Recent reports identified DGAT1 (EC 2.3.1.20) harboring a lysine to alanine substitution (K232A) as a candidate gene with a strong effect on milk production traits. Our objective was to estimate the frequency of the DGAT1 K232A polymorphism in the main Zebu and Taurine breeds in Brazil as well as in Zebu x Taurine crossbreds as a potential QTL for marker-assisted selection. Samples of 331 animals from the main Brazilian breeds, Nellore, Guzerat, Red Sindhi, Gyr, Holstein, and Gyr x Holstein F1 were genotyped for DGAT1 K232A polymorphism (A and K alleles) using the PCR-RFLP technique. The highest frequency of the A allele was found in the Holstein sample (73%) followed by Gyr x Holstein F1 (39%). Gyr and Red Sindhi showed low frequencies of A alleles (4 and 2.5%, respectively). The A allele was not found in the Nellore and Guzerat samples. Our results could be used to guide association studies between this locus and milk traits in these breeds.  相似文献

Distribution of parasitic plants is directly linked with the distribution of host species and behavioral patterns of seed dispersers. Psittacanthus robustus (Loranthaceae) is a neotropical hemiparasite that mainly colonizes species of the family Vochysiaceae. Vochysia thyrsoidea is the main host of P. robustus and is commonly found in areas of cerrado rupestre (rocky savanna), an abundant vegetation in our study site. We conducted the study in the ecological park Parque Ecológico Quedas do Rio Bonito (PEQRB), over an area of 2.82 ha of cerrado rupestre. The objective of this work was to investigate population structure, parasitic behavior (mistletoe grip height and circumference of host branch), and spatial distribution of Psittacanthus robustus on a population of Vochysia thyrsoidea. We sampled 267 V. thyrsoidea individuals and found that the population had a random distribution pattern. Seventy-nine individuals (29.6% of the sample population) hosted the hemiparasite, to a total of 193 P. robustus individuals. The number of mistletoe individuals per host plant ranged between 1 and 12. The V. thyrsoidea individuals most infested with mistletoes were those reaching greater heights. The correlation between height of host plant and preferred grip height was highly significant, with the preferred grip height being the uppermost portions of host plants. The crown size of P. robustus individuals ranged between 10 and 230 cm. The main disperser of P. robustus fruit was found to be swallow-tanager Tersina viridis viridis. Its activities led to a clumped pattern of spatial distribution of the hemiparasite along with higher infestation in larger trees.  相似文献

Laboratory studies are often criticized for not being representative of processes occurring in natural populations. One reason for this is the fact that laboratory populations generally do not capture enough of the genetic variation of natural populations. This can be mitigated by mixing the genetic background of several field populations when creating laboratory populations. From these outbred populations, it is possible to generate inbred lines, thereby freezing and partitioning part of their variability, allowing each genotype to be characterized independently. Many studies addressing adaptation of organisms to their environment, such as those involving quantitative genetics or experimental evolution, rely on inbred or outbred populations, but the methodology underlying the generation of such biological resources is usually not explicitly documented. Here, we developed different procedures to circumvent common pitfalls of laboratory studies, and illustrate their application using two haplodiploid species, the spider mites Tetranychus urticae and Tetranychus evansi. First, we present a method that increases the chance of capturing high amounts of variability when creating outbred populations, by performing controlled crosses between individuals from different field‐collected populations. Second, we depict the creation of inbred lines derived from such outbred populations, by performing several generations of sib‐mating. Third, we outline an experimental evolution protocol that allows the maintenance of a constant population size at the beginning of each generation, thereby preventing bottlenecks and diminishing extinction risks. Finally, we discuss the advantages of these procedures and emphasize that sharing such biological resources and combining them with available genetic tools will allow consistent and comparable studies that greatly contribute to our understanding of ecological and evolutionary processes.  相似文献

The objective of this study was to analyse the feeding habits and trophic interactions between four oceanic predatory fish around the Fernando de Noronha Archipelago (FNA), Brazil, in the western equatorial Atlantic (3.86°S/32.42°W), internationally recognized as an environment of high economic and ecological value. For this purpose, biological samples of yellowfin tuna (Thunnus albacares), wahoo (Acanthocybium solandri), barracuda (Sphyraena barracuda) and dolphinfish (Coryphaena hippurus) were collected for stomach content and stable isotope analysis. Values of the index of relative importance revealed varied diets, with a strong presence of teleost fishes (Diodontidae and Exocoetidae) for all species, with yellowfin tuna having a greater diversity of food items. Despite being generalists/opportunists, the feeding strategy of these predators showed a tendency towards a specialized diet in the use of the available resources around the FNA. They presented a narrow trophic niche width (Levin's index, Bi < 0.6) and low overlap between species, except between barracuda and wahoo (MacArthur and Levin's, R0 = 0.72). Isotopic compositions had broad values of δ13C and δ15N, and were significantly different between species. Our results provide information about the four species' trophic organization and suggest that the predators avoid competition by preying on different prey, thus allowing their coexistence.  相似文献

DNA methylation is linked to homocysteine metabolism through the generation of S-adenosylmethionine (AdoMet) and S-Adenosylhomocysteine (AdoHcy). The ratio of AdoMet/AdoHcy is often considered an indicator of tissue methylation capacity. The goal of this study is to determine the relationship of tissue AdoMet and AdoHcy concentrations to allele-specific methylation and expression of genomically imprinted H19/Igf2. Expression of H19/Igf2 is regulated by a differentially methylated domain (DMD), with H19 paternally imprinted and Igf2 maternally imprinted. F1 hybrid C57BL/6J x Castaneous/EiJ (Cast) mice with (+/−), and without (+/+), heterozygous disruption of cystathionine-β-synthase (Cbs) were fed a control diet or a diet (called HH) to induce hyperhomocysteinemia and changes in tissue AdoMet and AdoHcy. F1 Cast x Cbs+/− mice fed the HH diet had significantly higher plasma total homocysteine concentrations, higher liver AdoHcy, and lower AdoMet/AdoHcy ratios and this was accompanied by lower liver maternal H19 DMD allele methylation, lower liver Igf2 mRNA levels, and loss of Igf2 maternal imprinting. In contrast, we found no significant differences in AdoMet and AdoHcy in brain between the diet groups but F1 Cast x Cbs+/− mice fed the HH diet had higher maternal H19 DMD methylation and lower H19 mRNA levels in brain. A significant negative relationship between AdoHcy and maternal H19 DMD allele methylation was found in liver but not in brain. These findings suggest the relationship of AdoMet and AdoHcy to gene-specific DNA methylation is tissue-specific and that changes in DNA methylation can occur without changes in AdoMet and AdoHcy.  相似文献

Several species of arthropods inhabiting forest fragments interact with managed areas. The importance of such areas to biodiversity conservation, however, is not well established. Communities of solitary wasps and bees (Insecta: Hymenoptera) play a key role in agroecosystem functioning and they have been used in studies of biodiversity assessment in different land‐use types. We aimed to assess patterns of species richness and composition of solitary wasps and bees over a 1‐yr period in a gradient of decreasing land‐use intensity formed by pastures, alley croppings, young fallows, and old fallows using trap nests. Old fallows had the highest species richness of wasps and bees, harboring all bee species and 86 percent of wasp species occurring in the region, while the remaining land‐uses had similar species richness. Vegetation structure (tree richness) and relative humidity explained most of the variance for the species richness of wasps. For bees, however, there was no influence of environmental factors on the community among land‐use types, indicating better adaptability of this group to environmental variations related to land‐use. The composition of solitary wasp communities (but not those of bees) differed among land‐use types, and the occurrence of rare species in most cases was restricted to old fallow sites. In conclusion, the community of solitary wasps and bees is contingent on land‐use, with solitary wasps more sensitive to anthropized areas. For both groups, less anthropized areas harbor a greater richness and number of rare species while more intensively managed land‐use types harbor higher abundances.  相似文献

The adenovirus E4orf4 protein selectively kills human cancer cells independently of p53 and thus represents a potentially promising tool for the development of novel antitumor therapies. Previous studies suggested that E4orf4 induces an arrest or a delay in mitosis and that both this effect and subsequent cell death rely largely on an interaction with the B55 regulatory subunit of protein phosphatase 2A. In the present report, we show that the death of human H1299 lung carcinoma cells induced by expression of E4orf4 is typified not by an accumulation of cells arrested in mitosis but rather by the presence of both tetraploid and diploid cells that are arrested in G1 because they are unable to initiate DNA synthesis. We believe that these E4orf4-expressing cells eventually die by various processes, including those resulting from mitotic catastrophe.  相似文献

The cassava green mite, Mononychellus tanajoa, is a key pest of cassava, Manihot esculenta Crantz (Euphorbiaceae), and it may be kept in check by naturally occurring predatory mites of the family Phytoseiidae. In addition to predatory mites, abiotic factors may also contribute to regulate pest mite populations in the field. Here, we evaluated the population densities of both M. tanajoa and the generalist predatory mite Euseius ho DeLeon (Acari: Phytoseiidae) over the cultivation cycle (11 months) of cassava in four study sites located around the city of Miranda do Norte, Maranhão, Brazil. The abiotic variables rainfall, temperature and relative humidity were also recorded throughout the cultivation cycle of cassava. We determined the relative importance of biotic (density of E. ho) and abiotic (rainfall, temperature and relative humidity) factors to the density of M. tanajoa. The density of M. tanajoa increased whereas the density of E. ho remained constant throughout time. A hierarchical partitioning analysis revealed that most of the variance for the density of M. tanajoa was explained by rainfall and relative humidity followed by E. ho density and temperature. We conclude that abiotic factors, especially rainfall, were the main mechanisms driving M. tanajoa densities.  相似文献
