As the initiation step of bacterial infection or biofouling, bacterial adhesion on cells or substrates is generally an optimal target for antibacterial design. Phosphatidylethanolamine (PE) is the principal phospholipid in bacteria, and its function in bacterial adhesion remains unclear. In this study, four E. coli strains including two PE-deficient mutants (PE?PC? and PE?PC+?strains) and two PE-containing wild-type controls (PE?+?PC? strains) were recruited to investigate the influence of PE deficiency on bacterial adhesion. We found that PE deficiency could impair E. coli adhesion on macrophages (human THP-1-derived and mouse RAW264.7 macrophages) or glass coverslips by downregulating lipopolysaccharide (LPS) biosynthesis, which could be reversible by high galactose/lactose but not glucose cultivation. The data imply that PE play important role in bacterial adhesion probably via affecting LPS biosynthesis and suggest that targeting PE biosynthesis is also a potential antibacterial strategy. 相似文献

富亮氨酸重复区的类受体蛋白激酶(LRR-RLKs)在植物对多种信号的响应中发挥作用。应用电子克隆的方法在玉米中克隆了一个LRR-RLKs类基因的全长cDNA,命名为ZmLRRPK1(GenBank接受号为EU873320)。推断的ZmLRRPK1蛋白含有594个氨基酸,分子量为66kDa,等电点pI为5.42,具有典型的LRR-RLKs结构域。半定量RT-PCR结果表明,在ABA、甘露醇和氯化钠的诱导下,ZmLRRPK1在玉米胚芽鞘中表达并在24 h内保持较高的转录水平。 相似文献

With the development of functional genomics research, large-scale proteomics studies are now widespread, presenting significant challenges for data storage, exchange, and analysis. Here we present the Integrated Proteomics Exploring Database (IPED) as a platform for managing proteomics experimental data (both process and result data). IPED is based on the schema of the Proteome Experimental Data Repository (PEDRo), and complies with the General Proteomics Standard (GPS) drafted by the Proteomics Standards Committee of the Human Proteome Organization. In our work, we developed three components for the IPED platform: the IPED client editor, IPED server software, and IPED web interface. The client editor collects experimental data and generates an extensible markup language (XML) data file compliant with PEDRo and GPS; the server software parses the XML data file and loads information into a core database; and the web interface displays experimental results, to provide a convenient graphic representation of data. Given software convenience and data abundance, IPED is a powerful platform for data exchange and presents an important resource for the proteomics community. The refractive index resolution of a surface plasmon resonance (SPR) sensor has been significantly improved these years; however, higher sensing performance is always desired. In this work, we propose a line-monitoring, long-range SPR sensor whose resolution is much better than conventional SPR sensors. Also, in contrast to mono-channel detection, multichannel detection, using line-monitoring technique, can detect multiple channels concurrently. In this way, this system achieves a refractive index resolution of 4.0?×?10??7 refractive index units and can monitor multiple molecular interactions simultaneously. Finally, a model experiment detecting the Escherichia coli bacteria has demonstrated the potential for biomedical applications of this system. 相似文献

Due to a long-term transgression since the Early Cambrian, an extensive shallow-water carbonate platform was developed in the entire Tarim Basin (NW China). During the deposition of the Yingshan Formation (Early-Middle Ordovician), a carbonate ramp system was formed in the intrashelf basin in the Bachu-Keping area of the western basin. Four well-exposed outcrop sections were selected to investigate their depositional facies, cycles, and sequences, as well as the depositional evolution. Detailed facies analyses permit the recognition of three depositional facies associations, including peritidal, semi-restricted subtidal, and open-marine subtidal facies, and eleven types of lithofacies. These are vertically arranged into meter-scale, shallowing-upward peritidal, semi-restricted subtidal, and open-marine subtidal cycles, in the span of Milankovitch frequency bands, suggesting a dominant control of Earth’s orbital forcing on the cyclic sedimentation on the platform. On the basis of vertical facies (or lithofacies) and cycle stacking patterns, as well as accommodation changes illustrated graphically by Fischer plots at all studied sections, six third-order depositional sequences are recognized and consist of lower transgressive and upper regressive parts. In shallow depositional settings, the transgressive packages are dominated by thicker-than-average, shallow subtidal cycles, whereas the regressive parts are mainly represented by thinner-than-average, relatively shallow subtidal to peritidal cycles. In relatively deep environments, however, the transgressive and regressive successions display the opposite trends of cycle stacking patterns, i.e., thinner-than-average subtidal cycles of transgressive packages. Sequence boundaries are mainly characterized by laterally traceable, transitional zones without apparent subaerial exposure features. Good correlation of the long-term changes in accommodation space inferred from vertical facies and cycle stacking patterns with sea-level fluctuations elsewhere around the world suggests an overriding eustatic control on cycle origination, platform building-up and evolution during the Early-Middle Ordovician, although with localized influences of syndepositional faulting and depositional settings. 相似文献

Three new oplopane sesquiterpenes, knorringianalarins D – F ( 1 – 3 , respectively), and five known analogues ( 4 – 8 , respectively), were isolated from the roots and rhizomes of Ligularia knorringiana. The structures of three new compounds were identified as 4‐acetoxy‐11α,12‐epoxy‐2β‐hydroxy‐3β‐(2‐methylbutyryloxy)‐9α‐(4‐methylsenecioyloxy)oplop‐10(14)‐ene ( 1 ), 3β,4‐diacetoxy‐9α‐(4‐acetoxy‐4‐methylsenecioyloxy)‐11α,12‐epoxy‐8α‐(2‐methylbutyryloxy)oplop‐10(14)‐ene ( 2 ), and (1R,5R,6R,7R,9R)‐5,9,11‐trihydroxy‐4,15‐dinoroplop‐10(14)‐en‐3‐one ( 3 ) based on spectroscopic methods including 1D‐ and 2D‐NMR, mass spectrometry, and CD spectroscopy techniques. All compounds were evaluated for their anti‐complementary activity on the classical pathway of the complement system in vitro. Among which, three oplopane sesquiterpenes ( 3 , 7 , and 8 ) exhibited better anti‐complementary effects with CH50 values ranging from 0.33 to 0.89 mm , which are plausible candidates for developing potent anti‐complementary agents. 相似文献

ZAK (sterile alpha motif and leucine zipper containing kinase AZK), a serine/threonine kinase with multiple biochemical functions, has been associated with various cell processes, including cell proliferation, cell differentiation, and cardiac hypertrophy. In our previous reports, we found that the activation of ZAKα signaling was critical for cardiac hypertrophy. In this study, we show that the expression of ZAKα activated apoptosis through both a FAS‐dependent pathway and a mitochondria‐dependent pathway by subsequently inducing caspase‐3. ZAKβ, an isoform of ZAKα, is dramatically expressed during cardiac hypertrophy and apoptosis. The interaction between ZAKα and ZAKβ was demonstrated here using immunoprecipitation. The results show that ZAKβ has the ability to diminish the expression level of ZAKα. These findings reveal an inherent regulatory role of ZAKβ to antagonize ZAKα and to subsequently downregulate the cardiac hypertrophy and apoptosis induced by ZAKα. 相似文献

2010年10月8日-2011年2月26日,跟踪监测了闽江河口互花米草治理试验区人工种植的秋茄(Kandelia candel)、无瓣海桑(Sonneratia apetala)和拉关木(Leguncalaria racemosa)1年生幼苗叶片相溶性物质含量以及活性氧代谢等生理生化指标.结果表明:可溶性糖和脯氨酸含量均随气温的逐步降低而增加,秋茄可溶性糖含量最高,拉关木脯氨酸含量最高;整个监测期无瓣海桑和拉关木幼苗叶片超氧阴离子(O2)产生速率显著高于秋茄(P<0.01),而超氧化物歧化酶(SOD)和过氧化物酶(POD)活性显著低于秋茄(P<0.01);3种植物叶片丙二醛(MDA)含量及电解质渗透率均随着气温的降低而增加,其中无瓣海桑和拉关木MDA含量及电解质渗透率与日最低气温为显著和极显著的负相关关系.2010年冬季持续寒冷天气对闽江河口湿地1年生土著种秋茄幼苗无破坏,对引进种无瓣海桑和拉关木造成了严重的低温胁迫并使幼苗基本全部死亡. 相似文献
