Using agroinfection with a T-DNA vector carrying a hygromycin resistance marker, the recombinants were generated for the first time from the ectomycorrhizal basidiomycete Tricholoma matsutake, which produces commercially valuable fruit bodies, matsutake, during association with Pinus sp. plants. Denaturing high-performance liquid chromatography (DHPLC) is a relatively new technique, which utilizes heteroduplex formation between wild-type and mutated DNA strands to identify point mutations. Heteroduplex molecules are separated from homoduplex molecules by ion-pair, reverse-phase liquid chromatography on a special column matrix with partial heat denaturation of the DNA strands. In order to investigate the application of this method for point mutation detection in drug-resistant genes of Mycobacterium tuberculosis, katG, rpoB, embB, gyrA, pncA and rpsL genes, which are responsible for isoniazid, rifampicin, ethambutol, fluoroquinolone, pyrazinamide and streptomycin resistance, respectively, were detected by temperature-mediated DHPLC in 10 multidrug-resistant and 10 drug-susceptible clinical isolates. The DHPLC data were compared with those from a conventional MIC test. The results show that DHPLC is cost-effective with high capacity and accuracy, and is potentially useful for genotypic screening for mutations associated with anti-tuberculosis drug resistance.  相似文献

Inorganic nitrogen is a substrate for nitrogen assimilation and also functions as a signal triggering widespread changes in gene expression that modulate metabolism and development. To integrate the actions of the nitrogen signal at the whole plant level, plants use multiple signaling routes that communicate internal and external nitrogen status. One route depends on nitrate itself and one uses cytokinin as a messenger. Recent genome-wide research has shown that the nitrate-specific signal regulates a wide variety of metabolic processes including nitrogen and carbon metabolism, and cytokinin biosynthesis. Cytokinin-mediated signaling is related to the control of development, protein synthesis and acquisition of macronutrients. The coordination and interaction of both regulatory pathways is important for normal plant growth under variable nitrogen supply conditions.  相似文献

Habitat selection by tufted ducks (Aythya fuligula), a diving duck which swallows benthic prey organisms, was studied during winter at two neighboring lagoons (Lakes Nakaumi and Shinji, Honshu, Japan) which differ strongly in their benthic fauna and in their diving duck densities. The ducks fed overwhelmingly on the dominant bivalve found in each of the two lagoons, the mussel Musculista senhousia in L. Nakaumi and the clam Corbicula japonica in L. Shinji. In general, however, the ducks probably preferred the mussels to the clams because of: (i) their high (2.9 times) calorific content for their weight; (ii) their high digestibility; (iii) their greater accessibility; and (iv) their shorter handling time. An average tufted duck (850 g) was estimated to require 1.3 kg of mussels or 3.8 kg of clams to meet their daily energy requirements. As a result, the two wintering populations were estimated to consume 4970 t mussels and 4770 t clams during a single wintering season, amounting to some 20% of the standing clam crop. Throughout the winter the average gizzard weight (37 g), and gizzard–body mass ratio (4.2%) of the Lake Nakaumi population were half those of the Lake Shinji population (73 g, 8.1%, respectively), despite their significantly similar nutritive body condition (% body lipid > 12%). The need to maintain a specialized gizzard mass in order to be able to cope with the different prey species results in little opportunity for sampling movements of birds between lakes/prey types and as a result two subpopulations of ducks are indicated to be segregated.  相似文献

Preformed amyloid fibrils accelerate conformational changes of amyloid precursor proteins and result in rapid extension of amyloid fibrils in vitro. We injected various kinds of amyloid fibrils into mice with amyloidogenic apoAII gene (Apoa2(C)). The most severe amyloid depositions were detected in the tissues of mice injected with mouse AApoAII(C) amyloid fibrils. Mild amyloid depositions were also detected in the tissues of mice that were injected with other types of fibrils, including synthetic peptides and recombinant proteins. However, no amyloid depositions were found in mice that were injected with non-amyloid fibril proteins. These results demonstrated that a common structure of amyloid fibrils could serve as a seed for amyloid fibril formation in vivo.  相似文献

Following endoplasmic reticulum (ER) stress, which occurs via inhibition of the glycosylation of newly synthesized proteins, caspase family proteins are activated to promote ER stress-mediated apoptosis. Here we report that nerve growth factor (NGF) suppressed the ER stress-mediated apoptosis in tunicamycin-treated PC12 cells through an extensive decrease of the caspase-3/-9/-12 activity. Detailed analysis of the mechanism underlying the NGF-mediated cell survival revealed that the activities of all seriate caspases were reduced through the phosphatidylinositol 3-kinase (PI3-K) signaling pathway induced by NGF. Moreover, we found that the activity of c-Jun N-terminal kinase (JNK) was not essential for the tunicamycin-induced apoptosis of PC12 cells. These results demonstrate that the inactivation of caspase-12 via the NGF-mediated PI3-K signaling pathway leads to inactivation of the caspase cascade including caspase-3 and -9.  相似文献
