The presence of marrow cells within the cytoplasm of megakaryocytes has been documented and is attributed to the phenomenon of emperipolesis. In a previous study most patients demonstrating this phenomenon had or were suspected of having blood loss. To find out if these are causally related, we induced acute and chronic blood loss in rats and quantitatively measured the emperipolesis index (EI). EI dropped after acute blood loss but rose after chronic blood letting, indicating that blood loss can modulate this phenomenon. In the marrow, megakaryocytes are located preferentially on the abluminal side of sinus endothelium and it is postulated that in the state of heightened demand for cell delivery from marrow into the circulation, some cells take a transmegakaryocytic route to enter the circulation. It has been emphasized that specific bindings between membrane glycoproteins and membrane lectin-like substances are important in cell-to-cell interactions. We explored the surface of granulocyte-macrophage precursor cells (CFU-gm) by the differential agglutination technique. Enrichment of CFU-gm in the agglutinated fraction, containing the cells which have lectin receptors, from marrow treated with soybean agglutinin (SBA), peanut agglutinin (PNA) and concanavalin A (Con A), suggests the presence of reactive galactosyl and mannosyl residues on the surface of CFU-gm. On the other hand, wheat germ agglutinin (WGA), phytohemagglutinin (PHA) and ulex europaeus agglutinin (UEA), which bind to reactive N-acetylglucosamine, N-acetylgalactosamine and fucose, respectively, did not specifically agglutinate CFU-gm. Thus, reactive groups containing galactosyl and mannosyl structures on the surface of CFU-gm may possibly play a role in the process of cell-to-cell interactions between CFU-gm and marrow stromal cells. 相似文献

Through a process resembling receptor-mediated endocytosis, liver endothelium binds and internalizes the plasma glycoprotein ceruloplasmin (CP) on the luminal side. The protein is then transported via a vesicular system to the albuminal side where it is externalized to the space of Disse. In its path, the glycoprotein is fully desialylated. To determine if the endosomal compartment is involved in this transport, we used endosomal inhibitors NH4Cl, ethylamine as well as monensin to quantitatively measure the magnitude of radiolabeled CP transport across purified liver endothelial cells. All three reagents inhibited the transport of CP and its discharge by endothelium. The magnitude of inhibition was dose-related for all three reagents. We conclude that the endosomal compartment is involved in the transendothelial transport of CP across the liver endothelium. 相似文献
