Seventeen aporphines were tested for antiplatelet activity. L-(+)-hemovine HCl and 7-hydroxydehydrothalicsimidine strongly inhibited platelet aggregation induced by adenosine 5'-diphosphate (ADP), arachidonic acid (AA), collagen, and platelet-activating factor (PAF). Previous studies indicate that the H3 influenza virus has the ability to establish infection upon interspecies transmission and poses a threat to mammals. Therefore, it is important to enhance the surveillance of H3 avian influenza viruses (AIVs). In this study, A/duck/Shanghai/C84/2009(H3N2) (C84) was isolated from a live poultry market in Shanghai, China. Using PCR and sequencing analyses, we obtained the whole-genome sequence of this virus. The H3N2 virus proved to be a novel multiple-gene reassortant AIV whose genes were derived from H3N2, H4N6, H6N2, and H9N2. Knowledge regarding the complete genome sequence of the C84 virus will be useful for epidemiological surveillance. 相似文献

Cationic cotton was prepared by a designed two-bath pad-bake process with 3-chloro-2-hydroxypropyltrimethylammonium chloride as cationizing reagent to realize recycle utilization of the reagent and continuous processing of cationization. Experiments showed that 8.0% (o.w.bath) of the reagent, 1:1 of molar ratio of sodium hydroxide to the reagent, 60 °C and 6 min of baking temperature and time were selected for cationization and the obtained cationic cotton was suitable for application in salt-free reactive dyeing. The structures of both the untreated and cationic fibers were investigated by X-ray diffraction and scanning electronic microscopy. Higher dye utilization and color yields could be realized on the cationic cotton than that on the untreated one in the conventional dyeing. Levelness dyeing and good fastness properties of the dyes on the cationic fabrics were obtained. Besides, colorimetric properties and mechanical strength of the dyed fabrics were both evaluated to show applicability of this preparation process of cationic cotton. 相似文献

The α-acetolactate decarboxylase (ALDC) can reduce diacetyl fleetly to promote mature beer. A safe strain Bacillus subtilis WB600 for high-yield production of ALDC was constructed with the ALDC gene saald from Staphylococcus aureus L3-15. SDS-PAGE analysis revealed that S. aureus α-acetolactate decarboxylase (SaALDC) was successfully expressed in recombinant B. siutilis strain. The enzyme SaALDC was purified using Ni-affinity chromatography and showed a maximum activity at 45 °C and pH 6.0. The values of K m and V max were 17.7 μM and 2.06 mM min?1, respectively. Due to the unstable property of SaALDC at low pH conditions that needed in brewing process, site-directed mutagenesis was proposed for improving the acidic stability of SaALDC. Homology comparative modeling analysis showed that the mutation (K52D) gave rise to the negative-electrostatic potential on the surface of protein while the numbers of hydrogen bonds between the mutation site (N43D) and the around residues increased. Taken together the effect of mutation N43D-K52D, recombinant SaALDCN43D-K52D showed dramatically improved acidic stability with prolonged half-life of 3.5 h (compared to the WT of 1.5 h) at pH 4.0. In a 5-L fermenter, the recombinant B. subtilis strain that could over-express SaALDCN43D-K52D exhibited a high yield of 135.8 U mL?1 of SaALDC activity, about 320 times higher comparing to 0.42 U mL?1 of S. aureus L3-15. Among 21 human strains of Laribacter hongkongensis, small plasmids were observed in four strains, and large ones in six strains. The smallest, 3264-bp plasmid, pHLHK19, has only one ORF that encodes a putative replication initiator protein and a predicted origin of replication (ori) with a DnaA box, three 18-bp direct repeats and five pairs of inverted repeats. An Escherichia coli-L. hongkongensis shuttle vector was constructed by ligating the HindIII-digested pHLHK19, containing the replication initiator protein and ori of pHLHK19, to HindIII-digested pBK-CMV. We recently reported that the p12 subunit of human DNA polymerase δ (Pol δ4) is degraded by CRL4Cdt2 which regulates the licensing factor Cdt1 and p21WAF1 during the G1 to S transition. Presently, we performed multiparameter laser scanning cytometric analyses of changes in levels of p12, Cdt1 and p21WAF1, detected immunocytochemically in individual cells, vis-à-vis the initiation and completion of DNA replication. The latter was assessed by pulse-labeling A549 cells with the DNA precursor ethynyl-2′-deoxyribose (EdU). The loss of p12 preceded the initiation of DNA replication and essentially all cells incorporating EdU were p12 negative. Completion of DNA replication and transition to G2 phase coincided with the re-appearance and rapid rise of p12 levels. Similar to p12 a decline of p21WAF1 and Cdt1 was seen at the end of G1 phase and all DNA replicating cells were p21WAF1 and Cdt1 negative. The loss of p21WAF1 preceded that of Cdt1 and p12 and the disappearance of the latter coincided with the onset of DNA replication. Loss of p12 leads to conversion of Pol δ4 to its trimeric form, Pol δ3, so that the results provide strong support to the notion that Pol δ3 is engaged in DNA replication during unperturbed progression through the S phase of cell cycle. Also assessed was a correlation between EdU incorporation, likely reflecting the rate of DNA replication in individual cells, and the level of expression of positive biomarkers of replication cyclin A, PCNA and Ki-67 in these cells. Of interest was the observation of stronger correlation between EdU incorporation and expression of PCNA (r = 0.73) than expression of cyclin A (r = 0.47) or Ki-67 (r = 0.47). 相似文献

目的:探讨联合检测血清胸苷激酶1(TK1)与乳酸脱氢酶(LDH)水平在非霍奇金淋巴瘤(NHL)患者鉴别诊断及疗效监测中的临床意义。方法:收集2016年1月至2018年6月我院诊治的111例非霍奇金淋巴瘤的初诊患者血清标本,并选择50例正常人血清标本作为对照,采用免疫印迹增强发光法检测TK1浓度,比色法检测LDH浓度。所有患者随访至少1年,分析和比较惰性NHL与侵袭性NHL及各自四类分期之间血清TK1和LDH水平的差异,化疗后完全缓解、部分缓解与未缓解组LDH水平以及NHL患者中血清TK1和LDH的阳性率。结果:高度侵袭性NHL患者和侵袭性NHL患者血清TKI和LDH水平与惰性NHL患者相比显著增高(P0.05),但惰性NHL患者血清TK1和LDH水平与正常组之间差异无统计学意义(P0.05);Ⅲ、Ⅳ期侵袭性NHL患者血清TK1和LDH水平与Ⅰ、Ⅱ期患者相比显著增高(P0.05)。与化疗前相比,四次化疗后,完全缓解组NHL患者血清LDH水平下降21.05%,部分缓解组为16.66%,病情稳定组血清LDH水平升高至11.54%,三组NHL患者血清LDH水平比较差异具有统计学意义(P0.008),两组之间的差异均有统计学意义(P0.05)。结论:联合检测血清TK1和LDH水平对于NHL患者的鉴别诊断、疗效评估均具有重要参考价值。 相似文献
