On their way to the synapse and back, neuronal proteins are carried in cargo vesicles along axons and dendrites. Here, we demonstrate that the key parameters of axonal transport, i.e., particle velocities and pausing times can be read out from CCD-camera images automatically. In the present study, this is achieved via cross- and autocorrelation of kymograph columns. The applicability of the method was measured on simulated kymographs and data from axonal transport timeseries of mRFP-labeled synaptophysin. In comparing outcomes of velocity determinations via a performance parameter that is analogous to the signal-to-noise ratio (SNR) definition, we find that outcomes are dependent on sampling, particle numbers and signal to noise of the kymograph. Autocorrelation of individual columns allows exact determination of pausing time populations. In contrast to manual tracking, correlation does not require experience, a priori assumptions or disentangling of individual particle trajectories and can operate at low SNR. Only few factors influencing pest populations can be studied in the laboratory, but many population‐driving factors interact in the field. Therefore, complementary laboratory and field approaches are required for reliable predictions of real‐world patterns and processes. Laboratory and field experiments with the red spider mite, Oligonychus ilicis McGregor (Acari: Tetranychidae), and the coffee leaf miner, Leucoptera coffeella Guérin‐Méneville (Lepidoptera: Lyonetiidae), on coffee plants, Coffea arabica L. (Rubiaceae), were combined to study the relative importance of biotic interactions, including resource preferences and natural‐enemy impact, and habitat factors, such as agroforestry type and management intensity, on coffee pest densities. In the laboratory, leaf discs cut from undamaged coffee plants were significantly preferred by red spider mites over those from plants infested with conspecific mites, leaf rust pathogens [Hemileia vastatrix Berkeley & Broome (Uredinales)], or coffee leaf miners, resulting in higher reproductive success. Similarly, undamaged plants were preferred by coffee leaf miners over red spider mite‐infested plants. However, in the field, red spider mite densities were positively correlated with coffee leaf miner and leaf rust densities, thereby contrasting with laboratory predictions. Hence, our study suggests that the importance of resource preferences and fitness expected based on laboratory experiments was suppressed by environmental conditions in the field, though other unassessed biotic interactions could also have played a role. Furthermore, intensified agroforestry was characterized by higher red spider mite densities, whereas densities of its major natural enemy, the predatory mite Amblyseius herbicolus Chant (Acari: Phytoseiidae), were not related to agroforestry management. Densities of coffee leaf miner and its main natural enemy, a eulophid parasitoid (Hymenoptera), were not affected by management practices. In conclusion, patterns found in the laboratory did not hold for the field, emphasizing the difficulties of extrapolating small‐scale experiments to larger spatial scales and the need to combine both approaches.  相似文献

Prey from the decomposer subsystem may help sustain predator populations in arable fields. Adding organic residues to agricultural systems may therefore enhance pest control. We investigated whether resource addition (maize mulch) strengthens aboveground trophic cascades in winter wheat fields. Evaluating the flux of the maize-borne carbon into the food web after 9 months via stable isotope analysis allowed differentiating between prey in predator diets originating from the above- and belowground subsystems. Furthermore, we recorded aphid populations in predator-reduced and control plots of no-mulch and mulch addition treatments. All analyzed soil dwelling species incorporated maize-borne carbon. In contrast, only 2 out of 13 aboveground predator species incorporated maize carbon, suggesting that these 2 predators forage on prey from the above- and belowground systems. Supporting this conclusion, densities of these two predator species were increased in the mulch addition fields. Nitrogen isotope signatures suggested that these generalist predators in part fed on Collembola thereby benefiting indirectly from detrital resources. Increased density of these two predator species was associated by increased aphid control but the identity of predators responsible for aphid control varied in space. One of the three wheat fields studied even lacked aphid control despite of mulch-mediated increased density of generalist predators. The results suggest that detrital subsidies quickly enter belowground food webs but only a few aboveground predator species include prey out of the decomposer system into their diet. Variation in the identity of predator species benefiting from detrital resources between sites suggest that, depending on locality, different predator species are subsidised by prey out of the decomposer system and that these predators contribute to aphid control. Therefore, by engineering the decomposer subsystem via detrital subsidies, biological control by generalist predators may be strengthened.  相似文献

The relationship between density and area depends on local growth rates and the area-dependence of migration rates. These rates vary among taxa due to dispersal behaviour, plot productivity and natural enemy impact. Previous studies in aphids suggest that aphid densities are highest in patches of intermediate sizes, and lower in small and large patches. The suggested mechanism causing these patterns is that the dispersal behaviour in aphids creates a mixture of area- and perimeter-dependent migration rates. In this paper, we used these predictions to examine the additional consequences of nutrient availability and natural enemies on the density-area relationship. The derived predictions were compared to data from a system with three aphid species, a set of aphid parasitoids and generalist natural enemies, and at two levels of plant nutrient availability. We find that predictions from the model based only on dispersal and local growth agree with the temporal dynamics of density-area relationships for aphids in high nutrient patches. In patches with low nutrients, high parasitism rates appeared to cause a negative density-area relationship for aphids, thereby deviating from predictions driven by the aphids' dispersal behavior. Hence, the dispersal model with scale-dependent migration rates can provide a useful tool for understanding insect distribution in patch size gradients, but the relative importance of top-down effects can completely change with plot productivity.  相似文献

While investigating biodiversity patterns on different spatial scales, ecological processes determining these patterns have been rarely analysed. Flower visitation by bees is an important ecological process that is related to floral resource availability. However, little is known about whether responses of bee communities to floral resource availability change at different spatial scales. We studied density and species richness of flower-visiting bees in relation to floral resource availability, provided by coffee, in traditional agroforestry systems on a field, shrub, and branch scale. On a field scale, mean bee density per shrub increased with decreasing proportion of flowering coffee shrubs per site, showing a dilution effect. Conversely, on shrub and branch scales bee density per shrub, or shrub part, increased with increasing number of inflorescences, showing a concentration effect. Additionally, bee density per shrub was higher on those that were only partly, rather than totally surrounded by other flowering coffee shrubs. Species richness of flower-visiting bees was positively affected by high resource availability on a shrub and a branch scale, expressed by a high number of inflorescences, but at the field scale the proportion of flowering shrubs per site did not have any effect on species richness. Our results show contrasting responses of the community of flower-visiting bees to floral resource availability, depending on the spatial scale considered. We conclude that patterns of flower-visiting bee communities of only one spatial scale can not be generalized, since the number of pollinators may be limited on a field scale, but not on smaller scales.  相似文献

Curcumin is known for its anticancer properties, but its clinical application is limited due to its poor bioavailability and chemical stability. In this study we report the curcumin derivative, ST03 (1,2-bis[(3E,5E)-3,5-bis[(2-chlorophenyl)methylene]-4-oxo-1-piperidyl]ethane-1,2-dione) exhibits ∼ 14 fold better bioavailability compared to curcumin and is detectable in plasma up to 12 h. ST03 induces ROS, activates the intrinsic apoptotic pathway as evident by disruption of mitochondrial membrane potential, and induction of proapoptotic proteins in ovarian cancer lines PA1 and A2780. ST03 also blocked the migration of ovarian cancer cells. ST03 exerted its antitumor effect in-vivo in the EAC mouse model by activating the intrinsic apoptotic pathway. Our findings demonstrate ST03, a curcumin derivative, with better bioavailability and stability with no discernable toxicity in vivo to be a promising drug candidate for anticancer therapies.  相似文献

Concomitant with the rapid loss of tropical mature forests, the relative abundance of secondary forests is increasing steadily and the latter are therefore of growing interest for conservation. We analysed species richness of fruit-feeding nymphalid butterflies in secondary forest fragments of different age and isolation and in mature forest at the eastern margin of the Lore Lindu National Park in Central Sulawesi, Indonesia. From April to August 2001 we collected 2322 individuals of fruit-feeding butterflies, belonging to 33 species. Butterfly species richness increased with succession, but was significantly higher in mature forests than in all types of secondary forest. Isolation of the forest fragments did not have a significant effect on butterfly species richness in the range of distances (up to 1700 m) studied. Rather it appeared to affect only a few species. Species richness of endemic species was higher than of non-endemic species. Although endemic species were most diverse in mature forests, many species captured were restricted to secondary forests. Our results show that mature forest is essential for the conservation of nymphalid butterflies and for the endemic species in this area. However, considering the relatively large number of species found in these rather small habitat islands, secondary forest fragments, especially older successional stages, can be taken into account in conservation efforts and thus contribute to the preservation of tropical biodiversity on a landscape scale.  相似文献

Little is known about how tropical land-use systems contribute to the conservation of functionally important insect groups, including dung beetles. In a study at the margin of Lore Lindu National Park (a biodiversity hotspot in Central Sulawesi, Indonesia) dung-beetle communities were sampled in natural forest, young secondary forest, agroforestry systems (cacao plantations with shade trees) and annual cultures (maize fields), each with four replicates (n = 16 sites). At each site we used 10 pitfall traps, baited with cattle dung, along a 100 m transect for six 3-day periods. The number of trapped specimens and species richness at the natural forest sites was higher than in all land-use systems, which did not significantly differ. Each land-use system contained, on average, 75% of the species richness of the natural forest, thereby indicating their importance for conservation. However, a two-dimensional scaling plot based on NESS indices (m = 6) indicated distinct dung beetle communities for both forest types, while agroforestry systems and annual cultures exhibited a pronounced overlap. Mean body size of dung beetles was not significantly influenced by land-use intensity. Five of the six most abundant dung beetle species were recorded in all habitats, whereas the abundance of five other species was significantly related to habitat type. Mean local abundance and number of occupied sites were closely correlated, further indicating little habitat specialisation. The low dung beetle diversity (total of 18 recorded species) may be due to the absence of larger mammals in Sulawesi during historical times, even though Sulawesi is the largest island of Wallacea. In conclusion, the dung beetle fauna of the lower montane forest zone in Central Sulawesi appears to be relatively robust to man-made habitat changes and the majority of species did not exhibit strong habitat preferences.  相似文献

Aim Factors acting at various scales may affect biodiversity, demanding analyses at multiple spatial scales in order to understand how community richness is determined. Here, we adopted a hierarchical approach to test the contribution of region, landscape heterogeneity, local management (organic vs. conventional) and location within field (edge vs. centre) to the species richness and abundance of spiders in cereals. Location Three regions of western and central Germany: Leine Bergland, Soester Boerde, and Lahn‐Dill Bergland. Methods Forty‐two paired organic and conventional winter wheat fields were compared. Field pairs were located in areas ranging from structurally simple to structurally complex landscapes. In May and June 2003, spiders were sampled using pitfall traps. Linear mixed models were used to determine the relationship of spider diversity and abundance with regional spatial factors and landscape heterogeneity within a 500‐m radius, as well as with local management and within‐field location. Results Within‐field location of the traps and landscape heterogeneity were the best predictors of species richness: more species were found in field edges and in heterogeneous landscapes. Region and local management had no effect on species richness. Activity density was higher in field edges and differed among regions. Main conclusions The diversity of farmland spiders was influenced by differences at two of the spatial scales (edge vs. centre, simple vs. complex landscapes), but not at the two others (field management, region), emphasizing the importance of analyses at a variety of spatial scales for an adequate explanation of patterns in biodiversity. Our study suggests that promoting heterogeneity in land use at landscape scales is one of the keys to promoting spider diversity in agroecosystems.  相似文献

Swiprosin-1/EFhd2 (EFhd2) is a cytoskeletal Ca2+ sensor protein strongly expressed in the brain. It has been shown to interact with mutant tau, which can promote neurodegeneration, but nothing is known about the physiological function of EFhd2 in the nervous system. To elucidate this question, we analyzed EFhd2−/−/lacZ reporter mice and showed that lacZ was strongly expressed in the cortex, the dentate gyrus, the CA1 and CA2 regions of the hippocampus, the thalamus, and the olfactory bulb. Immunohistochemistry and western blotting confirmed this pattern and revealed expression of EFhd2 during neuronal maturation. In cortical neurons, EFhd2 was detected in neurites marked by MAP2 and co-localized with pre- and post-synaptic markers. Approximately one third of EFhd2 associated with a biochemically isolated synaptosome preparation. There, EFhd2 was mostly confined to the cytosolic and plasma membrane fractions. Both synaptic endocytosis and exocytosis in primary hippocampal EFhd2−/− neurons were unaltered but transport of synaptophysin-GFP containing vesicles was enhanced in EFhd2−/− primary hippocampal neurons, and notably, EFhd2 inhibited kinesin mediated microtubule gliding. Therefore, we found that EFhd2 is a neuronal protein that interferes with kinesin-mediated transport.  相似文献
