The thermotropic transitions of the plasma membrane and tonoplastfrom cultured cells of chilling-sensitive (CS) and chilling-insensitive(CI) strains of rice (Oryza sativa L.) were analyzed by monitoringthe fluorescence polarization of an embedded fluorophore, 1,6-diphenyl-1,3,5-hexatriene(DPH), and their relationship to the degree of unsaturationof fatty acids in phospholipids was examined. Polarization values (P) for the tonoplast from cultured cellsof CI rice, in contrast to those from CS rice, exceeded thosefor the plasma membrane. The values for the tonoplast and plasmamembrane from CI cells were somewhat higher than those fromCS cell. Thus, the tonoplast of CI cells has the lowest fluidity,while the fluidity of the tonoplast and plasma membrane of CIcells shows greater dependence on temperature. Arrhenius plotsof the fluorescence anisotropy parameter {(ro/r)–1}–1of DPH in the plasma membrane and tonoplast from CI cells gavea slope that was virtually linear throughout the entire rangeof temperatures from 50°C to 10°C. However, in the caseof CS cells, a discontinuity was sometimes noted in the curvebetween 35°C and 30°C for tonoplast membranes. The activationenergy (Ea) of the anisotropy parameter of DPH in both the plasmamembrane and tonoplast from CI cells was greater than that fromCS cells. Ea in both cases for CS cells increased with increasingduration of exposure to low tempera ture (5°C), becomingnearly the same as that for CI cells. The proportion of unsaturated fatty acids, such as linoleicacid (18:2) and linolenic acid (18:3), in the total phospholipidsof the plasma membrane and tonoplast from CI cells was muchhigher than that from CS cells. In membranes from CS cells,this proportion also increased with increasing duration of exposureto low temperature and reached the value for membranes fromCI cells. In particular, in CS cells, the most dramatic changewas the change in PE and PC that in volved a sharp decreasein levels of 18:1, accompanied by an increase in 18:3. Vacuolar-type H+-ATPase was solubilized from tonoplasts of mung bean (Vigna radiata L.) and purified on a Mono Q anion-exchange column by fast protein liquid chromatography. The purified enzyme was inactivated by the reactive adenine analog, 7-chloro-4-nitrobenzo-2-oxa-1,3-diazole (NBD-Cl). This inactivation was reversed by addition of dithiothreitol (DTT). Inactivation by NBD-Cl was prevented by Mg-ADP, a competitive inhibitor of ATPase. [14C]NBD-Cl predominantly modified the 68-kilodalton subunit and the degree of 14C incorporation was decreased in the presence of Mg-ADP or upon subsequent addition of DTT. The loss of activity followed pseudo first-order kinetics with respect to NBD-Cl concentration, and double log plots of pseudo first-order rate constants versus reagent concentration yielded a straight line with a slope of 0.957. The NBD-modified/inactivated enzyme showed an absorbance maximum at 418 nanometers and a fluorescence emission peak at 515 nanometers. The absorption and fluorescence emission spectra of the NBD-modified enzyme were essentially the same as those of the model compound, N-acetyl-S-NBD cysteine. Absorbance by the modified enzyme at 418 nanometers disappeared upon addition of DTT, which coincided with the restoration of ATPase activity and the decrease in bound [14C]NBD-Cl. These findings show that NBD-Cl modifies an essential cysteine residue(s) at or near the catalytic site in the 68-kilodalton subunit of tonoplast H+-ATPase and that the modification closely correlates with the loss of ATPase activity.  相似文献

In human platelets, thrombin activates Ca2+-activated, phospholipid-dependent protein kinase (protein kinase C) and mobilizes Ca2+ concomitantly, whereas 12-O-tetradecanoylphorbol-13-acetate (TPA) may be intercalated into membranes and directly activates protein kinase C without mobilization of Ca2+ in sufficient quantities. A series of experiments with TPA and Ca2+-ionophore (A23187) indicates that activation of protein kinase C is a prerequisite requirement for release of serotonin, and that this enzyme activation and Ca2+ mobilization act synergistically to elicit a full cellular response. Both cyclic AMP and cyclic GMP inhibit activation of protein kinase C by prohibiting the signal-dependent breakdown of inositol phospholipid to produce diacyl-glycerol, but none of these cyclic nucleotides prevents the TPA-induced activation of this enzyme.  相似文献

Borna disease virus (BDV) is a neurotropic, as yet unclassified, non-segmented, negative-sense, single-strand RNA virus. Natural infection with this virus has been reported to occur in horses and sheep. In addition, antibodies to BDV in plasma or BDV RNA in peripheral blood mononuclear cells (PBMCs) were also found in patients with neuropsychiatric diseases. We describe here the possible link between the patients with chronic fatigue syndrome (CFS) and infection with BDV.  相似文献
