Enzymatic cis-trans isomerization of nitrofuran derivatives was 3-(5-Nitro-2-furyl)-2-(2-furyl)-demonstrated with milk xanthine oxidase. acrylamide (AF-2) and 3-(5-nitro-2-furyl)-2-(5-bromo-2-furyl)acrylamide (NFBFA) were mainly converted from the cis to the trans form by this enzyme supplemented with an electron donor. This enzymatic reaction was further characterized with respect to its cofactor requirements. Finally, a new cis-trans isomerization mechanism, which is based on transfer of a single electron by a nitroreductase such as xanthine oxidase to a nitrofuran derivative to give the anion free radical, was proposed. 相似文献

Blood group H-active polysaccharide has been prepared from “smooth” strain Escherichia coli 2B-V by Freeman's method. a-Fucosidase derived from Bacillus fulminans caused the liberation of fucose from this polysaccharide, together with concomitant loss of blood group H activity. The results of quantitative microanalysis, borohydride reduction, the Morgan-Elson reaction and enzymic hydrolysis with β-galactosidase using isolated oligosaccharides obtained by partial acid hydrolysis indicated that the O-specific side chain of the polysaccharide has a pentasaccharide unit which is β-d-Gal-(1→3)-d-GalNAc-(1→3)-d-GalNAc-Fuc with a D -glucose residue bound at some undetermined point on this structure. The present study provides the first evidence that nitropolycyclic aromatic hydrocarbons such as 1-nitropyrene and 2-nitrofluorene are metabolized to the corresponding formylamino compounds in animal bodies. The study shows that the formation of such novel metabolites, 1-formylaminopyrene and 2-formylaminofluorene, is due to N-formylation of the nitroreduction products, 1-aminopyrene and 2-aminofluorene, by liver formamidase in the presence of N-formyl-L-kynurenine. 相似文献

N-Hydroxy-2-acetylaminofluorene has been found to be an effective inhibitor of aldehyde oxidase. At concentrations of 1 X 10(-6) M and 1 X 10(-5) M, 38% and 88% inhibition was observed on the oxidase activity towards N1-methylnicotinamide. The inhibition was of noncompetitive type and had a Ki value of 4.4 X 10(-6) M. We investigated the effects of the N-ethylmaleimide (NEM), a sulfhydryl(SH) radical blocker, on platelet activation. Platelet aggregation and ATP release was suppressed by 0.2 mM NEM during ADP (20 microM) stimulation and by 0.5 mM NEM during A23187 (4 microM) stimulation. However the agent had no effect on actin polymerization in stimulated platelets. In the absence of a stimulant, NEM (over 1 mM) induced shape changes and slight (5%) actin polymerization, but not aggregation or ATP release. Although platelet aggregation and ATP release were suppressed by the addition of 1 mM NEM during the process of both reactions, the amount of polymerized actin was not influenced by the addition. The reconstructed system consisting of actin and partially purified regulatory proteins without myosin showed a dose-dependent increase in turbidity by the addition of NEM. From these findings, we concluded that NEM enhances actin polymerization, although actin molecules contain SH-radicals, and that actin polymerization has little affect on aggregation and release reaction. 相似文献
