification by means of weight loss (or maintenance of healthy body weight), regular exercise and low alcohol consumption will reduce the blood pressure of appropriately selected normotensive and hypertensive people. Sodium restriction and stress management will reduce the blood pressure of appropriately selected hypertensive patients. The side effects of these therapies are few, and the indirect benefits are well known. There are certainly costs associated with lifestyle modification, but they were not measured in the studies reviewed. Supplementing the diet with potassium, calcium and magnesium has not been associated with a clinically important reduction in blood pressure in people consuming a healthy diet. RECOMMENDATIONS: (1) It is recommended that health care professionals determine the body mass index (weight in kilograms/[height in metres]2) and alcohol consumption of all adult patients and assess sodium consumption and stress levels in all hypertensive patients. (2) To reduce blood pressure in the population at large, it is recommended that Canadians attain and maintain a healthy body mass index. For those who choose to drink alcohol intake should be limited to 2 or fewer standard drinks per day (maximum of 14/week for men and 9/week for women). Adults should exercise regularly. (3) To reduce blood pressure in hypertensive patients, individualized therapy is recommended. This therapy should emphasize weight loss for overweight patients, abstinence from or moderation in alcohol intake, regular exercise, restriction of sodium intake and, in appropriate circumstances, individualized cognitive behaviour modification to reduce the negative effects of stress. VALIDATION: The recommendations were reviewed by all of the sponsoring organizations and by participants in a satellite symposium of the fourth international Conference on Preventive Cardiology. They are similar to those of the World Hypertension League and the Joint National committee, with the exception of the recommendations on stress management, which are based on new information. They have not been clinically tested. The guidelines are intended for use in clinical practice and public education campaigns. OPTIONS: Restriction of dietary salt intake may be an alternative to antihypertensive medications or may supplement such medications. Other options include other nonpharmacologic treatments for hypertension and no treatment. OUTCOMES: The health outcomes considered were changes in blood pressure and in morbidity and mortality rates. Because of insufficient evidence, no economic outcomes were considered. EVIDENCE: A MEDLINE search was conducted for the period 1966-1996 using the terms hypertension, blood pressure, vascular resistance, sodium chloride, sodium, diet, sodium or sodium chloride dietary, sodium restricted/reducing diet, clinical trials, controlled clinical trial, randomized controlled trial and random allocation. Both trials and review articles were obtained, and other relevant evidence was obtained from the reference lists of the articles identified, from the personal files of the authors and through contacts with experts. The articles were reviewed, classified according to study design and graded according to level of evidence. In addition, a systematic review of all published randomized controlled trials relating to dietary salt intake and hypertension was conducted. VALUES: A high value was placed on the avoidance of cardiovascular morbidity and premature death caused by untreated hypertension. BENEFITS, HARMS AND COSTS: For normotensive people, a marked change in sodium intake is required to achieve a modest reduction in blood pressure (there is a decrease of 1 mm Hg in systolic blood pressure for every 100 mmol decrease in daily sodium intake). For hypertensive patients, the effects of dietary salt restriction are most pronounced if age is greater than 44 years. A decrease of 6.3 mm Hg in systolic blood pressure and 2.2 mm Hg in diastolic blood pressure per 100 mmol decrease in daily sodium intake was observed in people of this age group. For hypertensive patients 44 years of age and younger, the decreases were 2.4 mm Hg for systolic blood pressure and negligible for diastolic blood pressure. A diet in which salt is moderately restricted appears not to be associated with health risks. RECOMMENDATIONS: (1) Restriction of salt intake for the normotensive population is not recommended at present, because of insufficient evidence demonstrating that this would lead to a reduced incidence of hypertension. (2) To avoid excessive intake of salt, people should be counselled to choose foods low in salt (e.g., fresh fruits and vegetables), to avoid foods high in salt (e.g., pre-prepared foods), to refrain from adding salt at the table and minimize the amount of salt used in cooking, and to increase awareness of the salt content of food choices in restaurants. (3) For hypertensive patients, particularly those over the age of 44 years, it is recommended that the intake of dietary sodium be moderately restricted, to a target range of 90-130 mmol per day (which corresponds to 3-7 g of salt per day). (4) The salt consumption of hypertensive patients should be determined by interview. VALIDATION: These recommendations were reviewed by all of the sponsoring organizations and by participants in a satellite symposium of the fourth International Conference on Preventive Cardiology. They have not been clinically tested. Lethal concentration (LC) has been widely used to estimate pesticide toxicity. However, it does not consider the sub-lethal effects. Therefore we included the instantaneous rate of increase in association with LC to estimate population-level effects of the acaricides fenbutatin oxide and sulfur on the predator Iphiseiodes zuluagai and its prey, the phytophagous southern red mite, Oligonychus ilicis. The predator was 32.84x and 17.20x more tolerant to fenbutatin oxide and sulfur, respectively, than its prey, based on LC50 estimates obtained from acute concentration–mortality bioassays. The instantaneous rate of population growth in both mite species decreased with increasing acaricide concentration. Both acaricides provided effective control of O. ilicis at their recommended concentrations, but sulfur drastically compromised the predator populations quickly leading them to extinction due to the low reproductive potential of this species compared with its prey.  相似文献

Cerebellar granule neurons undergo apoptosis when switched from a medium containing high potassium (HK) to one that has low potassium (LK). LK-induced cell death is blocked by GW5074 [5-Iodo-3-[(3,5-dibromo-4-hydroxyphenyl) methylene]-2-indolinone], a synthetic drug that inhibits c-Raf activity in vitro. GW5074 has no direct effect on the activities of several apoptosis-associated kinases when assayed in vitro. In contrast to its effect in vitro, treatment of neurons with GW5074 causes c-Raf activation (when measured in vitro in the absence of the drug) and stimulates the Raf-MEK-ERK pathway. Treatment of neurons with GW5074 also leads to an increase in the activity of B-Raf, which is not inhibited by GW5074 in vitro at concentrations at which the drug exerts its neuroprotective effect. PD98059 and U0126, two distinct inhibitors of MEK, block the activation of ERK by GW5074 but have no effect on its ability to prevent cell death. Overexpression of a dominant-negative form of Akt does not reduce the efficacy of GW5074, demonstrating an Akt-independent mechanism of action. Neuroprotection is inhibited by SN-50, a specific inhibitor of nuclear factor-kappa B (NF-kappaB) and by the Ras inhibitor S-trans, trans-farnesylthiosalicylic acid (FTS) implicating NF-kappaB and Ras in the neuroprotective signaling pathway activated by GW5074. In addition to preventing LK-induced apoptosis, treatment with GW5074 protects against the neurotoxic effects of MPP+ and methylmercury in cerebellar granule neurons, and glutathione depletion-induced oxidative stress in cortical neurons. Furthermore, GW5074 prevents neurodegeneration and improves behavioral outcome in an animal model of Huntington's disease. Given its neuroprotective effect on distinct types of cultured neurons, in response to different neurotoxic stimuli, and in an animal model of neurodegeneration, GW5074 could have therapeutic value against neurodegenerative pathologies in humans.  相似文献

Billfishes from the Brazilian oceanic northeastern region feed on a large food spectrum composed mainly of epipelagic species of fish and cephalopods, with occasional occurrences of mesopelagic organisms and crustaceans. The oceanic pomfret, Brama brama, and the squid, Ornithoteuthis antillarum, were the main prey items in the diet of the four species, occurring in at least 50% of the stomachs containing food. There was no correlation between body length of prey and predator, or between mass of stomach content and body mass in the individuals from 100 to 330cm fork length. The feeding of the four species was characterized by constant ingestion of small quantities of food, and constant presence of food in the stomachs, with a predominance of epipelagic organisms of small sizes with an average of 8.0cm. The reduced stomach size forces the predators to feed constantly on several meals during the day, and consequently provide constant energy for migration.  相似文献
