Light absorbance and reflectance were measured on representative shrubs of two dominant shrub species (Pistacia lentiscus and Juniperus phoenicea) of coastal Mediterranean macchia ecosystems in Sardinia (Italy). An array of 64 calibrated photodiodes was adopted for both calculations of leaf area index (LAI) and canopy transmittance of photosynthetic active radiation (PAR). PAR and NIR reflectance of canopies having similar LAI allowed us to establish the particular optical properties of each species. Lentisk reflected twice as much as juniper in the 700-1,100 spectral region. Water indexes of the two species were also different in relation to leaf water content and red edge amplitude appeared to be related to chlorophyll content. In this study, we detected Leishmania (Viannia) braziliensis infection in equids living in endemic regions of cutaneous leishmaniasis. To determine the role of these animals in the Leishmania cycle, we used two approaches: serological and molecular methods. Antibodies to the parasite were assayed using the Enzyme Linked Immunosorbent Assay (ELISA). Blood samples were collected and tested by polymerase chain reaction (PCR), and the positive products were sequenced. The results showed that 11.0% (25/227) of the equids were seropositive for Leishmania sp, and 16.3% (37/227) were PCR positive. Antibodies were detected in 20 horses, 3 donkeys, and 2 mules, and the parasite DNA was detected in 30 horses, 5 donkeys, and 2 mules. Sequencing the amplified DNA revealed 100% similarity with sequences for Viannia complex, corroborating the results of PCR for L. braziliensis. Our results show that equids are infected with L. braziliensis, which could be food sources for phlebotomines in the peridomiciliary environment and consequently play a role in the cutaneous leishmaniasis cycle.  相似文献

In vitro culture of explants were used to apply genetic or cell engineering techniques to the sexually incompatible potato relative Solanum commersonii (2n=2x=24) Three accessions of S. commersonii were tested for regeneration from leaf explants using six different protocols. A two step-regeneration procedure gave the best results. Genetic variability for regeneration ability was found between accessions, and between clones within accessions. The accession PI 472834 regenerated at highest frequency. Clones with high regeneration ability were selected. Approximately 60% of regenerated plants were diploids and 40% were tetraploids. A very low frequency of chimeras was found. Leaf shape and chloroplast counts in guard cells were shown to be quick and reliable methods for estimating ploidy levels. Somatic fusion of Solanum commersonii, a frost tolerant wild potato species not crossable with Solanum tuberosum, relies on the possibility to isolate and culture protoplasts. This study was conducted to determine whether protoplasts could be isolated and plants regenerated in three S. commersonii accessions. Shoot cultures for protoplast isolation were maintained on Murashige and Skoog medium. Mesophyll protoplasts were isolated and cultured using a protocol originally described for S. tuberosum with some modifications. Differences were evident among the three accessions for protoplast yield, plating efficiency and regeneration frequency. Protoplast yield ranged from 3.0 to 8.5 × 106 protoplasts per g of fresh tissue. At 1–2 × 104 protoplasts ml−1, which was the optimal plating density, 10–20% of plated protoplasts gave multicellular colonies. Regeneration of shoots was observed in two accessions only, the maximum regeneration frequency being 66%. In one of these accessions the reduction of sucrose concentration in regeneration media improved the regeneration frequency from 14 to 35%. About three hundred plants were rooted in vitro and successfully transferred to soil.  相似文献

Lifetime dairy production, reproduction and growth traits of 75 females sired by Holstein, Jersey or Brown Swiss bulls and Holstein-Friesian x Gir dams of 1/2 to 3/4 Holstein-Friesian fractions were compared. The animals were in a single herd under the same management. Milk, fat and protein yields, concentrates fed, reproduction, and weights were recorded throughout the lifetime of the cows. The data were analyzed by least squares techniques under models including the fixed effects of breed of sire, Bos taurus fraction and year of birth. Herd lifes for Holstein, Jersey and Brown Swiss crosses were 6.006 +/- 0.812, 8.129 +/- 0.863 and 7.247 +/- 0.777 years. Milk yields per day of herd life were 7.150 +/- 0.266, 6.757 +/- 0.282 and 6.249 +/- 0.254 kg. Weights of cull cows sold were 458 +/- 15, 415 +/- 15 and 457 +/- 13 kg. Based on these and on previously reported results of the same experiment, intakes of roughage and pasture were estimated from energy requirements. Lifetime expenditures on concentrates, roughages, pastures, milking, reproduction, and heifer rearing were calculated based on mean performance of each breed of sire, as well as on receipts from animals and milk sold (the latter with four sets of prices of protein, fat and carrier). The conclusion was that in systems of artificial female calf rearing and male calf wastage, the Jersey crosses appear to offer important economic benefits to farmers, which would be even greater if payment on milk protein and fat becomes effective.  相似文献

By structure–activity relationship studies on the tilorone scaffold, the ‘one armed’ substituted dibenzothiophenes and the fluoren-9-ones were identified as the most potential α7 nAChR ligands. While the suitability of dibenzothiophene derivatives as PET tracers is recognized, the potential of fluoren-9-ones is insufficiently investigated. We herein report on a series of fluoren-9-one based derivatives targeting α7 nAChR with compounds 8a and 8c possessing the highest affinity and selectivity. Accordingly, with [18F]8a and [18F]8c we designed and initially evaluated the first fluoren-9-one derived α7 nAChR selective PET ligands. A future application of these radioligands is facilitated by the herein presented successful implementation of fully automated radiosynthesis.  相似文献

Respiratory electron transport system (ETS) activity was measuredin plankton samples (<200 µm) collected in the NW AlboranSea. Sampling was carried out during seasonal cruises (summerand autumn 2003 and winter and spring 2004) in 12 stations locatedin transects off the coast of Malaga (southern Spain). Thiswork reports for the first time seasonal variations of the Arrheniusactivation energy (Ea) as well as being the first study to addressCO2 balance in the NW Alboran Sea. These variations were relatedto changes in the phytoplankton community assemblage, whichcould ultimately be caused by the seasonal variability of hydrologicalconditions. ETS activity was significantly higher in summer,coinciding with a higher chlorophyll a (Chl a) concentrationand relatively high levels of particulate organic matter. TheETS:Chl atotal ratios were low during the four seasons, suggestinga high contribution of autotrophic phytoplankton to the respiratoryactivity of planktonic community. Respiratory CO2 production(RCP) calculated from ETS activity ranged from 4.6 to 28.1 mgC m–3 day–1 during the four cruises. Chl a-specificRCP was lower than the maximum photosynthetic rates reportedin the literature for the studied area, suggesting that primaryproduction (PP) and respiration in the water column might beunbalanced.  相似文献

The Opuntia (prickly pear) genus, an important horticultural crop in Mexico, is essentially a fruit crop with two variants: sweet (“tunas”) or acid (“xoconostles”) fruits; it is also a source of vegetables “nopalitos” or fodder for livestock, among other uses. Its taxonomical classification has been reported as complex, although few studies on the genetic structure of Mexican Opuntia are available, and genetic differences between the two types of fruits are unknown. Opuntia genotype identification and classification are still mainly based on morphological characters. In this study, the genetic diversity of Mexican Opuntia germplasm with agronomic and economic importance was revealed, using 88 accessions and 13 SSR markers, in an attempt to explore the genetic relationships among them. A total of 159 alleles were detected ranging from 7 to 23 per locus with an average of 12.2. The SSR markers generated unique fingerprints for each Opuntia accession confirming their usefulness for genetic analysis. The accessions’ grouping was defined by several complementary clustering methods, and the moderate incongruences between the different methods did not influence the overall clustering. DAPC and STRUCTURE analyses grouped the accessions into five groups, thus confirming the incorrect delimitation of species in this genus. The following species had no clear boundaries: Opuntia ficus-indica, Opuntia albicarpa, Opuntia megacantha, Opuntia streptacantha, Opuntia lasiacantha, and Opuntia hyptiacantha. However, Opuntia robusta was separated from the rest of the species. Opuntia joconostle and Opuntia matudae, which produce acid fruits, tended to differ from the others. Median-joining simulation classified all genotypes into a complex network, and both linear and reticular ties between Mexican Opuntia genotypes were revealed. The genetic distance revealed in the present study shows the importance of Mexican accessions for conservation and use in breeding programs.  相似文献
