A comparative study of tetraheme cytochrome c3 molecules from several species was carried out using recently developed theoretical methods based on continuum electrostatics. The binding joint equilibrium of electrons and protons was simulated, revealing the complete thermodynamic aspects of electron-proton coupling in these molecules. The method yields excellent accuracy in terms of midpoint potentials, giving the correct reduction orders in all molecules examined, except for one heme site. The coupling between electrons and protons is shown to be present and significant at physiological pH in all cases. This phenomenon, known as the redox-Bohr effect, though of thermodynamic nature, is shown to have an intrinsic "dynamic" character at the molecular level (in the sense of the empty/occupied fluctuations at the microscopic level), with the binding states of redox and protonatable sites displaying both correlated averages and correlated fluctuations. The protonatable sites more directly involved in the redox-Bohr effect are identified using, among other properties, the statistical correlation between pairs of sites, which automatically reflects indirect effects mediated by other sites. Several sites are identified in this analysis. Propionate D of heme I seems to be the most interesting, generally showing a high correlation not only with its own heme, but also with heme II, corresponding to an indirect stabilization of the reduced forms of both hemes. Other interesting sites are the free histidines of two of the cytochromes and propionate D of heme IV, the latter being potentially associated with redox-induced structural changes. Among the set of cytochromes c3 analyzed in this study, significant differences are observed for several properties of the acidic cytochrome included in the set, from Desulfovibrio africanus, supporting the hypothesis of a different functional role.  相似文献

The Bacillus subtilis endospore coat protein CotA shows laccase activity. By using comparative modeling techniques, we were able to derive a model for CotA based on the known x-ray structures of zucchini ascorbate oxidase and Cuprinus cereneus laccase. This model of CotA contains all the structural features of a laccase, including the reactive surface-exposed copper center (T1) and two buried copper centers (T2 and T3). Single amino acid substitutions in the CotA T1 copper center (H497A, or M502L) did not prevent assembly of the mutant proteins into the coat and did not alter the pattern of extractable coat polypeptides. However, in contrast to a wild type strain, both mutants produced unpigmented colonies and spores unable to oxidize syringaldazine (SGZ) and 2'2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS). The CotA protein was purified to homogeneity from an overproducing Escherichia coli strain. The purified CotA shows an absorbance and a EPR spectra typical of blue multicopper oxidases. Optimal enzymatic activity was found at < or =pH 3.0 and at pH 7.0 for ABTS or SGZ oxidation, respectively. The apparent K(m) values for ABTS and SGZ at 37 degrees C were of 106 +/- 11 and 26 +/- 2 microm, respectively, with corresponding k(cat) values of 16.8 +/- 0.8 and 3.7 +/- 0.1 s(-1). Maximal enzyme activity was observed at 75 degrees C with ABTS as substrate. Remarkably, the coat-associated or the purified enzyme showed a half-life of inactivation at 80 degrees C of about 4 and 2 h, respectively, indicating that CotA is intrinsically highly thermostable.  相似文献

This article reviews the history and current state of ethanol production from sugarcane in Brazil and presents a strategy for improving ecosystem services and production. We propose that it is possible to produce ethanol from sugarcane while maintaining or even recovering some of Brazil's unique neotropical biodiversity and ecosystem climate services. This approach to the future of sustainable and responsible ethanol production is termed the ‘midway’ strategy. The ‘midway’ strategy involves producing the necessary biotechnology to increase productivity while synergistically protecting and regenerating rainforest. Three main areas of scientific and technological advance that are key to realizing the ‘midway’ strategy are: (i) improving the quality of scientific data on sugarcane biology as pertains to its use as a bioenergy crop; (ii) developing technologies for the use of bagasse for cellulosic ethanol; and (iii) developing policies to improve the ecosystem services associated with sugarcane landscapes. This article discusses these three issues in the general context of biofuels production and highlights examples of scientific achievements that are already leading towards the ‘midway’ strategy.  相似文献

A study was conducted to assess Typha spp.'s ability to withstand and remove, from water, a metabolite of blood lipid regulator drugs, clofibric acid (CA). At a concentration of 20 microg L(-1), Typha had removed >50% of CA within the first 48h, reaching a maximum of 80% by the end of the assay. Experimental conditions assured that photodegradation, adsorption to vessel walls and microbial degradation did not contribute to the removal. Exposure to higher CA concentrations did not affect Typha's photosynthetic pigments but the overall increase in enzyme activity (ascorbate and guaiacol peroxidases, catalase, superoxide dismutase) indicates that both roots and leaves were affected by the xenobiotic. Eventually, Typha seemed able to cope with the CA's induced oxidative damage suggesting its ability for phytoremediation of CA contaminated waters.  相似文献

The microorganisms associated with sugary Brazilian kefir beverage were investigated using a combination of culture-dependent and -independent methods. A total of 289 bacteria and 129 yeasts were identified via phenotypic and genotypic methods. Lb. paracasei (23.8%) was the major bacterial isolate identified, followed by Acetobacter lovaniensis (16.31%), Lactobacillus parabuchneri (11.71%), Lactobacillus kefir (10.03%) and Lactococcus lactis (10.03%). Saccharomyces cerevisiae (54.26%) and Kluyveromyces lactis (20.15%) were the most common yeast species isolated. Scanning electron microscopy showed that the microbiota was dominated by lemon-shaped yeast cells growing in close association with Lactobacillus (long and curved). Some lactic acid bacteria detected by sequence analysis of DGGE (denaturing gradient gel electrophoresis) bands were not recovered at any time through fermentation by plating. Conversely, DGGE fingerprints did not reveal bands corresponding to some of the species isolated by culturing methods. The bacteria Acetobacter lovaniensis and the yeast Kazachstania aerobia are described for the first time in sugary kefir. During the 24 h of fermentation, the concentration of lactic acid ranged from 0.2 to 1.80 mg/ml, and that of acetic acid increased from 0.08 to 1.12 mg/ml. The production of ethanol was limited, reaching a final mean value of 1.24 mg/ml.  相似文献

Since historical times, the inherent human fascination with pearls turned the freshwater pearl mussel Margaritifera margaritifera (Linnaeus, 1758) into a highly valuable cultural and economic resource. Although pearl harvesting in M. margaritifera is nowadays residual, other human threats have aggravated the species conservation status, especially in Europe. This mussel presents a myriad of rare biological features, e.g. high longevity coupled with low senescence and Doubly Uniparental Inheritance of mitochondrial DNA, for which the underlying molecular mechanisms are poorly known. Here, the first draft genome assembly of M. margaritifera was produced using a combination of Illumina Paired-end and Mate-pair approaches. The genome assembly was 2.4 Gb long, possessing 105,185 scaffolds and a scaffold N50 length of 288,726 bp. The ab initio gene prediction allowed the identification of 35,119 protein-coding genes. This genome represents an essential resource for studying this species’ unique biological and evolutionary features and ultimately will help to develop new tools to promote its conservation.  相似文献

Patterns of range size evolution are important for developing an evolutionary biogeographical theory and supporting conservation actions. Determining those patterns is however hampered by multiple factors acting on range size and by our uncertainty with regard to species' phylogenetic relationships and geographic distribution. In addition, given the diversity of analytic approaches existing in the literature, there are concerns regarding whether different methods might lead to different patterns. Here, we addressed these uncertainties in order to test for correlations between the evolution of morphology (body size and shape) and range size. We studied lizards of the family Gymnophthalmidae, representing changes of an order of magnitude in body size and also two independent lineages with limb‐reduced (snake‐like) morphs. We used phylogenetic multivariate methods (pPCA) to control for allometric effects of body size over shape. Then, we performed multiple regressions under three approaches: ‘naive’ least squares, Bayesian comparative methods that accounted for uncertainties in trait optimization, tree topology and branch length, and a novel permutative procedure based on phylogenetic generalized least squares (pGLS) to assess the robustness of our relationships to uncertainties in the range sizes of the species studied. All approaches led to the same answer: only body size was related to range size. While bigger gymnophthalmids tend to have relatively large ranges, small species can have either small or large ones. Our results’ robustness to strong uncertainties in both phylogenetic relationships and range sizes shows there are opportunities for overcoming those problems and produce reliable patterns of range size evolution. However, interpretation of the processes driving patterns of range size evolution will still require advances in phylogenetic and taxonomic knowledge. We discuss the application of morphology–range size relationships to conservation planning in the light of existing uncertainties in the geographic knowledge of our studied species group and workarounds for data availability.  相似文献

In the context of ongoing climatic warming, certain landscapes could be near a tipping point where relatively small changes to their fire regimes or their postfire forest recovery dynamics could bring about extensive forest loss, with associated effects on biodiversity and carbon‐cycle feedbacks to climate change. Such concerns are particularly valid in the Klamath Region of northern California and southwestern Oregon, where severe fire initially converts montane conifer forests to systems dominated by broadleaf trees and shrubs. Conifers eventually overtop the competing vegetation, but until they do, these systems could be perpetuated by a cycle of reburning. To assess the vulnerability of conifer forests to increased fire activity and altered forest recovery dynamics in a warmer, drier climate, we characterized vegetation dynamics following severe fire in nine fire years over the last three decades across the climatic aridity gradient of montane conifer forests. Postfire conifer recruitment was limited to a narrow window, with 89% of recruitment in the first 4 years, and height growth tended to decrease as the lag between the fire year and the recruitment year increased. Growth reductions at longer lags were more pronounced at drier sites, where conifers comprised a smaller portion of live woody biomass. An interaction between seed‐source availability and climatic aridity drove substantial variation in the density of regenerating conifers. With increasing climatic water deficit, higher propagule pressure (i.e., smaller patch sizes for high‐severity fire) was needed to support a given conifer seedling density, which implies that projected future increases in aridity could limit postfire regeneration across a growing portion of the landscape. Under a more severe prospective warming scenario, by the end of the century more than half of the area currently capable of supporting montane conifer forest could become subject to minimal conifer regeneration in even moderate‐sized (10s of ha) high‐severity patches.  相似文献

Calmodulin is a Ca(+2)-binding protein important in a variety of cell functions. The Ca(+2)/calmodulin complex interacts with and regulates various enzymes and target proteins, known as calmodulin-binding proteins (CaMBPs). In this study, we revealed a comparative identification of the CaMBPs composition in the worker honeybee (Apis mellifera) brain, considering two different honeybee behaviors in the colony. To this end, the CaMBPs of forager and nurse workers were purified by affinity chromatography, separated in 1D gel, digested and submitted to peptide mass fingerprinting (PMF). In the PMF analysis, 15 different proteins, considered behavior-specific proteins, were identified, one of them exclusively in forager workers and 10 in nurses. All the proteins were classified in terms of their function and cell localization, revealing a greater expression of metabolism-related CaMBPs in both worker subcastes. Protein sequences were then analyzed for the presence of the calmodulin-binding sites. Therefore, the honeybee brain CaMBPs profiles presented differences between worker subcastes. This is the first identification of calmodulin-binding proteins in the brain of A. mellifera upon nursing and foraging behaviors in the colony and this diversity of target proteins for Ca(+2)/CaM may be involved in terms of the function of these proteins in the nervous system.  相似文献

This paper aims at identifying variables associated with inadequate work ability among nursing personnel at a public hospital, considering factors related to socio-demographic, lifestyles, working conditions, and health outcomes. A cross-sectional study was conducted in a university hospital in S?o Paulo, Brazil, as part of a larger research study on tolerance to 12 h night work. Nursing staff included registered nurses, nurse technicians, and nurse aides; in total, there were 996 healthcare workers (878 female; 118 male) at the time of the study. Some 696 workers (69.9%) of the population agreed to participate. Data collection (October 2004-July 2005) was based on a comprehensive questionnaire about living and working conditions (including incivility at work, work demands, work control, and support), mental and physical health symptoms (fatigue and sleep problems), and work ability. This report presents analyses of the adapted Brazilian version of the Work Ability Index (WAI) and associated variables. The study population worked one of the following shift schedules at this hospital: 12 h nights followed by 36 h off or 9 h or 6 h day (morning or afternoon) shifts. The mean age of the respondents was 34.9 (S.D.+/-10.4) years of age; 31.5% of the participants held two jobs. Statistical analyses using a hierarchical multiple logistic regression model were performed to evaluate the factors associated with inadequate (moderate and low scores) of the WAI. The significantly associated factors were socio-demographic (income responsibility, sole breadwinner, raising kids, age group), working conditions (thermal discomfort, organization of the workplace, and verbal abuse), and health outcomes (high body mass index, obesity, sleep problems, and fatigue). In spite of limitations of the study design, results indicate that the nursing profession is associated with stressful working conditions, contributing to inadequate WAI. This is in addition to bad living conditions and precarious work. Intervention measures, either at the workplace or at individual levels, are necessary to prevent a decrease in work ability, even in this quite young working population.  相似文献
