Anthropogenic pollution causes oxidative stress in plants and reactive oxygen species (ROS) are diminished by antioxidative enzymes and small molecular antioxidants. Pollution may also affect the performance of plant-eating animals by increasing or decreasing their performance. The effects of pollution cannot be fully understood without knowledge of how pollution affects the interactions with the third trophic level, namely natural enemies and diseases of herbivores. In this study, we examined how long-term (19 yr) acid rain pollution affects (i) the oxidative responses in mountain birch foliage and (ii) the growth and immune responses of autumnal moth larvae. We found that pollution caused a 50% increase (p<0.05) in the peroxidase activities (PODs) in birch leaves whereas polyphenoloxidase (PPO) or catalase (CAT) activities were not affected, suggesting that PODs play an important role in the quenching of the oxidative stress in birches. In polluted trees, phenoloxidases probably acted as antioxidative not prooxidative enzymes, which was shown as positive relations between enzyme activities (PPO, CAT) and larval performance (pupal weights). Although acid rain pollution did not have any direct effect on either pupal weight or the length of larval period, the stronger acid rain treatment reduced slightly (6% in females) the encapsulation response of pupae. A decrease of this magnitude might be too small to have measurable effects on the incidence of moth outbreaks.  相似文献

Terrestrial dissolved organic carbon (DOC) provides an external carbon source to lake ecosystems. However, there is ongoing debate about whether external DOC that enters a lake can pass up the food web to support top consumers. We show, from experimental manipulation of a whole lake, that externally loaded DOC can contribute appreciably to fish biomass. Monthly additions of cane sugar with a distinct carbon stable isotope value during 2 years rapidly enriched the 13C content of zooplankton and macroinvertebrates, with a more gradual 13C enrichment of fish. After sugar addition stopped, the 13C content of consumers reverted towards original values. A simple isotope mixing model indicated that by the end of the sugar addition almost 20% of fish carbon in the lake was derived from the added sugar. Our results provide the first direct experimental demonstration at relevant ecological spatial and temporal scales that externally loaded DOC to lakes can indeed transfer to top consumers.  相似文献

Constitutive expression of the Pim-1 kinase prolongs survival of cytokine-deprived FDCP1 cells, partly via maintenance of Bcl-2 expression. Here, we show that Pim-1 colocalizes and physically interacts with the pro-apoptotic Bad protein and phosphorylates it in vitro on serine 112, which is a gatekeeper site for its inactivation. Furthermore, wild-type Pim-1, but not a kinase-deficient mutant, enhances phosphorylation of this site in FDCP1 cells and protects cells from the pro-apoptotic effects of Bad. Our results suggest that phosphorylation of Bad by Pim-1 is one of several mechanisms via which the Pim-1 kinase can enhance Bcl-2 activity and promote cell survival.  相似文献

This is a report of a workshop held on the establishment of human research tissue banking which was held in Levi, Finland 21–24 March 2002.There were 21 participants from 7 European countries. This meeting was attended by representatives from academia, research tissue banks and from the Biotech and Pharmaceutical Industries. The principal aim of the workshop was to find a way to progress the recommendations from ECVAM workshop 44 (ATLA 29, 125–134,2001) and ECVAM workshop 32 (ATLA 26, 763–777, 1998). The workshop represented the first unofficial meeting of the European Network of Research Tissue Banks (ENRTB) steering group. It is expected that in the period preceding the next workshop the ENRTB steering group will co-ordinate the ethical,legislative and organisational aspects of research tissue banking. Key issues dealt with by the Levi workshop included the practical aspects of sharing expertise and experiences across the different European members. Such collaboration between research tissue banks and end users of such material seeks to ultimately enable shared access to human tissue for medical and pharmaco-toxicological research while maintaining strict adherence to differences in legal and ethical aspects related to the use of human tissue in individual countries. This study compared the developmental potential of bovine nuclear transfer embryos with varying amounts of cytoplasm. Embryos formed from single cytoplasts fused to blastomeres by a single electrical pulse or from double cytoplasts using a double electrical pulse resulted in reconstituted embryos containing 75% and 150% of the original oocyte volume. No differences in fusion, cleavage, or development rates to blastocysts were observed between the groups. Mean cell numbers 2 days after fusion were significantly lower in single-cytoplast clones. Cell numbers of resulting blastocysts were likewise significantly lower in single-cytoplast clones. Embryos formed by fusion of blastomeres with single cytoplasts using a single electrical pulse or from double cytoplasts using either a single or a double pulse resulted in reconstituted embryos containing 50%, 100% and 100% of the original oocyte volume. Again, no differences in fusion or cleavage rates were observed between groups, but the development to blastocysts at day 7 was significantly higher in double cytoplasts constructed with one fusion pulse than in single cytoplasts (P< 0.05). Mean cell numbers 2 days after fusion were significantly lower in single-cytoplast clones (P< 0.05), but at the blastocyst stage, no statistically significant differences in cell numbers were observed. The results of this study show that cytoplasmic volume plays a role in the development of nuclear transfer embryos. When using crude enucleation methods such as oocyte bisection, normal cytoplasmic volumes can be achieved by fusing double cytoplasts with embryonic blastomeres. Mol. Reprod. Roentgen stereophotogrammetric analysis (RSA) can be utilized to accurately describe joint kinematics, but even when measuring small displacements within radiographically discernible structures, standardized reference frames are imperative for useful comparison across patients and across studies. In the current paper, accurately controlled laboratory models demonstrated the considerable influence that a mere 1.9-cm offset of the origin of the coordinate system from the rotation axes could exert on translation measures when rotations were occurring. In addition, the use of two different coordinate systems to gauge translation on a radiographic anterior-posterior (A-P) knee laxity exam resulted in a significant correlation (R(2)=0.562) between the two systems; however, differences of up 9.28 mm were found between corresponding measurements. This implies that clinical conclusions can potentially be upheld or refuted, based on the same data set, subject to coordinate system definition. Although the data analyzed presently involved the knee joint, similar issues surround the RSA motion analysis of other joints as well.  相似文献

The microfibril angle (MFA) distribution and the size of cellulose crystallites in isolated double cell walls of Norway spruce (Picea abies [L.] Karst.) tracheids were determined by synchrotron X-ray microdiffraction using the reflections 200 and 004. Samples were 25 μm thick longitudinal sections of earlywood from annual rings 6–18 of several stems. The asymmetric MFA distributions extended from ?20° to 90°. The mean MFA of tangential cell walls decreased from an average of 24° into 19° from the pith to the bark. The mode of the MFA distribution was about 10° smaller than the mean MFA. The standard deviation of the MFA distribution varied between 18° and 25°. The mean MFA and the mode of the MFA distribution were larger in radial than in tangential cell walls. MFA distributions of mature wood samples exhibited a separate small peak at around 90°. The average width and length of cellulose crystallites varied between 28.9–30.9 Å and 192–284 Å, respectively. Both increased slightly as a function of annual ring number from the pith up to the 15th annual ring. An irrigation–fertilisation treatment of some of the stems resulted in longer cellulose crystallites compared to the untreated stems.  相似文献

In contrast to the growth of fungi, the growth of mycobacteria in moisture-damaged building materials has rarely been studied. Environmental mycobacteria were isolated from 23% of samples of moisture-damaged materials (n = 88). The occurrence of mycobacteria increased with increasing concentrations of fungi. Mycobacteria may contribute to indoor exposure and associated adverse health effects.  相似文献

A force balance between the ligaments, articular contact, muscles and body weight maintains knee joint stability. Thus, it is important to study anterior cruciate ligament (ACL) biomechanics, in vivo, under weightbearing conditions. Our objective was to compare the ACL strain response under weightbearing and non-weightbearing conditions and in combination with three externally applied loadings: (1) anterior-posterior shear forces, (2) internal-external torques, and (3) varus-valgus moments. A strain transducer was implanted on the ACL of 11 subjects. All joint loadings were performed with the knee at 20 degrees of flexion. A significant increase in ACL strain was observed as the knee made the transition from non-weightbearing to weightbearing. During anterior shear loading, the strain values produced during weightbearing were greater than those of the non-weightbearing knee (shear loads <40N). At higher shear loads, the strain values became equal. During axial torsion, an internal torque of 10Nm strained the ACL when the knee was non-weightbearing while an equivalent external torque did not. Weightbearing significantly increased ACL strain values in comparison to non-weightbearing with the application of external torques and low internal torques (<3Nm). The strains became equal for higher internal torques. For V-V loading, the ACL was not strained in the non-weightbearing knee. However, weightbearing increased the ACL strain values over the range of moments tested. These data have important clinical ramifications in the development of rehabilitation protocols following ACL reconstruction since weightbearing has been previously thought to provide a protective mechanism to the healing graft.  相似文献
